Formulation of Sodium Valproate Nanospanlastics as a Promising Approach for Drug Repurposing in the Treatment of Androgenic Alopecia

Sodium valproate (SV) is an antiepileptic drug that is widely used in the treatment of different seizure disorders. The topical SV has a hair regenerative potential through activating the Wnt/beta-catenin pathway and anagen phase induction. The aim of the current investigation was to fabricate nanospanlastics of SV for improving its dermal delivery by providing prolonged drug effect and increasing its permeability for treatment of androgenic alopecia (AGA). SV-loaded nanospanlastics were formulated according to 2(3) factorial design by ethanol injection method using a non-ionic surfactant (Span 60) and edge activators (EAs), such as Tween 80 and Cremophor RH 40, to explore the influence of different independent variables on entrapment efficiency (EE%) and percentage drug released after 12 h (Q(12h)) in order to choose the optimized formula using Design-Expert software. The optimized formula (F8) appeared as spherical deformable vesicles with EE% of 90.32 +/- 2.18% and Q(12h) of 90.27 +/- 1.98%. F8 exhibited significant improvement of ex vivo permeation than free SV. The clinical study exhibited no comparable difference between F8 and marketed minoxidil lotion. However, F8 demonstrates less adverse effects than minoxidil lotion. Nanospanlastics could be a safe and effective method for improving the topical delivery of SV in the management of AGA.

Synthesis of Spirooxindole Analogs Tethered Pyrazole Scaffold as Acetylcholinesterase Inhibitors

Summary: In this study, a new series of spirooxindole analogs were synthesized and evaluated for their neuroprotective properties through the inhibition of acetylcholine esterase enzyme. Three compounds showed the strongest inhibitory activity, with one compound exhibiting the most potent inhibition. The incorporation of NO2 group and N-methylglycine were found to play a crucial role in the activity of these compounds.

Evaluation of antimicrobial, antiquorum sensing, and cytotoxic activities of new vanillin 1,2,3-triazole derivatives

Summary: In this study, vanillin was used as a starting material to synthesize five new triazole derivatives and one known derivative through click chemistry. The newly synthesized compounds showed enhanced antimicrobial and antifungal activities, while compounds 1 and 2 exhibited high antiquorum-sensing activity. Compounds 4-7 demonstrated selective cytotoxicity against cancer cells.

Novel spirooxindole based benzimidazole scaffold: In vitro, nanoformulation and in vivo studies on anticancer and antimetastatic activity of breast adenocarcinoma

Summary: This study investigated the anticancer activity of the compound SP1 and its nanoformulation against breast adenocarcinoma. The results showed that SP1 exhibited strong inhibitory effects on multiple cancer cell lines and induced apoptosis. The use of a nanoformulation improved the selectivity of the compound against cancer cells. In vivo studies on mice confirmed the ability of SP1 to halt the proliferation of breast cancer cells and suppress metastasis.

An emerging flavone glycoside from Phyllanthus emblica L.: As promiscuous enzyme inhibitor and potential therapeutic in chronic diseases

Summary: In current drug discovery, there is a growing focus on designing drug molecules that interact with multiple targets simultaneously (exhibiting compound promiscuity). Previous screening research on 54 medicinal plants found that the methanol extract of Phylanthus emblica L. leaves showed promising inhibitory activity against alpha amylase, tyrosinase, and hyaluronidase enzymes. Further fractionation of the extract led to the isolation of two flavone glycosides, with one of them, luteolin-7-O-a-L-rhamnoside, exhibiting strong inhibitory activity against the enzymes. This study suggests that luteolin-7-O-a-L-rhamnoside could be used as a potential promiscuous enzyme inhibitor for the management and treatment of certain chronic diseases.

Hyaluronidase Inhibitory Activity and In Silico Docking Study of New Eugenol 1,2,3-triazole Derivatives

Summary: Chemical transformation study is an important tool for new drugs discovery. In this study, new eugenol 1,2,3-triazole derivatives were synthesized via click chemistry using the Cu(I)-catalyzed azide-alkyne 1,3-dipolar cycloaddition method. Their inhibitory effects on hyaluronidase enzyme were examined, and one derivative showed enhanced inhibitory effect compared to its parent compound eugenol.

Microbial Transformation of the Sesquiterpene Lactone, Vulgarin, by Aspergillus niger

Summary: The biotransformation of vulgarin, a sesquiterpene lactone obtained from Artemisia judaica, was performed by Aspergillus niger to produce three more polar metabolites. The structures of these metabolites were determined using NMR spectroscopy and HRESIMS. The anti-inflammatory activity of vulgarin and its metabolites was evaluated, and vulgarin showed preferential inhibitory activity against COX-2, while its metabolite (3a) exhibited selective inhibition against COX-1.
