Cyclodextrins are cyclic oligosaccharides constituted by glucopyranose units linked by alpha-(1,4) bonds that show a characteristic donut shape with a hydrophobic cavity and a hydrophilic outer surface. Cyclodextrins are capable of forming water-soluble host: guest complexes with lipophilic molecules that hide in the their cavity. The aim of the review is to describe the historical background, the structure and proprieties of natural cyclodextrins and their derivatives, their physico-chemical properties and toxicological profile. We also describe the different methods to prepare the inclusion complexes and give an overview of the various applications of such complexes in the different fields. The review discusses and summarizes the most recent literature of cyclodextrins and their application in order to give a comprehensive treatment of the main features of these tools widely used to improve the solubility and bioavailability of many compounds used in the biotechnological and pharmaceutical fields.

The Impact of the COVID-19 Outbreak on Patients' Adherence to PCSK9 Inhibitors Therapy

Valentina Maria Caso, Simona Sperlongano, Biagio Liccardo, Emanuele Romeo, Sergio Padula, Fortunato Arenga, Antonello D'Andrea, Pio Caso, Paolo Golino, Gerardo Nigro, Vincenzo Russo

Summary: During the COVID-19 pandemic, adherence to PCSK9 inhibitors decreased among patients at high cardiovascular risk, leading to elevated LDL cholesterol levels, with few patients maintaining within the therapeutic range. Non-adherence to drug therapy was mainly due to drug prescription failure and fear of infection during the pandemic.

Summary: The crucial role of integrin in pathological processes such as tumor progression and metastasis formation has inspired intense efforts to design novel pharmaceutical agents modulating integrin functions in order to provide new tools for potential therapies. Researchers have discovered a chimeric peptide RGDechi that can specifically recognize alpha v beta 3 integrin and have altered or abolished its binding through chemical modifications. By studying the effects of these modifications, a better understanding of the structural and dynamical determinants involved in integrin recognition mechanism can be obtained.

Copper (I) or (II) Replacement of the Structural Zinc Ion in the Prokaryotic Zinc Finger Ros Does Not Result in a Functional Domain

Martina Dragone, Rinaldo Grazioso, Gianluca D'Abrosca, Ilaria Baglivo, Rosa Iacovino, Sabrina Esposito, Antonella Paladino, Paolo Pedone, Luigi Russo, Roberto Fattorusso, Gaetano Malgieri, Carla Isernia

Summary: There is a strict interaction between copper and zinc in cellular processes. The results of copper's interaction with zinc binding proteins have great significance. In this study, the effects of replacing zinc ions with copper ions of different electronic configurations and ionic radii were explored in the prokaryotic zinc finger protein Ros. The replacement did not lead to a functional domain.

Summary: Humans are estimated to consume several grams per week of nanoplastics through exposure to various sources of contamination. However, the effects of these polymeric particles on living systems are still largely unknown. In this study, the interaction of polystyrene nanoplastics (PS-NPs) with ubiquitin is described at an atomic resolution using CD, NMR, and TEM analyses, revealing the formation of a hard protein corona. Additionally, exposure to PS-NPs in human HeLa cells leads to a significant reduction in ubiquitination. These findings suggest that PS-NPs can cause structural effects on ubiquitin, thereby affecting a vital metabolic process for cell viability.

Summary: This study investigates the mechanism of thermal unfolding of human prion protein and reveals that the transient electrostatic interactions between the N- and C-terminal domains play a key role in regulating the folding process. Summary: This study found that pre-existing atrial fibrillation (AF) is a common comorbidity in COVID-19 patients, accounting for 25% of cases. Even without significant differences in cardiovascular comorbidities or treatments, pre-existing AF was independently associated with an increased risk of developing severe acute respiratory distress syndrome (ARDS) during hospitalization. However, pre-existing AF did not increase the risk of in-hospital mortality.
