Involvement of Essential Signaling Cascades and Analysis of Gene Networks in Diabesity

(1) Aims: Diabesity, defined as diabetes occurring in the context of obesity, is a serious health problem that is associated with an increased risk of premature heart attack, stroke, and death. To date, a key challenge has been to understand the molecular pathways that play significant roles in diabesity. In this study, we aimed to investigate the genetic links between diabetes and obesity in diabetic individuals and highlight the role(s) of shared genes in individuals with diabesity. (2) Methods: The interactions between the genes were analyzed using the Search Tool for the Retrieval of Interacting Genes (STRING) tool after the compilation of obesity genes associated with type 1 diabetes (T1D), type 2 diabetes (T2D), and maturity-onset diabetes of the young (MODY). Cytoscape plugins were utilized for enrichment analysis. (3) Results: We identified 546 obesity genes that are associated with T1D, T2D, and MODY. The network backbone of the identified genes comprised 514 nodes and 4126 edges with an estimated clustering coefficient of 0.242. The Molecular Complex Detection (MCODE) generated three clusters with a score of 33.61, 16.788, and 6.783, each. The highest-scoring nodes of the clusters were AGT, FGB, and LDLR genes. The genes from cluster 1 were enriched in FOXO-mediated transcription of oxidative stress, renin secretion, and regulation of lipolysis in adipocytes. The cluster 2 genes enriched in Src homology 2 domain-containing (SHC)-related events triggered by IGF1R, regulation of lipolysis in adipocytes, and GRB2: SOS produce a link to mitogen-activated protein kinase (MAPK) signaling for integrins. The cluster 3 genes ere enriched in IGF1R signaling cascade and insulin signaling pathway. (4) Conclusion: This study presents a platform to discover potential targets for diabesity treatment and helps in understanding the molecular mechanism.

Non-coding RNAs as biomarkers of myocardial infarction

Summary: Non-coding RNAs (ncRNAs) are RNA molecules that have tissue-specific expression and lack protein-coding potential. Aberrant expression of ncRNAs is critical in disease onset and development, making them promising biomarkers for prognosis and diagnosis. Compared to standard biomarkers, ncRNAs, such as miRNAs and lncRNAs, provide better sensitivity and specificity for diagnosing acute myocardial infarction (AMI), leading to earlier treatment and better prognosis for patients. This review summarizes and discusses the potential clinical use of circulating ncRNAs as MI biomarkers, highlighting their specificity and sensitivity, limitations, and possible solutions.

Effects of the pathological E200K mutation on human prion protein: A computational screening and molecular dynamics approach

Summary: The human prion protein gene (PRNP) is located on chromosome 20 and mutations in this gene are associated with prion disease. The G127V mutation increases protein stability, while the E200K mutation, which has the highest mutation rate, causes Creutzfeldt-Jakob disease (CJD) and induces protein aggregation. This study aimed to identify the structural mechanisms of these mutations. Bioinformatic algorithms were used to predict the association of the E200K mutation with prion disease. Molecular dynamics simulations were performed to confirm the stability of the protein structures. The results showed that the E200K mutation led to increased fluctuations in the protein structure, while the G127V mutation stabilized the protein structure.

Summary: Carbapenem-resistant Acinetobacter baumannii is a common hospital-acquired pathogen with a high mortality rate. Combining beta-lactam antibiotics with beta-lactamase inhibitors can enhance their effectiveness. Through MIC testing and in silico analysis, we identified promising drug combinations, including eravacycline, cefepime/zidebactam, cefiderocol/zidebactam, and eravacycline with durlobactam or zidebactam, for treating A. baumannii infections expressing specific oxacillinases.

Prevalence, predictors, and outcomes of major congenital anomalies: A population-based register study

Summary: This study aimed to assess the incidence, risk factors, and outcomes of major congenital anomalies (CAs) in Qatar. The study found that the incidence of any CA among live births in Qatar was 1.3%, with cardiovascular system (CVS) anomalies being the most common. Multiple pregnancies, parity, and maternal BMI were associated with specific CAs. CAs were significantly associated with high rates of caesarean deliveries, low Apgar scores, in-hospital mortality, and neonatal intensive care unit admission.

Unraveling the Dysbiosis of Vaginal Microbiome to Understand Cervical Cancer Disease Etiology-An Explainable AI Approach

Summary: Microbial dysbiosis is linked to the development of diseases, including cervical cancer, which necessitates the study of vaginal microbiome. This study characterizes the microbial pathogenesis involved in cervical cancer and identifies specific microbial species associated with disease progression. Analysis of diversity, richness, and dominance reveals differences between cervical cancer and control samples, while Linear discriminant analysis Effect Size (LEfSe) predicts the association of certain microbial species and genera with cervical cancer. The results also provide functional enrichment evidence of microbial disease associations. The dataset is trained and validated using a random forest algorithm, and SHapley Additive exPlanations (SHAP) method is employed to analyze the predicted results, highlighting the potential significance of Ralstonia in predicting cervical cancer. The study confirms the presence of pathogenic microbiomes and their relationship with microbial imbalance in cervical cancer vaginal samples.

Airway and Oral microbiome profiling of SARS-CoV-2 infected asthma and non-asthma cases revealing alterations-A pulmonary microbial investigation

Summary: New evidence reveals the role of host-associated microbiomes in respiratory diseases and how their dysbiosis affects lung behavior and respiratory system functioning. Various exogenous and environmental factors impact the development of asthma and chronic lung disease. The relationship between asthma and microbes is better understood but still requires further investigation. The study analyzes sequencing data from the saliva and nasopharyngeal regions of asthma and non-asthma patients infected with SARS-CoV-2, focusing on microbial pathogenesis, exacerbation of respiratory disease, and potential diagnostic biomarkers. Analysis of the data identifies specific microbial taxa enriched in different groups and demonstrates the efficacy of a random forest algorithm for classification and identification of subgroups.

Effect of Biogenic Selenium Nanocomposite on Okra and Metagenomic Analysis of Bacterial Community Associated with Rhizosphere and Phyllosphere

Summary: This study investigated the effects of bio-synthesized Se nanocomposite on the metabolic pathway in Okra leaf and soil compared to bio-fertilizer. Se nanocomposites with an average diameter of 220-330 nm were synthesized and characterized. Metagenomic sequencing revealed changes in the microbial community and metabolic functions in Okra. Compared to biofertilizer-treated Okra, Se treatment significantly increased metabolism in Okra root. Furthermore, exclusive effects were observed in Se-treated Okra roots compared to Se-treated Okra leaves and biofertilizer-treated Okra. Overall, this study enhances our understanding of the impact of Se nanocomposites and biofertilizers on Okra.
