Circulating MicroRNAs: Biogenesis and Clinical Significance in Acute Myocardial Infarction

Acute myocardial infarction (AMI) causes many deaths around the world. Early diagnosis can prevent the development of AMI and provide theoretical support for the subsequent treatment. miRNAs participate in the AMI pathological processes. We aim to determine the early diagnostic and the prognostic roles of circulating miRNAs in AMI in the existing studies and summarize all the data to provide a greater understanding of their utility for clinical application. We reviewed current knowledge focused on the AMI development and circulating miRNA formation. Meanwhile, we collected and analyzed the potential roles of circulating miRNAs in AMI diagnosis, prognosis and therapeutic strategies. Additionally, we elaborated on the challenges and clinical perspectives of the application of circulating miRNAs in AMI diagnosis. Circulating miRNAs are stable in the circulation and have earlier increases of circulating levels than diagnostic golden criteria. In addition, they are tissue and disease-specific. All these characteristics indicate that circulating miRNAs are promising biomarkers for the early diagnosis of AMI. Although there are several limitations to be resolved before clinical use, the application of circulating miRNAs shows great potential in the early diagnosis and the prognosis of AMI.

Long noncoding RNA NONMMUT015745 inhibits doxorubicin-mediated cardiomyocyte apoptosis by regulating Rab2A-p53 axis

Summary: This study demonstrates that doxorubicin induces apoptosis in cardiomyocytes and identified a dysregulated long noncoding RNA (lncRNA), lnc5745, in doxorubicin-treated cardiomyocytes. Overexpression of lnc5745 alleviates cardiomyocyte apoptosis induced by doxorubicin both in vitro and in vivo, while silencing lnc5745 promotes cardiomyocyte apoptosis. Additionally, Rab2A, a direct target of lnc5745, has a protective effect in doxorubicin-induced cardiotoxicity. Mechanistically, Rab2A interacts with p53 and promotes its phosphorylation, thereby activating the apoptotic pathway.

Autophagy regulation in teleost fish: A double-edged sword

Summary: Autophagy is a conserved cellular process essential for maintaining metabolism and energy balance, with implications in various physiological functions and clinical diseases. However, little is known about autophagy in teleost fish. This review provides an overview of the current knowledge of autophagy in teleosts, focusing on its physiological and pathological regulation in different processes. The potential options for targeting autophagy in teleosts for domestication, farming, or therapeutic strategies are also explored.

Noncoding RNAs (ncRNAs), including microRNAs (miRNAs), long noncoding RNAs (lncRNAs) and circular RNAs (circRNAs), play essential roles in modulating pyroptosis and affecting various hallmarks of cancer. This review provides an overview of the complex roles of ncRNAs in regulating pyroptosis, with a focus on their underlying mechanisms and potential applications as biomarkers and therapeutic targets in cancer.

Nanomaterial-based CT contrast agents and their applications in image-guided therapy

Summary: Computed tomography (CT) is a widely used diagnostic tool in hospitals worldwide, but it faces limitations in distinguishing soft tissues from normal organs and tumors. To improve imaging sensitivity and contrast, various nanomaterial-based CT contrast agents have been developed. In this review, we discuss CT contrast agents based on gold (Au), bismuth (Bi), lanthanide (Ln), and transition metal (TM) nanomaterials, along with their applications in image-guided therapy. Additionally, we summarize the use of CT image-guided therapy for various diseases and highlight the future perspectives of CT contrast agents and their biomedical applications.

Construction of rolling circle amplification products-based pure nucleic acid nanostructures for biomedical applications

Summary: Nucleic acid nanomaterials, which have good biocompatibility, biodegradability, and programmability, are often combined with inorganic nanomaterials to improve their biological stability. However, the undefined toxic side effects of composite nanocarriers hinder their in vivo application. Recently, nanostructures composed entirely of DNA oligonucleotides have been rapidly developed as a nanotool to avoid potential biotoxicity and have been applied in biomedicine. This review discusses the assembly strategies of pure nucleic acid nanomaterials based on rolling circle amplification products and their applications in biosensing, bioimaging, and biomedicine.

miR-124 inhibits cardiomyocyte apoptosis in myocardial ischaemia-reperfusion injury by activating mitochondrial calcium uniporter regulator 1

Summary: Mitochondria-mediated apoptosis is the primary cause of cardiomyocyte death. MCUR1 and miR-124 play important roles in regulating cardiomyocyte apoptosis. This study shows that miR-124 can inhibit cardiomyocyte apoptosis by activating MCUR1, suggesting their potential as biomarkers for myocardial injury and infarction.

The potential regulatory role of the lncRNA-miRNA-mRNA axis in teleost fish

Summary: Research in the past two decades has confirmed that noncoding RNAs (ncRNAs), present in cells from yeast to vertebrates, are functional regulators involved in cellular and physiological processes. Dysregulation of ncRNAs is linked to imbalances in cellular homeostasis and the development of diseases. In mammals, ncRNAs like long noncoding RNAs (lncRNAs) and microRNAs (miRNAs) serve as biomarkers and intervention targets in growth, development, immunity, and disease progression. The lncRNA-miRNA-mRNA axis, in which lncRNAs act as competing endogenous RNAs (ceRNAs), is the predominant mode of lncRNA-miRNA crosstalk. However, the role and mechanism of the lncRNA-miRNA-mRNA axis in teleost species have received little attention. This review provides an overview of the physiological and pathological regulation of the teleost lncRNA-miRNA-mRNA axis in growth and development, reproduction, skeletal muscle, immunity to bacterial and viral infections, and stress-related immune responses. It also explores the potential application of the lncRNA-miRNA-mRNA axis in the aquaculture industry.

Protein-Gold Nanoclusters Loaded with Ferrous Ions for Inducing Ferroptosis of Hepatoma Cells

CRISPR/Cas9 therapeutics: progress and prospects

However, effectively delivering the CRISPR system to diseased cells in vivo is currently lacking, and nonviral vectors with target recognition functions may be the focus of future research. Pathological and physiological changes resulting from disease onset can be used for targeted delivery or gene editing.
