Bushen-Tiansui Formula Improves Cognitive Functions in an Aβ1-42 Fibril-Infused Rat Model of Alzheimer's Disease

Bushen-Tiansui Formula (BTF) was empirically updated from a classical prescription named Kong-Sheng-Zhen-Zhong pill. It is based on the traditional Chinese medicine theory of the mutual relationship between the brain and the kidney and is intended to treat neurodegenerative diseases. This formulation has been used for several years to treat patients with Alzheimer's disease- (AD-) like symptoms in our clinical department. However, the medicinal ingredients and the mechanisms by which BTF improves cognition and memory functions have not been characterized. In this study, we used UPLC-MS to generate a chromatographic fingerprinting of BTF and identified five possible active ingredients, including stilbene glycoside; epimedin A1, B, and C; and icariin. We also showed that oral administration of BTF reversed the cognitive defects in an A beta(1-42)fibril-infused rat model of AD, protected synaptic ultrastructure in the CA1 region, and restored the expression of BDNF, synaptotagmin (Syt), and PSD95. These effects likely occurred through the BDNF-activated receptor tyrosine kinase B (TrkB)/Akt/CREB signaling pathway. Furthermore, BTF exhibited no short-term or chronic toxicity in rats. Together, these results provided a scientific support for the clinical use of BTF to improve learning and memory in patients with AD.

Summary: This study investigated the effects of Bushen Tiansui formula (BSTSF) on cognitive performance and transfer RNA-derived small RNAs (tsRNAs) in a rat model of Alzheimer's disease (AD). The results showed that BSTSF treatment improved learning and memory deficits in AD rats and regulated the expression of tsRNAs in the hippocampus. Additionally, bioinformatic analyses indicated that the BSTSF treatment-related tsRNAs may have therapeutic roles in AD through various signaling pathways and biological functions.

Systematic Pan-Cancer Analysis Identifies TREM2 as an Immunological and Prognostic Biomarker

Summary: TREM2 is highly expressed in most cancers and is associated with prognosis, tumor mutational burden, microsatellite instability, and DNA methylation. In 6 selected tumor types, TREM2 gene expression was negatively correlated with most immune cell infiltration levels but positively correlated with M1 and M2 macrophages infiltration levels. The correlation with TREM2 expression differed according to T cell subtype.

Summary: A novel GI-related miRNA signature (GIMiSig) was developed to stratify gastric cancer patients into distinct risk groups, with different survival outcomes and immunotherapy efficacy. The high-risk group showed decreased UBQLN4 expression, higher immune cell accumulation, lower TTN mutation frequency, worse immunotherapy efficacy, and cancer-associated pathways, while the low-risk group exhibited UBQLN4 overexpression, lower immune cell fraction, higher TTN mutation frequency, better response to immunotherapy, and GI-related pathways. The results may provide new clues for improving gastric cancer outcomes.

Systematic Pan-Cancer Analysis of KLRB1 with Prognostic Value and Immunological Activity across Human Tumors

The results show that KLRB1 is downregulated in multiple cancers while upregulated in kidney cancer. Patients with high expression of KLRB1 have better prognosis in most cancer types, and the expression of KLRB1 is associated with tumor immune signatures and chemotherapeutic drug sensitivity.

Gene set enrichment analysis revealed enrichment of the lncRNAs in CRC-related signalling pathways, supporting their potential as biomarkers for prognosis prediction in CRC.
