Heavy metal pollution and risk assessment by the battery of toxicity tests

The current study was carried out on dominant fish Oreochromis niloticus and water collected from the polluted Yamuna River, Agra, India. The heavy metals in water, recorded as follows: Fe>Mn>Zn>Cu>Ni>Cr>Cd and all were found to be above the prescribed limits. According to metal pollution index, exposed muscle (49.86), kidney (47.68) and liver (45.26) have been recorded to have higher bioaccumulation. The blood biochemical analysis of exposed O. niloticus indicated significant increase in activities of aspartate aminotransferase (+343.5%), alkaline phosphatase (+673.6%), alanine aminotransferase (+309.1%), and creatinine (+494.3%) over the reference. However, a significant decrease in albumin (A): globulins (G) ratio (-87.86%) was observed. Similarly, the exposed fish also showed significant increase in total leucocyte count (+121%), differential leucocyte count, respiratory burst (+1175%), and nitric oxide synthase (+420%). The histological examination of liver and kidney showed tissue injury. Moreover, micronuclei (0.95%), kidney shaped nuclei (1.2%), and lobed nuclei (0.6%) along with DNA damage in the form of mean tail length in the liver (20.7 mu m) and kidney (16.5 mu m) was observed in the exposed O. niloticus. Potential health risk assessments based on estimated daily intake, target hazard quotient, hazard index, and target cancer risk indicated health risks associated with the consumption of these contaminated fishes. In conclusion, the present study showed that exposure to heavy metals contaminated water can alter immunological response; induce histopathological alterations and DNA damage in the studied fish. The consumption of this contaminated water or fish could have serious impact on human health.

Redox Status, Immune Alterations, Histopathology, and Micronuclei Induction in Labeo rohita Dwelling in Polluted River Water

Summary: In this study, various parameters of oxidative stress, immune response, and abnormalities in the erythrocyte nucleus of Labeo rohita inhabiting the polluted Kshipra River, India, were measured. The heavy metal concentrations in the river water were in the order of Ni > Fe > Cd > Cr > Mn > Zn > Cu. The fish showed increased levels of antioxidant enzymes in gills, liver, and gut compared to the reference fish, but decreased levels of GST and GSH. Heavy metals also caused immunosuppression and histopathological changes in the fish. These findings highlight the potential impact of heavy metal-induced stress on fish inhabiting polluted river water and the importance of pollution control.

In this study, the effects of CTZ on hen egg white lysozyme (HEWL) aggregation were examined using various methods. The results showed that CTZ can reduce the formation of amyloid fibrils and prevent aggregation.

Structure-guided identification of potent inhibitors of ROS1 kinase for therapeutic development against non-small cell lung cancer

Summary: Proto-oncogene tyrosine-protein kinase ROS (ROS1) is a member of a receptor family that affects epithelial cell differentiation and is highly expressed in various tumors. This study identified two potential inhibitors of ROS1 through virtual screening of phytochemicals. The findings highlight the role of ROS1 in multiple physiological processes and suggest the potential use of phytochemicals for therapeutic development targeting ROS1-associated non-small cell lung cancer.

Computational investigation of the impact of potential AT2R polymorphism on small molecule binding

Summary: The renin-angiotensin system (RAS) is crucial for regulating arterial pressure and fluid balance, and the angiotensin type 2 receptor (AT2R) plays a protective role in pathophysiological conditions. This study investigated the impact of single nucleotide polymorphisms (SNPs) on AT2R and its interactions with agonist and antagonist molecules. Computational analysis and molecular dynamics simulations revealed that certain mutations affect the protein conformation and ligand binding. These findings have implications for the development of better medications and future in vitro and in vivo studies.

Summary: This study aimed to identify potential new drug targets/candidates for multi-drug-resistant Mycobacterium tuberculosis (Mtb) through computational techniques. The protein structure of ribonuclease VapC2 of Mtb H37Rv was analyzed, and docking studies were performed with 22 anti-TB drugs. Rifampicin showed the best binding energy and was selected as the parent molecule for designing derivatives. One of the derivatives, RIF-155841, showed activity in inhibiting VapC2. Molecular dynamics simulations confirmed the stability of the VapC2-RIF155841 complex, and a chemical synthesis scheme was designed for future validation.

Assessment of the Anticancer Effect of Chlorojanerin Isolated from Centaurothamnus maximus on A549 Lung Cancer Cells

Summary: The aim of this study was to evaluate the anticancer efficacy of chlorojanerin against different cancer cells. The effects of chlorojanerin on cell cytotoxicity, cell cycle arrest, and cell apoptosis were investigated using various assays. RT-PCR was used to analyze the expression levels of apoptosis-related genes. Docking simulations were employed to study the binding preferences of chlorojanerin with Bcl-2. The results showed that chlorojanerin inhibited cell proliferation dose-dependently and had a promising effect against A549 lung cancer cells. The inhibition of cell growth was associated with G2/M phase cell cycle arrest and apoptosis induction. We also found that chlorojanerin altered the expression of genes involved in apoptosis initiation and could fit into the active site of Bcl-2 according to docking simulations. These findings suggest that chlorojanerin has potential as a therapeutic agent in lung cancer treatment.

Cathepsin-B inhibitor CA-074 attenuates retinopathy and optic neuritis in experimental autoimmune encephalomyelitis induced in SJL/J mice

Summary: This study explored the therapeutic outcome of Cathepsin B inhibitor (CA-074) against retinopathy and optic neuritis caused by experimental autoimmune encephalomyelitis (EAE). The findings showed that CA-074 could ameliorate optic neuritis and retinopathy by reducing inflammation, as well as reducing demyelination and axonal injuries in EAE mice.

Study of Amiloride Binding to Human Serum Albumin: Insights from Thermodynamic, Spectroscopic, and Molecular Docking Investigations

Summary: This study investigates the interaction between the sodium channel blocker amiloride and human serum albumin. The results show that amiloride specifically binds to a specific site on HSA and the complex is stabilized by van der Waals hydrogen-bonding and hydrophobic interactions. FRET analysis demonstrates that the distance between amiloride and Trp214 is crucial for efficient quenching. UV-Vis spectroscopy and circular dichroism indicate minor structural changes in HSA after binding. Molecular dynamics simulations provide further evidence of stable HSA-AML complex formation.

Structure-based virtual screening and molecular docking approaches to identify potential inhibitors against KIF2C to combat glioma

Summary: In this study, new drug candidates that may function as KIF2C enzyme inhibitors were discovered. Through virtual screening and molecular docking experiments, the best compounds that bind with KIF2C protein were identified and further analyzed for stability and energy contribution. These findings indicate the potential of these compounds to inhibit the biological function of KIF2C and call for further research.
