Analysis of Hepatic Fibrosis Characteristics in Cirrhotic Patients with and without Hepatocellular Carcinoma by FTIR Spectral Imaging

The evolution of cirrhosis is marked by quantitative and qualitative modifications of the fibrosis tissue and an increasing risk of complications such as hepatocellular carcinoma (HCC). Our purpose was to identify by FTIR imaging the spectral characteristics of hepatic fibrosis in cirrhotic patients with and without HCC. FTIR images were collected at projected pixel sizes of 25 and 2.7 mu m from paraffinized hepatic tissues of five patients with uncomplicated cirrhosis and five cirrhotic patients with HCC and analyzed by k-means clustering. When compared to the adjacent histological section, the spectral clusters corresponding to hepatic fibrosis and regeneration nodules were easily identified. The fibrosis area estimated by FTIR imaging was correlated to that evaluated by digital image analysis of histological sections and was higher in patients with HCC compared to those without complications. Qualitative differences were also observed when fibrosis areas were specifically targeted at higher resolution. The partition in two clusters of the fibrosis tissue highlighted subtle differences in the spectral characteristics of the two groups of patients. These data show that the quantitative and qualitative changes of fibrosis tissue occurring during the course of cirrhosis are detectable by FTIR imaging, suggesting the possibility of subclassifying cirrhosis into different steps of severity.

Summary: This study utilized infrared spectral imaging (IRSI) to investigate the expression and distribution changes of glypicans in different hair follicle compartments during various phases of the hair growth cycle, providing a new perspective on hair shaft growth. Hepatotoxicity of metronidazole in Cockayne syndrome: A clinical report

Summary: Cockayne syndrome (CS) is a rare autosomal recessive genetic disorder characterized by growth failure and multisystem dysfunction. A case study reported here presents a 21-year-old CS patient who developed jaundice following treatment with metronidazole (MTZ) combined with spiramycin for dental care. Liver biopsy showed portal and lobular inflammation with severe cholestasis, but no bile duct damage. Jaundice regressed over 6 weeks. This case underscores the importance of strictly avoiding the prescription of MTZ in CS patients.

Summary: This study analyzed the progressive development of cirrhosis in two mouse models using Fourier transform infrared spectral histology. Summary: This study aimed to investigate the occurrence of alveolar echinococcosis (AE) in transplant patients. The research found that AE had a higher mortality and faster clinical progression in transplant patients, possibly due to the reactivation of latent liver lesions through immunosuppression, and suggested the use of Western blot serology as the preferred diagnostic method. Surgery had a low success rate and high mortality, while conservative treatment with albendazole was well tolerated.

Exploration of utility of combined optical photothermal infrared and Raman imaging for investigating the chemical composition of microcalcifications in breast cancer

Summary: Microcalcifications play a key role in cancer detection, but linking their morphology, composition, and the nature of a specific breast lesion is challenging. By utilizing vibrational spectroscopy and multiphoton imaging techniques, we explored the composition of microcalcifications in depth. Through O-PTIR and Raman spectroscopy, we confirmed the presence of carbonate ions in microcalcifications for the first time, and multiphoton imaging allowed us to create stimulated Raman histology (SRH) images that provide chemical information similar to histological images. In conclusion, we established an efficient protocol for analyzing microcalcifications through iterative refinement of the area of interest.
