In the last decades, finding new methods to preserve the quality of wheat during post-harvest storage has become of primary importance to guarantee high-quality food products. Attacks by rodents and microorganisms, as well as humidity and high temperatures, are well-known factors affecting the qualitative and quantitative features of stored grains. The use of a controlled atmosphere saturated with different gases (such as carbon dioxide-CO2 and nitrogen-N-2) is known to decrease the growth of microorganisms. Furthermore, the use of N-2 is free of pollutants, thus being an environmental-friendly method. In this study, the use of a controlled atmosphere saturated with N-2 was investigated as a method to preserve the nutraceutical qualities in two old Italian wheat cultivars (i.e. Verna and Cappelli). In particular, vitamin E (alpha-tocopherol), polyphenols and antiradical activity were quantified to highlight the differences between traditional storage methods and the use of a controlled atmosphere saturated with N-2. The data obtained showed that the use of a N-2-saturated atmosphere reduced the loss of functional molecules, especially vitamin E, in both cultivars, during long storage periods. Moreover, the present system is suitable for the storage of seeds of old varieties cultivated under organic agriculture. Overall, the results validate the efficacy of the storage method with N-2 and suggest its possible use for wheat on an industrial scale. (C) 2020 Elsevier Ltd. All rights reserved.

Computational Analysis of Thermal Adaptation in Extremophilic Chitinases: The Achilles' Heel in Protein Structure and Industrial Utilization

Dale L. Ang, Mubasher Zahir Hoque, Md. Abir Hossain, Gea Guerriero, Roberto Berni, Jean-Francois Hausman, Saleem A. Bokhari, Wallace J. Bridge, Khawar Sohail Siddiqui

Summary: This study analyzed amino acid sequences and structures of chitinases from different temperature-adapted organisms using computational and molecular dynamics simulation methods. The more stable mesophilic and thermophilic chitinases were found to have fewer and/or shorter loops, oligomerization, and less flexible surface regions, while colder-adapted chitinases had longer and more flexible surface loops. No consistent trends were observed between stability and amino acid composition or structural features, indicating unique elements affecting stability in different chitinases.

Summary: Histochemistry is a vital analytical tool in plant science for deciphering physiological processes, studying cell structures, and localizing biomolecules and organelles. It is also used to study plant responses to environmental stresses by detecting reactive oxygen species. Additionally, histochemical techniques allow for the identification of specific cell components and the in vivo localization of promoters in different tissues.

Summary: Stinging nettle is a multi-purpose plant with functional ingredients of medicinal interest in its leaves and roots, and valuable extraxylary fibers in its stems. The stem of nettle serves as a useful model for cell wall research, with differences in the occurrence and distribution of cell wall polysaccharides and proteins between young and older internodes, particularly evident in the bast fibers.

Identification of Novel Candidate Genes Involved in Apple Cuticle Integrity and Russeting-Associated Triterpene Synthesis Using Metabolomic, Proteomic, and Transcriptomic Data

Christelle M. Andre, Gea Guerriero, Marc Lateur, Sophie Charton, Celine C. Leclercq, Jenny Renaut, Jean-Francois Hausman, Sylvain Legay

Summary: This study investigated the molecular events and gene expression changes associated with russeting in apple skin. Using metabolomics, proteomics, and transcriptomics data, the study revealed lower expression of cuticle biosynthesis genes and alteration of specialized metabolism pathways in russet apple. A lipid transfer protein was identified as a novel player in cuticle formation, and a diverse range of triterpene-hydroxycinnamates were found to accumulate in russeted tissues. This study enhances our understanding of the interaction between triterpene and suberin pathways.

Biochemical and cytological interactions between callose synthase and microtubules in the tobacco pollen tube

Luigi Parrotta, Claudia Faleri, Cecilia Del Casino, Lavinia Mareri, Iris Aloisi, Gea Guerriero, Jean-Francois Hausman, Stefano Del Duca, Giampiero Cai

Summary: This article investigates the association between callose synthase and cytoskeleton in the pollen tube. The study confirms this association and demonstrates colocalization using immunogold labeling. The results suggest that callose synthase interacts with microtubules and exerts its activity in cooperation with the microtubular cytoskeleton.

Silicon reduces zinc absorption and triggers oxidative tolerance processes without impacting growth in young plants of hemp (Cannabis sativa L.)

Marie Luyckx, Jean-Francois Hausman, Gea Guerriero, Stanley Lutts

Summary: Hemp (Cannabis sativa L.) is a promising crop for non-food agricultural production on soils contaminated by moderate doses of heavy metals. Silicon, as a beneficial element, is reported to improve stressed plant behavior. This study found that applying silicon to Cannabis sativa plants reduced zinc absorption and alleviated zinc-induced membrane lipid peroxidation, while increasing total antioxidant activities.

Impact of Pseudomonas sp. SVB-B33 on Stress- and Cell Wall-Related Genes in Roots and Leaves of Hemp under Salinity

Roberto Berni, Jean-Francois Hausman, Silas Villas-Boas, Gea Guerriero

Summary: Salinity negatively affects plant growth and development. Textile hemp shows salt stress sensitivity in a genotype- and developmental stage-dependent manner. In this study, the effects of a Pseudomonas isolate on salt-stressed hemp were investigated, and the application of both living and dead bacteria had impacts on root biomass and stress-related gene expression. These findings provide insights for future research on enhancing hemp's resilience to exogenous stresses through the use of Pseudomonas sp. SVB-B33 and silica.
