Communications among cells can be achieved either via direct interactions or via secretion of soluble factors. The emergence of extracellular vesicles (EVs) as entities that play key roles in cell-to-cell communication offer opportunities in exploring their features for use in therapeutics; i.e., management and treatment of various pathologies, such as those used for cancer. The potential use of EVs as therapeutic agents is attributed not only for their cell membrane-bound components, but also for their cargos, mostly bioactive molecules, wherein the former regulate interactions with a recipient cell while the latter trigger cellular functions/molecular mechanisms of a recipient cell. In this article, we highlight the involvement of EVs in hallmarks of a cancer cell, particularly focusing on those molecular processes that are influenced by EV cargos. Moreover, we explored the roles of RNA species and proteins carried by EVs in eliciting drug resistance phenotypes. Interestingly, engineered EVs have been investigated and proposed as therapeutic agents in various in vivo and in vitro studies, as well as in several clinical trials.

Kinetics of D-lactate and ischemia-modified albumin after abdominal aortic surgery and their ability to predict intestinal ischemia

Marie Tomandlova, Tomas Novotny, Robert Staffa, Jindra Smutna, Tomas Krivka, Peter Kruzliak, Ondrej Slaby, Lubos Kubicek, Robert Vlachovsky, Lenka Radova, Josef Tomandl

Summary: This study investigates the perioperative kinetics of D-lactate and ischemia-modified albumin (IMA) and their predictive value in early diagnosis of acute intestinal ischemia. The results suggest that serum levels of IMA and D-lactate can serve as influential predictive markers for postoperative intestinal ischemia, especially after 3 hours from complete declamping of vascular reconstruction.

Co-delivery of gemcitabine and salinomycin in PEGylated liposomes for enhanced anticancer efficacy against colorectal cancer

Lucia Ruxandra Tefas, Ioana Toma, Alina Sesarman, Manuela Banciu, Ancuta Jurj, Ioana Berindan-Neagoe, Lucia Rus, Rares Stiufiuc, Ioan Tomuta

Summary: Colorectal cancer is a major cause of illness and death worldwide, and targeted elimination of both cancer stem cells (CSCs) and non-CSCs is essential for successful treatment. This study developed and optimized a liposomal drug delivery system for co-loading the anti-CSC drug salinomycin (SAL) and the conventional anticancer drug gemcitabine (GEM). The co-loaded liposomes demonstrated sustained release of both drugs and superior in vitro cytotoxic effects compared to single GEM treatment, indicating that this combination therapy may be a promising strategy for treating colorectal cancer.

Summary: Cardiac-urogenital syndrome is a rare syndrome associated with pathogenic variants in the MYRF gene, leading to loss of protein function. It is characterized by complex congenital cardiac defects, pulmonary hypoplasia, diaphragmatic hernia, and urogenital malformations. We report a case of two siblings with extensive developmental defects, who were found to have a novel splicing pathogenic variant in the MYRF gene. The presence of the identical de novo event in siblings suggests gonadal mosaicism, which mimics recessive inheritance. This is the first published case indicating gonadal mosaicism in familial cardiac-urogenital syndrome.

Summary: This case report describes a 43-year-old female patient with frontal glioblastoma who developed intraspinal metastasis five months after surgery. Next-generation sequencing analysis revealed a higher number of single nucleotide variants in the metastatic sample compared to the primary tumor. Mutations in NF1, NOTCH3, and ARID1A were identified and may contribute to the invasiveness and metastatic potential of this particular glioblastoma case.

Discovering the Biological Significance and Therapeutic Potential of miR-29b-3p in Triple-Negative Breast Cancer

Summary: The lack of estrogen or progesterone receptors and absence of HER2 amplification/overexpression limit treatment options for triple-negative breast cancer (TNBC). MicroRNAs (miRNAs), particularly miR-29b-3p, have been of interest for TNBC and correlated with overall survival rates. This study explored the potential therapeutic effects of miR-29b-3p inhibition in TNBC cell lines, observing decreased cell proliferation, activation of apoptosis and autophagy, and altered miRNA expression patterns. Target analysis revealed ECM receptor interaction and TP53 signaling as the main predicted targets, with additional validation showing upregulation of MCL1 and TGFB1.

Summary: This review focuses on the functional and tumor progression role of miRNA-3652 in ovarian cancer. miRNA-3652 is involved in regulating ovarian cancer cell proliferation, migration, and invasion, and its dysregulation is associated with altered expression of critical genes involved in tumor growth and metastasis. Additionally, miRNA-3652 expression levels are correlated with clinical stages, prognosis, and response to therapy in ovarian cancer patients.

The Effect of Different Anesthetic Techniques on Proliferation, Apoptosis, and Gene Expression in Colon Cancer Cells: A Pilot In Vitro Study

Summary: This study aimed to compare the effects of different anesthetic drugs on postoperative outcomes in colorectal cancer patients and investigate the potential role of intravenous lidocaine on colon cancer cell functions. The results showed that different anesthesia techniques did not significantly affect apoptosis, migration, and cell cycle of colorectal cancer cells. However, the propofol group showed significantly increased TP53 gene expression, while the lidocaine with sevoflurane group showed increased cell viability. Further studies are needed to explore the effects of propofol and lidocaine on different colon cancer cell lines and assess the clinical implications.

LncRNA SNHG6 role in clinicopathological parameters in cancers

Khushbukhat Khan, Muhammad Irfan, Areej Abdul Sattar, Manal Bint Faiz, Anees Ur Rahman, Hafsa Athar, Daniela Calina, Javad Sharifi-Rad, William C. Cho

Summary: RNA sequencing has shown that a large part of the human genome undergoes transcription, but only a small fraction of these transcripts translate into proteins. LncRNA SNHG6, once considered as transcriptional noise, has now been recognized as a crucial regulator in various cellular processes and cancer development. This review explores the biogenesis, roles, and mechanisms of SNHG6 in cancer, highlighting its potential as a biomarker and therapeutic target.

Summary: This study investigated whether sublobectomy was comparable to lobectomy in elderly patients (>= 75 years old) with peripheral solid-dominant and diameter <= 2 cm non-small cell lung cancer (NSCLC). The results showed that sublobectomy was associated with a shorter operation time and had equivalent survival outcomes compared to lobectomy.
