Low lethal doses of Streptococcus iniae caused enteritis in Siberian sturgeon (Acipenser baerii)

In aquaculture, the incidence of enteritis due to Streptococcus iniae infection in Siberian sturgeon (Acipenser baerii) has increased in recent years. The pathogenesis of S. iniae is largely unknown due to the paucity of experimental studies on fish intestinal inflammation. In this study, S. iniae infection of A. baerii juveniles was induced by anal intubation of 0.15 mL at a low lethal dose (2 x 10(7) CFU/mL). Intestinal pathology and gene expression studies were conducted within 10 days of the experiment. Histopathological examination showed severe intestinal lesions, inflammatory cell infiltration, intestinal submucosa edema, epithelial cell shedding and necrosis. Predominant symptoms of exudative inflammation, metamorphic inflammation and proliferative inflammation on days 1-3, 4-6, and 7-10 post infection were shown, respectively. Ultrastructural observations also revealed fractured microvilli and shedding on days 4-6. Intestinal villi gradually repaired during the subsequent 7-10 days post infection. Expression of the pro-inflammatory cytokines, tumor necrosis factor and interleukin 1 beta were up-regulated on days 1-3 followed by a significant decrease on day 5, ultimately reaching control levels on day 10 post infection. A similar pattern was shown in mucus cells, involving mucin secretion and expression of the mucin encoding gene, Mucin-2. These results showed the cellular response to S. iniae infection associated with inflammatory genes expression in the Siberian sturgeon.

Summary: This study found that Salvia miltiorrhiza polysaccharide (SMP) can improve the immune function and disease resistance of sturgeon. In vitro experiments showed that SMP could activate the macrophages of sturgeon, while in vivo experiments demonstrated that dietary supplementation with SMP enhanced immune cell activity and resistance to bacterial infection in sturgeon.

Summary: Receptor-interacting serine/threonine-protein kinase 2 (RIPK2) plays an important role in regulating inflammatory response and resisting pathogenic microbial infection. The study identified AbRIPK2 in Acipenser baerii and found that it is highly expressed in the gill and immune tissues. AbRIPK2 was significantly upregulated after infection and cellular stimulation, indicating its potential mechanism in resisting pathogenic infection in Siberian sturgeon.

Transcriptome analysis of the immunomodulatory effects of Salvia miltiorrhiza polysaccharide on hemocyte immune response in Procambarus clarkii

Summary: This study analyzed the immunological effects of Salvia miltiorrhiza polysaccharides (SMP) on the hemocytes of Procambarus clarkii. The results showed that SMP treatment upregulated the activity of total superoxide dismutase (T-SOD), phenoloxidase (PO), and respiratory burst. Differential gene expression analysis revealed that SMP affected crustacean cellular immune-related signaling pathways. Furthermore, diets supplemented with SMP improved the cellular and humoral immunity of crayfish. This study provides molecular evidence for the immunopotentiator effects of plant polysaccharides in crustaceans.

Identification and characterization of high mobility group box 1 and high mobility group box 2 in Siberian sturgeon (Acipenser baerii)

Summary: The study identified HMGB1 and HMGB2 in Acipenser baerii and found that these proteins play important roles in DNA damage repair, immune response, and inflammation. The expression of these proteins was detected in multiple tissues and was significantly induced after infection and PAMPs treatment, suggesting their involvement in the pathogenic infection process. This provides a basis for further understanding the mechanism of enteritis induced by Streptococcus iniae in Acipenser baerii.

Summary: The study found that Salvia miltiorrhiza polysaccharides (SMP) have positive effects on the growth performance, antioxidant capacity, non-specific immunity, and resistance to Streptococcus iniae in hybrid sturgeon. The dietary supplementation of SMP can enhance the antioxidant and non-specific immune capacity of hybrid sturgeon, improve disease resistance, and promote healthy breeding. These findings provide novel recommendations for the application of SMP supplementation in farmed fish.

Transcriptome Revealed the Macrophages Inflammatory Response Mechanism and NOD-like Receptor Characterization in Siberian Sturgeon (Acipenser baerii)

Summary: In this study, the inflammatory response mechanism and characterization of NOD-like receptors (NLRs) family in Siberian sturgeon were investigated using transcriptome analysis. The results showed differential expression of genes related to pattern recognition receptors, adaptor proteins, cytokines, and cell adhesion molecules. The downregulation of NOD-like receptor family CARD domains containing 3-like (NLRC3-like) and upregulation of pro-inflammatory cytokines were observed. Furthermore, 19 NLRs with NACHT structural domains were identified in Siberian sturgeon. This study provides insights into the inflammatory response and NLRs family in teleost fish.

Co-isolated bacteria from septicemia of female Siberian sturgeon broodstock: Lactococcus petauri resisting Aeromonas hydrophila infection

Summary: The rapid development of sturgeon aquaculture has met the market demand for sturgeon products and addressed the issue of declining wild resources. However, infectious deaths after artificial propagation of female Siberian sturgeon pose a threat to sturgeon farming. This study identified Aeromonas hydrophila as the etiological agent causing the infection, while co-isolated Lactococcus petauri showed resistance to A. hydrophila infection. The findings contribute to a better understanding of parental sturgeon infection and provide insights for the prevention of Aeromonas infection through the use of antagonistic strains.
