Some cancers are related to atherosclerotic diseases; therefore, these two types of disease may share some antibody biomarkers in common. To investigate this, a first screening of sera was performed from patients with esophageal squamous cell carcinoma (ESCC) or acute ischemic stroke (AIS) for serological identification of antigens using recombinant cDNA expression cloning (SEREX). The amplified luminescent proximity homogeneous assay-linked immunosorbent assay (AlphaLISA) method, which incorporates glutathione donor beads and anti-human IgG acceptor beads, was used to evaluate serum antibody levels. SEREX screening identified low-density lipoprotein receptor-related protein-associated protein 1 (LRPAP1) as a target antigen of serum IgG antibodies in the sera of patients with ESCC or AIS. Antigens, including recombinant glutathione S-transferase-fused LRPAP1 protein, were prepared to examine serum antibody levels. AlphaLISA revealed significantly higher antibody levels against the LRPAP1 protein in patients with solid cancers such as ESCC and colorectal carcinoma and some atherosclerosis-related diseases such as AIS and diabetes mellitus compared with healthy donors. Correlation analysis revealed that the elevated serum antibody levels against LRPAP1 were associated with smoking, a well-known risk factor for both cancer and atherosclerosis. Serum LRPAP1 antibody is therefore a common marker for the early diagnosis of some cancers and atherosclerotic diseases and may reflect diseases caused by habitual smoking.

Serum Anti-BRAT1 is a Common Molecular Biomarker for Gastrointestinal Cancers and Atherosclerosis

Liubing Hu, Jiyue Liu, Hideaki Shimada, Masaaki Ito, Kazuo Sugimoto, Takaki Hiwasa, Qinghua Zhou, Jianshuang Li, Si Shen, Hao Wang

Summary: Atherosclerosis (AS) and cancers are major global causes of mortality and morbidity. They share common risk factors and researchers have discovered potential serum biomarkers, such as BRAT1-Abs and WDR1-Abs, for AS and GI cancers. BRAT1-Ab shows better predictive potential for GI cancers compared to WDR1-Ab. High levels of BRAT1-Ab are also associated with poor prognosis in esophageal squamous cell carcinoma (ESCC) patients.

Ketone bodies: A double-edged sword for mammalian life span

Issei Tomita, Hiroaki Tsuruta, Mako Yasuda-Yamahara, Kosuke Yamahara, Shogo Kuwagata, Yuki Tanaka-Sasaki, Masami Chin-Kanasaki, Yukihiro Fujita, Eiichiro Nishi, Hideki Katagiri, Hiroshi Maegawa, Shinji Kume

Summary: Accumulating evidence suggests that ketone bodies have health benefits, especially for longevity. However, the role of endogenous ketogenesis in mammalian life span and the safety and efficacy of long-term exogenous ketone body supplementation remain unclear. This study shows that a deficiency in endogenous ketogenesis shortens life span in mice, and this can be prevented by daily ketone body supplementation. It is also found that the timing of ketone body supplementation has different effects on mortality rates in normal and aged mice. In conclusion, endogenous ketogenesis affects mammalian survival, and the administration method and health status are important factors in the effects of ketone body supplementation.

Anthropogenic Pollutants Induce Changes in Peroxyacetyl Nitrate Formation Intensity and Pathways in a Mountainous Background Atmosphere in Southern China

Yu Wang, Tao Liu, Daocheng Gong, Hao Wang, Hai Guo, Minping Liao, Shuo Deng, Huang Cai, Boguang Wang

Summary: This study found that PAN was photochemically formed in the Nanling mountains of South China, with methylglyoxal, acetaldehyde, radicals, and other OVOCs as the main precursors. When polluted air masses invaded the area, anthropogenic aromatics intensified PAN formation. The net PAN formation reduced hydroxyl radical level, impaired local radical cycling, and suppressed local O3 production. These findings deepen our understanding of PAN photochemistry and the impact of anthropogenic intrusions on mountainous regions' background atmosphere.

Summary: Psoriasis, a chronic inflammatory skin disease, is associated with obesity and myocardial infarction. Obesity-induced changes in lipid metabolism promote the differentiation of T helper 17 (Th17) cells, leading to chronic inflammation. This study found that weight loss through diet and exercise decreased Th17 cells and improved psoriasis in an obese patient with type 2 diabetes. This supports the hypothesis that obesity increases Th17 cells and chronic inflammation, contributing to psoriasis and atherosclerosis.

Immunohistochemical and clinicopathological study regarding nardilysin on extramammary Paget's disease

Akihiko Yamaguchi, Toshifumi Takahashi, Takeshi Kato, Toshihiro Tanaka, Eiichiro Nishi, Noriki Fujimoto

Summary: It has been reported that nardilysin (NRDC) is associated with malignancies in a conflicting manner, but its role in cutaneous malignancies has not been investigated. Immunohistochemical staining revealed that NRDC expression is observed in all extramammary Paget's disease (EMPD) cases, while other cutaneous malignancies did not show increased NRDC expression. It was found that NRDC staining was weaker in the marginal parts of EMPD lesions than in the central parts, and tumor cells tended to be distributed beyond the macroscopic skin lesions in these cases, suggesting that decreased NRDC expression in the marginal zones may be associated with the ability of tumor cells to produce the cutaneous manifestation of EMPD.
