Summary: The study demonstrates that treatment with 25HC3S can reduce mortality and decrease plasma levels of liver injury markers in APAP overdosed mouse models. It also shows that 25HC3S treatment may alleviate APAP induced liver injury by up-regulating master signaling pathways and maintaining mitochondrial membrane polarization.

Summary: Sleep insufficiency may lead to metabolic dysregulation and increase the risk of diabetes. Research has found that paternal sleep deprivation can affect the metabolic phenotype of offspring, and this is believed to be due to epigenetic inheritance. Male offspring of sleep-deprived fathers show glucose intolerance, insulin resistance, and impaired insulin secretion.

Summary: Porcine deltacoronavirus (PDCoV) is an emerging enteropathogenic coronavirus in pigs that poses a threat to human health. A derivative of cholesterol called 25-hydroxycholesterol (25HC) has been found to exhibit antiviral effects against PDCoV and its mechanism involves interference with cholesterol metabolism and suppression of TGF-beta 1.

Summary: Metabolic disorders are a growing epidemic disease, mainly caused by environmental factors. This study reveals that DNA hydroxymethylation plays a crucial role in early-life environmental stress-induced metabolic disorders. The findings show that early-life exposure to arsenic leads to glucose intolerance and hepatic lipid accumulation in adulthood, altering the expression of lipid metabolism-related genes and DNA hydroxymethylation reprogramming in the liver.

Summary: The presence of 25-hydroxycholesterol interferes with viruses' entry into target cells by inhibiting cholesterol's lipid-condensing effects, ultimately making the bilayers less rigid. This study suggests that incorporating 25-hydroxycholesterol in plasma membranes or converting some of their cholesterol content into 25-hydroxycholesterol can lead to morphological alterations and promote inhomogeneities in lipid packing, resulting in changes in membrane stiffness.

Summary: This study investigated the effects of Lactobacillus acidophilus NX2-6-fermented yogurt (LA-Y) on lipid and glucose levels in high-fat diet-fed mice. The results showed that LA-Y improved lipid profile, reduced fasting blood glucose levels, and enhanced insulin sensitivity. LA-Y also alleviated glucose metabolism disorders by activating the insulin signaling pathway, reducing inflammation, and promoting glycolysis and gluconeogenesis in the small intestine. Additionally, LA-Y promoted fat browning and increased energy expenditure through adiponectin/AMPK alpha/PGC-1 alpha/UCP1 pathway and mitochondrial biogenesis via adiponectin/AdipoR1/APPL1/AMPK alpha/PGC-1 alpha pathway.

Summary: DNA methylation is a process of adding methyl groups to the DNA molecule, which can alter the function of DNA. The importance and role of DNA methylation vary across different organisms, with mammals showing a strong relationship between DNA methylation and diseases such as cancer and placental development.

Summary: CaMelia, a CatBoost gradient boosting method, predicts missing methylation states based on locally paired intercellular methylation similarity. It significantly improves imputation performance on real single-cell methylation datasets, helping to discover intercellular differentially methylated loci masked by sparsity in raw data. The results demonstrate that CaMelia preserves cell-cell relationships, enhances cell type identification, and improves subpopulation detection.

Summary: UHRF1 SRA can recognize non-mCpG DNA, with contrasting mechanisms for hemi-mCpG and fully-mCpHpG DNA recognition. While the binding affinity for fully-mCpG is weaker, UHRF1 SRA exhibits higher affinity for fully-mCpHpG.

Summary: Efficient antiviral drug discovery is a pressing issue in global public health. Recent studies have shown that 25-hydroxycholesterol (25HC) has broad-spectrum antiviral activity with high efficiency and low toxicity. It restricts viral internalization and disturbs the maturity of viral proteins through multiple mechanisms. Understanding the role of 25HC in innate immunity may contribute to the development of urgently needed broad-spectrum antiviral drugs.
