microRNAs and Markers of Neutrophil Activation as Predictors of Early Incidental Post-Surgical Pulmonary Embolism in Patients with Intracranial Tumors

Venous thromboembolism (VTE) is a common complication of cancer that severely increases morbidity and mortality. Patients with intracranial tumors are more likely to develop VTE than patients with cancers at other sites. Conversely, limited tools exist to identify patients with high thrombotic risk. Upon activation, neutrophils release their content through different mechanisms triggering thrombosis. We explored the ability of microRNAs (miRNAs) and plasma markers of neutrophil activation measured before surgery to predict the risk of early post-surgical pulmonary embolism (PE) in glioma and meningioma patients. We recruited and prospectively followed 50 patients with glioma and 50 with meningioma, 34% of whom in each group developed an early objectively-diagnosed post-surgical PE. We measured miRNA expression and neutrophil markers (cell-free DNA, nucleosomes, calprotectin and myeloperoxidase) before surgery. In glioma patients, we adjusted and validated a predictive model for post-surgical PE with 6 miRNAs: miR-363-3p, miR-93-3p, miR-22-5p, miR-451a, miR-222-3p and miR-140-3p (AUC = 0.78; 95% Confidence Interval (CI) [0.63, 0.94]) and another with cfDNA and myeloperoxidase as predictors (AUC = 0.71; 95% CI [0.52, 0.90]). Furthermore, we combined both types of markers and obtained a model with myeloperoxidase and miR-140-3p as predictors (AUC = 0.79; 95% CI [0.64, 0.94]). In meningioma patients we fitted and validated a predictive model with 6 miRNAs: miR-29a-3p, miR-660-5p, miR-331-3p, miR-126-5p, miR-23a-3p and miR-23b-3p (AUC = 0.69; 95% CI [0.52, 0.87]). All our models outperformed the Khorana score. This is the first study that analyzes the capability of plasma miRNAs and neutrophil activation markers to predict early post-surgical PE in glioma and meningioma patients. The estimation of the thrombotic risk before surgery may promote a tailored thromboprophylaxis in a selected group of high-risk patients, in order to minimize the incidence of PE and avoid bleedings.

Neutrophil Extracellular Traps and Platelet Activation for Identifying Severe Episodes and Clinical Trajectories in COVID-19

Summary: The role of NETs and platelet activation in COVID-19 is not well known. This study aimed to evaluate their impact on clinical trajectories. The levels of cfDNA, CitH3, and P-selectin were found to be associated with disease severity and analytical parameters. Higher levels of CitH3 and P-selectin increased the risk of transitioning from the emergency department to the ICU, while higher levels of P-selectin increased the risk of transitioning from the ward to death. Elevated levels of CitH3, cfDNA, and P-selectin decreased the probability of transitioning from the ward to discharge. These findings suggest that NETs and P-selectin levels can be useful in predicting different clinical trajectories.

Summary: Neutrophils play a crucial role in defense against pathogens through various mechanisms, including the release of neutrophil extracellular traps (NETs). These NETs, composed of DNA, histones, and other molecules, have been implicated in both immune defense and the progression of diseases like cancer. In this review, the focus is on ovarian cancer (OC), exploring the involvement of NETs in metastasis establishment and progression, as well as their role in therapy resistance. Additionally, the potential of NETs as diagnostic and prognostic markers for OC is discussed.

On the Oral Microbiome of Oral Potentially Malignant and Malignant Disorders: Dysbiosis, Loss of Diversity, and Pathogens Enrichment

Summary: This study aims to compare and describe the composition and diversity of the oral microbiome in patients with homogeneous leucoplakia, proliferative verrucous leukoplakia, oral squamous cell carcinoma, and OSCC preceded by PVL. The number of bacterial species was lower in cancer patients, with Fusobacteriota being the dominant group. PVL and PVL-OSCC patients had a higher abundance of Campilobacterota and lower Proteobacteria compared to other groups. This is the first study comparing oral microbiome alterations in these groups, and further research is needed.

Oral cancers preceded by proliferative verrucous leukoplakia exhibit distinctive molecular features

Summary: This study investigated the pathophysiologic differences between proliferative verrucous leukoplakia-associated oral squamous cell carcinoma (PVL-OSCC) and non-PVL-OSCC through transcriptomic and DNA methylation analyses. The results showed that PVL-OSCC patients had lower expression of cancer-related genes and hypermethylation of the promoter region of many genes, suggesting that DNA methylation could be a regulatory mechanism.

Summary: Ovarian cancer has a high prevalence and mortality rate in women, often diagnosed at an advanced stage due to non-specific symptoms. The interaction between cancer cells and other molecules plays a crucial role in the development of the disease. Syndecans, a family of four transmembrane proteins, are dysregulated in various cancers, including ovarian cancer, and show promise as diagnostic and prognostic biomarkers. Additionally, studying these proteins in ovarian cancer could lead to the discovery of new drugs for treatment.

Effect and mechanisms of resveratrol in animal models of ischemic stroke: A systematic review and Bayesian meta-analysis

Summary: This systematic review evaluates the effects and mechanisms of Resveratrol (RSV) in animal models of ischemic stroke. The results show that RSV has beneficial effects on structural and functional outcomes, including reducing infarct size, edema size, BBB impairment, neurofunctional impairment, and improving motor performance. These effects may be attributed to the reduction of oxidative stress, inflammation, and apoptosis. However, further high-quality preclinical research is needed to better inform clinical research.
