Comparative Analysis of Primer-Probe Sets for RT-qPCR of COVID-19 Causative Virus (SARS-CoV-2)

Coronavirus disease 2019 (COVID-19) is a newly emerging human infectious disease caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2, also previously known as 2019-nCoV). Within 8 months of the outbreak, more than 10,000,000 cases of COVID-19 have been confirmed worldwide. Since human-to-human transmission occurs easily and the rate of human infection is rapidly increasing, sensitive and early diagnosis is essential to prevent a global outbreak. Recently, the World Health Organization (WHO) announced various primer-probe sets for SARS-CoV-2 developed at different institutions: China Center for Disease Control and Prevention (China CDC, China), Charite '(Germany), The University of Hong Kong (HKU, Hong Kong), National Institute of Infectious Diseases in Japan (Japan NIID, Japan), National Institute of Health in Thailand (Thailand NIH, Thailand), and US CDC (USA). In this study, we compared the ability to detect SARS-CoV-2 RNA among seven primer-probe sets for the N gene and three primer-probe sets for the Orf1 gene. The results revealed that NIID_2019-nCOV_N from the Japan NIID and ORF1ab from China CDC represent a recommendable performance of RT-qPCR analysis for SARS-CoV-2 molecular diagnostics without nonspecific amplification and cross-reactivity for hCoV-229E, hCoV-OC43, and MERS-CoV RNA. Therefore, the appropriate combination of NIID_2019-nCOV_N (Japan NIID) and ORF1ab (China CDC) sets should be selected for sensitive and reliable SARS-CoV-2 molecular diagnostics.

This rapid biosensor could be important for detecting SARS-CoV-2 NP in the presence of spreading variants.

(S)-5-Methylmellein Isolated from an Endogenous Lichen Fungus Rosellinia corticium as a Potent Inhibitor of Human Monoamine Oxidase A

Summary: This study evaluated the inhibitory activities of lichen fungi against human monoamine oxidases (hMAOs). A compound called (S)-5-methylmellein (5MM) was isolated from an extract and exhibited strong inhibitory activity against hMAO-A. It also showed moderate inhibitory activity against acetylcholinesterase. Molecular docking analysis indicated that 5MM had higher binding affinity for hMAO-A compared to hMAO-B. In silico pharmacokinetic analysis suggests that 5MM has good absorption and permeability properties. These findings suggest that 5MM may be a potential candidate for the treatment of neuropsychiatric disorders.

Versatile role of ACE2-based biosensors for detection of SARS-CoV-2 variants and neutralizing antibodies

Interaction Analysis of the Spike Protein of Delta and Omicron Variants of SARS-CoV-2 with hACE2 and Eight Monoclonal Antibodies Using the Fragment Molecular Orbital Method

Summary: This study proposes a method that combines statistical analysis and molecular quantum mechanics simulations to efficiently predict the binding affinity between SARS-CoV-2 variants and hACE2 or monoclonal antibodies. The method accurately predicts the trend of binding affinity and interaction energy changes, demonstrating its effectiveness in predicting the effects of mutations.

Identification of 3-Oxindole Derivatives as Small Molecule HIV-1 Inhibitors Targeting Tat-Mediated Viral Transcription

Trovafloxacin drives inflammation-associated drug-induced adverse hepatic reaction by changing macrophage polarization

Summary: Drug-induced liver injury (DILI) is a serious concern for public healthcare systems and pharmaceutical companies. This study investigates the mechanism by which DILI drugs alter the activation and phenotype of hepatic macrophages. The findings suggest that proinflammatory M1-like macrophages contribute to hepatotoxicity mediated by DILI drugs, and the interferon type I pathway, cytokines, and apoptosis pathway are involved in the initiation of liver injury. Transcriptional changes in genes related to macrophage polarization could be used as indicators to reflect the severity of DILI during drug development.

Regulation of Circadian Genes Nr1d1 and Nr1d2 in Sex-Different Manners during Liver Aging

Summary: This study aimed to evaluate the effects of age and sex on gene expression and identify sex-specific target molecules in the regulation of aging. The results showed that the expression of genes related to circadian rhythms changed more in males than in females during liver aging, and Nr1d1 and Nr1d2 were upregulated in males. These findings suggest that age-related upregulation of Nr1d1/2 may play a more crucial role in the change in circadian rhythms in males during liver aging.

Summary: Thirteen compounds were isolated from Canavalia lineata pods and their inhibitory activities against hMAO-A and -B were evaluated. Compounds 8 and 13 showed potent inhibitory activity against hMAO-B, while most compounds weakly inhibited MAO-A. Compounds 8 and 13 were reversible competitive inhibitors against hMAO-B.

Ag-ZnO Nanocomposites as a 3D Metal-Enhanced Fluorescence Substrate for the Fluorescence Detection of DNA

Summary: Fluorescence enhancement, including metal-enhanced fluorescence (MEF), has been widely explored in biosensor technology. In this study, a novel 3D MEF substrate using Ag-ZnO nanocomplex was developed, showing significant fluorescence enhancement compared to the glass substrate. The Ag-ZnO nanocomplex exhibited a 50-fold higher sensitivity for the detection of double-stranded DNA compared to a 2D MEF substrate. This research provides a potential platform for the development of sensitive MEF-based biosensors for DNA detection.

A Novel Time-Resolved Fluorescence Resonance Energy Transfer Assay for the Discovery of Small-Molecule Inhibitors of HIV-1 Tat-Regulated Transcription

Summary: Designed a homogeneous time-resolved fluorescence resonance energy transfer (TR-FRET) assay using europium cryptate as a fluorescence donor to investigate the interaction between Tat and TAR RNA for HIV-1 replication. The optimized assay showed specificity and successfully identified two small molecules (460-G06 and 463-H08) capable of inhibiting Tat activity and HIV-1 infection. This assay is suitable for high-throughput screening and the identified compounds may serve as potential molecular scaffolds for new HIV-1 drug development.

A Comparative Inflammation-On-A-Chip with a Complete 3D Interface: Pharmacological Applications in COPD-Induced Neutrophil Migration

Summary: Chronic obstructive pulmonary disease (COPD) is a deadly lung disease with limited drug discovery due to lack of appropriate preclinical models. Researchers have developed a comparative inflammation-on-a-chip (IoC) model that mimics the key feature of COPD, neutrophil transendothelial migration (NTEM). Using this model, the effects of CXCR2 inhibitors on neutrophil migration in the presence of COPD patient samples were evaluated. This IoC model provides a valuable tool for studying COPD severity and advancing research on NTEM.
