Pelvic organ prolapse (POP) is a hidden women's health disorder that impacts 1 in 4 women across all age groups. Surgical intervention has been the only treatment option, often involving non-degradable meshes, with variable results. However, recent reports have highlighted the adverse effects of meshes in the long term, which involve unacceptable rates of erosion, chronic infection and severe pain related to mesh shrinkage. Therefore, there is an urgent unmet need to fabricate of new class of biocompatible meshes for the treatment of POP. This review focuses on the causes for the downfall of commercial meshes, and discusses the use of emerging technologies such as electrospinning and 3D printing to design new meshes. Furthermore, we discuss the impact and advantage of nano-/microstructured alternative meshes over commercial meshes with respect to their tissue integration performance. Considering the key challenges of current meshes, we discuss the potential of cell-based tissue engineering strategies to augment the new class of meshes to improve biocompatibility and immunomodulation. Finally, this review highlights the future direction in designing the new class of mesh to overcome the hurdles of foreign body rejection faced by the traditional meshes, in order to have safe and effective treatment for women in the long term.

Tissue engineering utilizes the electrospinning method to create scaffolds, with the addition of ceramics to enhance their mechanical properties and biological behavior, as well as induce other effects such as improved hydrophilicity, antibacterial and antioxidant properties.

Improved osteoblast function on titanium implant surfaces coated with nanocomposite Apatite-Wollastonite-Chitosan- an experimental in-vitro study

Summary: In this study, a composite coating of ceramic apatite-wollastonite (AW) and polymer chitosan was synthesized by electrophoretic deposition on titanium implants. The coated substrates showed significant positive effects on cell proliferation, growth, and mineralization compared to uncoated titanium samples. The coating was non-toxic to cells and enhanced cell adhesion and protein production.

Platelet lysate can support the development of a 3D-engineered skin for clinical application

Summary: This study found that platelet lysate (PL) can be used as a safe and effective serum alternative for bioengineering skin for cell therapies. PL supports the expansion of fibroblasts and directed epidermal stratification, and promotes better transplantation outcomes by regulating the expression of CD90 and FAP. Moreover, PL is rich in growth factors, including TGF-beta 1, which contributes to skin maturation. This research is of great importance for the development of bioengineered skin.

Subtle changes in perivascular endometrial mesenchymal stem cells after local endometrial injury in recurrent implantation failure

Summary: Improvements in reproductive techniques have increased live birth rates from IVF procedures, but the rate has plateaued recently. Endometrial receptivity deficiencies are identified as the main hindrance for successful implantation in patients with recurrent implantation failure (RIF). The impact of local endometrial injury (LEI) on perivascular endometrial mesenchymal stem/progenitor cells (eMSCs) is unknown, but it may play a role in improving IVF outcomes for RIF patients.

6-Shogaol Exhibits Anti-viral and Anti-inflammatory Activity in COVID-19-Associated Inflammation by Regulating NLRP3 Inflammasomes

Summary: Recent global health concern has led to the investigation of natural medicinal plant resources as alternative treatments for COVID-19 infection and inflammation. In this study, a phytochemical called 6-shogaol was explored as a potential anti-inflammatory and anti-COVID-19 agent. 6-shogaol was found to efficiently inhibit SARS-CoV2 replication and regulate the expression of inflammasome pathway molecules. Compared to existing drugs, 6-shogaol demonstrated similar efficacy and better safety in terms of physicochemical properties and clinical toxicity. This study provides important insights for the further development of 6-shogaol as a potential therapeutic agent against inflammation and viral infections.

New concepts on the etiology of endometriosis

Summary: Endometriosis is a serious and chronic condition where tissue similar to the lining of the uterus grows outside of it, leading to severe pelvic pain and infertility. It affects 11% of women and has multiple causes, with retrograde menstruation being a major factor. There is a genetic component to endometriosis, with over 40 genetic risk factors identified, although their specific mechanisms are still being understood.

Radiotherapy exposure directly damages the uterus and causes pregnancy loss

Summary: Female cancer survivors are at a higher risk of infertility due to the damaging effects of chemotherapy and radiotherapy on the ovaries. However, the effects of cancer treatments on the uterus have been understudied. This study shows that total-body gamma-irradiation (TBI) damages the uterus, leading to compromised pregnancy success. The pro-apoptotic protein Puma is identified as a key mechanism in TBI-induced uterine damage, suggesting that inhibiting PUMA may be a potential fertility preservation strategy.
