Summary: Heat stroke is a devastating pathology with high morbidity and mortality rates. Understanding the cytotoxic mechanisms of heat can help prevent or treat stroke-related diseases. The role of lysosomes in heat stroke is poorly understood due to a lack of tools, but recent studies have shown a correlation between lysosome HClO and heat stroke development. To better interpret and treat heat stroke, a reliable real-time tool for tracking HClO levels is necessary, and MB-HClO, a near-infrared probe, shows promise in rapidly detecting lysosome HClO.

Summary: A water-soluble 1,2,4,5-tetrazine-substituted carbon-dipyrromethene (C-DIPY) was synthesized and showed fluorescence quenching in phosphate-buffered saline. However, the fluorescence could be restored up to 6.6-fold by adding a bicyclo[6.1.0]non-4-yne (BCN) derivative through an inverse electron-demand Diels-Alder reaction. This bioorthogonal activation was also observed in human glioblastoma cells, enhancing the fluorescence intensity of C-DIPY by 8.7-fold.

Summary: Breast cancer has a high incidence rate and mortality rate among women. A second near-infrared (NIR-II) fluorescent probe (BP-A) was developed in this study to detect breast cancer by monitoring the concentration changes of H2S. BP-A showed excellent NIR-II emission and the ability to sensitively and specifically detect H2S in living mice. It also demonstrated the ability to distinguish between healthy mice and those with breast cancer. Therefore, this probe represents a promising and powerful tool for the early clinical detection of breast cancer.

Summary: Steatosis is the underlying cause of non-alcoholic steatohepatitis and chronic kidney disease. Endoplasmic reticulum (ER) stress induced by steatosis leads to kidney injury. This study investigated the therapeutic potential of 5-aminolevulinic acid (5-ALA) in alleviating ER stress and lipotoxicity in human renal cells. The results showed that 5-ALA treatment increased the expression of heme oxygenase-1 (HO-1) and reduced the expression of ER stress markers, thereby protecting against lipotoxicity-induced renal injury.
