Risk Governance of Emerging Technologies Demonstrated in Terms of its Applicability to Nanomaterials

Nanotechnologies have reached maturity and market penetration that require nano-specific changes in legislation and harmonization among legislation domains, such as the amendments to REACH for nanomaterials (NMs) which came into force in 2020. Thus, an assessment of the components and regulatory boundaries of NMs risk governance is timely, alongside related methods and tools, as part of the global efforts to optimise nanosafety and integrate it into product design processes, via Safe(r)-by-Design (SbD) concepts. This paper provides an overview of the state-of-the-art regarding risk governance of NMs and lays out the theoretical basis for the development and implementation of an effective, trustworthy and transparent risk governance framework for NMs. The proposed framework enables continuous integration of the evolving state of the science, leverages best practice from contiguous disciplines and facilitates responsive re-thinking of nanosafety governance to meet future needs. To achieve and operationalise such framework, a science-based Risk Governance Council (RGC) for NMs is being developed. The framework will provide a toolkit for independent NMs' risk governance and integrates needs and views of stakeholders. An extension of this framework to relevant advanced materials and emerging technologies is also envisaged, in view of future foundations of risk research in Europe and globally.

Extra-Pulmonary Translocation of Exogenous Ambient Nanoparticles in the Human Body

Summary: Ambient fine particles, as a major air pollutant, have generated significant concerns regarding public health. Recent studies have made progress in understanding their biological effects and mechanisms. However, challenges remain in investigating exposure pathways, translocation in the human body, and the correlation between environmental exposure concentration and internal fate of particles. Furthermore, more research is needed on particle biotransformation and differentiation from biological macromolecules. This review highlights the current knowledge gaps and proposes future research directions to support fundamental studies and policy making.

Self-Assembled Oleic Acid-Modified Polyallylamines for Improved siRNA Transfection Efficiency and Lower Cytotoxicity

Summary: Small interference RNA (siRNA) is a gene modulation tool that can silence genes associated with genetic disorders. However, the instability of siRNA in the blood and its difficulty in reaching the cytosol limit its therapeutic potential. Polyamine-based nanoparticles, particularly polyallylamine hydrochloride (PAH), show promise in gene delivery. In this study, PAH was chemically modified with oleic acid to improve cell viability and siRNA transfection efficiency. The PAH-oleic acid substitutes were effective in silencing the CD47 protein, making them a potential candidate for immunotherapy.

Design and fabrication of a microfluidic system with embedded circular channels for rotary cell culture

Summary: This article describes the manufacturing and functionalization of a microfluidic device to simulate blood vessels for testing new treatments for pulmonary arterial hypertension. The device is made using a wire with a circular section to determine the channel dimensions, and cells are seeded using rotary cell culture for homogeneous cell seeding on the inner wall. This method provides a simple and reproducible way to generate in vitro blood vessel models.

Sampling strategies and analytical techniques for assessment of airborne micro and nano plastics

This review extracts data from PubMed and Embase databases to analyze sampling strategies, identification methods, and reporting data for quantifying M/NPs. It primarily focuses on different methods for collecting atmospheric M/NPs and the analytical tools used for identification. The review identifies challenges and recommends further method development to support standardized and comparable future research.

Summary: Retrosynthesis is a valuable tool for simplifying the synthesis planning of organic molecules, and computer-assisted methods have enhanced its effectiveness. With the integration of computer technologies, retrosynthesis can now include database mining, reaction prediction, machine learning, and other data science tools, enabling the consideration of inorganic compounds and nanoparticles. The development of greener and more sustainable chemistry is also supported by retrosynthesis, as it allows for the selection of environmentally friendly processes and reactants. Additionally, recent advancements in computer technologies provide a chance to accurately control green and sustainable metrics at an early stage, leading to the emergence of predictive green and sustainable methods.

Towards higher scientific validity and regulatory acceptance of predictive models for PFAS

Comparing [3H] thymidine LPT and CFSE assay to assess lymphocyte proliferation in beryllium-exposed sarcoidosis patients

Summary: The detection of antigen specific lymphocyte responses plays a vital role in diagnosing various diseases. This study evaluated the suitability of the Be-specific LPT and CFSE assay in detecting beryllium sensitivity in sarcoidosis patients with suspected beryllium exposure. The results showed that both LPT and CFSE assay are suitable for detecting beryllium sensitivity in sarcoidosis patients, but CFSE assay has an advantage in identifying the relevant proliferating lymphocyte populations.
