Exercise dose affects the circulating microRNA profile in response to acute endurance exercise in male amateur runners

The systemic response to exercise is dose-dependent and involves a complex gene expression regulation and cross-talk between tissues. This context ARISES the need for analyzing the influence of exercise dose on the profile of circulating microRNAs (c-miRNAs), as emerging posttranscriptional regulators and intercellular communicators. Thus, we hypothesized that different exercise doses will determine specific c-miRNA signatures that will highlight its potential as exercise dose biomarker. Nine active middle-aged males completed a 10-km race (10K), a half-marathon (HM), and a marathon (M). Blood samples were collected immediately before and after races. Plasma RNA was extracted, and a global screening of 752 microRNAs was analyzed using RT-qPCR. Three different c-miRNA profiles were defined according to the three doses. In 10K, 14 c-miRNAs were found to be differentially expressed between pre- and post-exercise, 13 upregulated and 1 downregulated. Regarding HM, 13 c-miRNAs were found to be differentially modulated, in all the cases upregulated. A total of 28 c-miRNAs were found to be differentially expressed in M, 21 overexpressed and 7 repressed after this race. We had also found 3 common c-miRNAs between 10K and M and 2 common c-miRNAs between 10K and HM. In silico analysis supported a close association between exercise dose c-miRNA profiles and cellular pathways linked to energy metabolism and cell cycle. In conclusion, we have observed that different exercise doses induced specific c-miRNA profiles. So, our results point to c-miRNAs as emerging exercise dose biomarkers and as one of regulatory mechanisms modulating the response to endurance exercise.

Summary: This study measured serum iron markers in naturally menstruating and oral contraceptive athletes. The results showed that naturally menstruating athletes had lower levels of serum iron compared to OC athletes during bleeding phase. However, the differences disappeared when comparing the different phases of the menstrual cycle with the withdrawal phase of the OC cycle.

Summary: Although genomes from many edible mushrooms have been sequenced, studies on fungal micro RNAs (miRNAs) are rare. This study investigated the RNA-seq of the white button mushroom and identified potential novel miRNA-like small RNAs that were found in the edible parts. The results suggest that these small RNAs are involved in fungal metabolic regulation and may interfere with important biological processes in humans.

Summary: The implementation of cell-free microRNAs as biomarkers is limited due to lack of reproducibility. Technical variability and analytical caveats make the interpretation of miRNA data challenging. Standardization efforts and consideration of specific aspects in sample handling and data analysis are important for reproducibility. Providing a checklist can facilitate the control of miRNA quantification and analysis.

miR-24-3p and Body Mass Index as Type 2 Diabetes Risk Factors in Spanish Women 15 Years after Gestational Diabetes Mellitus Diagnosis

Summary: The aim of this study was to investigate the glucose tolerance status after 15 years of pregnancy in women diagnosed with gestational diabetes mellitus (GDM) and to assess the long-term effect of GDM on circulating miRNA profile. The study included 30 randomly selected women diagnosed with GDM during 2005-2006, and glucose tolerance was measured using the National Diabetes Data Group criteria. Four miRNAs were analyzed in the plasma of women 15 years after the diagnosis of GDM. The study found that 50% of women had some degree of glucose intolerance 15 years after the diagnosis of GDM, which was directly related to body weight and body mass index during pregnancy. Dysglycemic women also showed significantly increased levels of circulating hsa-miR-24-3p. Thus, initial weight and BMI, along with circulating expression levels of hsa-miR-24-3p, could be good predictors of the future development of dysglycemia in women with a previous diagnosis of GDM.

Hydroxytyrosol decreases EDNRA expression through epigenetic modification in colorectal cancer cells

Summary: The Mediterranean diet is associated with lower incidence of cardio-vascular and cerebrovascular diseases as well as cancer. Extra virgin olive oil, with its phenolic components such as hydroxytyrosol, has been attributed with many healthful effects. Recent studies show that extra virgin olive oil and hydroxytyrosol modulate epigenetic mechanisms, specifically DNA methylation, in human tissues. In this study, colorectal cancer cells treated with hydroxytyrosol demonstrated an increase in global DNA methylation, suggesting a potential gene target for this compound.

Omega-3 and Omega-6 Polyunsaturated Fatty Acid Intakes, Determinants and Dietary Sources in the Spanish Population: Findings from the ANIBES Study

Summary: The dietary intake of omega-3 polyunsaturated fatty acids among the Spanish population is inadequate, especially in children, adolescents, and younger women. The intake of omega-6 polyunsaturated fatty acids is relatively acceptable, but the ratio of omega-6 to omega-3 is high. Meat and meat products are the main source of essential fatty acids, while fish and shellfish are the only sources of EPA and DHA. The combined degree of inadequacy for omega-3 PUFA, folic acid, vitamin B-12, and choline is 21.3% in the ANIBES population.

Circulating lipoprotein-carried miRNome analysis reveals novel VLDL-enriched microRNAs that strongly correlate with the HDL-microRNA profile

Summary: This study examined the expression profiles of miRNAs in lipoprotein fractions (VLDL, LDL, and HDL) isolated from the serum through various methods. The results showed stable detection of different miRNAs in each fraction, with VLDL-specific miRNAs detected for the first time. Functional enrichment analysis indicated the potential roles of lipoprotein-carried miRNAs in cardiovascular disease-related mechanisms.

Single vesicle analysis reveals the release of tetraspanin positive extracellular vesicles into circulation with high intensity intermittent exercise

Summary: Small extracellular vesicles (sEVs) are released and participate in the intercellular exchange of proteins, lipids, metabolites, and nucleic acids. Proteomic, flow cytometry, and nanoparticle tracking analyses suggest that sEVs are released into circulation with exercise. HIIT leads to a decrease in the number of platelets and CD9, CD63, and CD41a positive sEVs in platelet-free plasma, supporting the concept of sEV release with exercise. Platelet-free plasma is the preferred sample to study sEV dynamics and phenotype during exercise.

Nutritional Composition of Breakfast in Children and Adolescents with and without Celiac Disease in Spain-Role of Gluten-Free Commercial Products

Summary: This study assessed the ingredients and nutrient composition of gluten-free and gluten-containing breakfast products, and analyzed the nutritional quality of breakfast in Spanish children and adolescents with celiac disease. The results showed that breakfast in celiac disease patients was balanced in terms of energy and key food groups, but their fruit intake needed improvement. Compared to the control group, breakfast in the celiac disease group had less protein and saturated fat, similar carbohydrates and fiber, and more salt. Overall, breakfast on a gluten-free diet can be adequate but can be improved through product reformulation and reducing consumption of processed foods.

Circulating plasma levels of miR-106b-5p predicts maximal performance in female and male elite kayakers

Summary: This study aimed to analyze the predictive value of plasma miR-106b-5p levels on sports performance in elite female and male kayakers. Results showed that miR-106b-5p levels correlated negatively with performance in men, while the miR-106b-5p/progesterone ratio correlated negatively with performance in women. The study highlights the importance of analyzing the molecular response to exercise separately in men and women, taking into account the stage of the menstrual cycle in women as a relevant factor.
