The genusYoungiaCass. is taxonomically complex and widely distributed in East Asia. Despite the high diversity of species, only two species,Youngia japonica(L.) DC. andYoungia longiflora(Babc. & Stebbins) C.Shih, are currently found in Korea. Among them,Y. longiflorawas once recognized as a subspecies ofY. japonica. However, based on chloroplast and nuclear DNA sequences, as well as morphology,Y. longiflorawas found to be a distinct species. Here, the two KoreanYoungiaspecies were analyzed using scanning electron microscope and light microscope for chromosome number, pollen, cypselae, and leaf micromorphological characters to evaluate if the examined characters are informative for taxonomy. Pollen morphology is described, and the micromorphological characters ofY. longifloraare described for the first time. Cypselae and pollen surface ornamentation, as well as trichome morphology and stomatal size varied between the two species.Y. japonicahad longer chromosomes thanY. longiflora, while other chromosomal characters including the localization of the NOR (nucleolar organizer region) were similar. The present micromorphological and cytological results are of great taxonomic value forYoungiaspecies identification in Korea.

Impact of parasitic lifestyle and different types of centromere organization on chromosome and genome evolution in the plant genus Cuscuta

Pavel Neumann, Ludmila Oliveira, Jana Cizkova, Tae-Soo Jang, Sonja Klemme, Petr Novak, Katarzyna Stelmach, Andrea Koblizkova, Jaroslav Dolezel, Jiri Macas

Summary: The parasitic genus Cuscuta stands out for its unique centromere organization and genome size variations among plants. Monocentric Cuscuta species show a remarkable 102-fold variation in genome sizes, while holocentric species have more moderate sizes. The transition to holocentric chromosomes in Cuscuta is associated with changes in epigenetic marks, chromosome number, and repetitive DNA sequence composition.

Micromorphological differentiation of Korean Disporum species using light and scanning electron microscopy

Hyeonjin Kim, Bokyung Choi, Changyoung Lee, Scott A. M. McAdam, Jin-Hyub Paik, Tae-Soo Jang

Summary: This study provides a detailed description of leaf, floral, pollen, and orbicule micromorphology of four Korean Disporum species using light and scanning electron microscopy. The presence of orbicules and their taxonomic implications in Korean Disporum species are described for the first time, and phylogenetically informative characters are identified to improve understanding of the systematic relationships of Korean species in the genus Disporum.

Cytological Study of Cypripedium japonicum Thunb. (Orchidaceae Juss.): An Endangered Species from Korea

Bokyung Choi, Geun-Hye Gang, Hyeonjin Kim, Hyejoo Byun, Minyeong Kwak, Soonku So, Hyeon-Ho Myeong, Tae-Soo Jang

Summary: The study investigated the chromosome number, karyotype, and genome size of the Korean lady's slipper orchid, finding that the species is diploid with 2n = 22 and x = 11 as the base chromosome number. The karyotypes were similar among individuals and differentiated the Korean population from the Japanese lady's slipper orchid. Results were consistent with Korean populations being more closely related to Chinese populations than Japanese populations, aiding in the discrimination of Eastern Asian lady's slipper orchid species.

Chromosome Evolution in Korean Carduus-Cirsium Taxa (Asteraceae)

Seongyeon Kang, Bokyung Choi, Tae-Soo Jang

Summary: The genus Cirsium is a member of the tribe Cardueae in the Asteraceae family, distributed in the northern hemisphere. There is a history of taxonomic confusion within the Carduus-Cirsium group due to complex morphological and molecular data. This study reports chromosome numbers and karyotypes of Korean Cirsium species, aiding in understanding chromosomal evolution.

Implications of foliar epidermal micromorphology using light and scanning electron microscopy: A useful tool in taxonomy of Korean irises

Bokyung Choi, Yu-Eun Ahn, Tae-Soo Jang

Summary: In this study, the leaf micromorphological characteristics of Korean irises were examined, providing valuable information for species identification. The presence of polymorphic stomatal types was reported for the first time, and the potential correlation between genome size and epidermal guard cell length was discussed.

Can pollen exine ornamentation contribute to species delimitation in Korean Iris L. taxa (Iridaceae)?

Bokyung Choi, Jangseung Ryu, Tae-Soo Jang

Summary: This study used scanning electron microscopy to describe the pollen morphology of Korean Iris. The observed pollen micromorphological characters, including size, shape, and exine ornamentation, enabled species distinction and taxonomic delimitation. The pollen morphological data presented in this study are of systematic significance to Korean Iris species.

Bokyung Choi, Inkyu Park, Soonku So, Hyeon-Ho Myeong, Jangseung Ryu, Yu-Eun Ahn, Kyu-Chan Shim, Jun-Ho Song, Tae-Soo Jang

Summary: This study compared the complete plastid genome sequences and micromorphological features of Iris ruthenica Ker Gawl. and I. uniflora Pall. ex Link, and identified molecular markers for their discrimination. The phylogenetic analysis showed their close relationship, and the comparison of plastomes and micromorphological features provided valuable information for understanding their taxonomic and evolutionary relationships.

A study of the chromosome number and genome size of the rare species Rhododendron keiskei var. hypoglaucum in Korea

Bokyung CHOI, Hyeonjin KIM, Hye-Joo BYUN, Geun-Hye GANG, Yongsoon LEE, Hyeon-Ho MYEONG, Soonku SO, Tae-Soo JANG

Summary: This study reports the chromosome number, karyotype structure, and genome size of Rhododendron keiskei var. hypoglaucum for the first time. The chromosome number of this plant was found to be 2n = 26 with x = 13 as the base chromosome number. The karyotype consisted of metacentric and sub-metacentric chromosomes of similar length. These findings provide important insights for further molecular, cytogenetic, and phylogenomic analyses of Rhododendron.

New Chromosome Counts in Juncus (Juncaceae) Taxa from Korea

Young -Min Choi, Bokyung Choi, Tae -Soo Jang

Summary: Juncus L. is a genus of herbs with approximately 310 species globally, but less commonly found in the tropics. This study presents detailed information on chromosome numbers from 36 individuals of 10 Korean Juncus taxa, revealing variations in chromosome numbers with no systematic relevance. The study suggests that this chromosome number variation could provide additional evidence for morphological diversification.

Disruption of the standard kinetochore in holocentric Cuscuta species

Pavel Neumann, Ludmila Oliveira, Tae-Soo Jang, Petr Novak, Andrea Koblizkova, Veit Schubert, Andreas Houben, Jiri Macas

Summary: The transition from monocentric to holocentric organization in Cuscuta was associated with loss or truncation of kinetochore-related genes, disruption of centromeric localization of kinetochore proteins, and degeneration of the spindle assembly checkpoint. This indicates that holocentric Cuscuta species lack a standard kinetochore and do not employ the SAC to control microtubule attachment to chromosomes.
