Summary: In this study, multivariable logistic regression analysis was used to analyze the baseline data of anal HPV 16 infection, and it was found that current smoking, nadir CD4 count, prevalent cervical HPV 16 infection, and anogenital herpes in the previous 6 months were associated with prevalent anal HPV 16 infection.

Summary: This study compared the early natural history of different HPV types in the development of cervical cancer, using prospective data from three research projects. The results showed that the time to clearance of HPV infections was similar across different HPV types, indicating that relative prevalence can predict relative incidence. The study also found that the time to clearance of incident HPV infections was similar among different risk classes and did not differ based on age groups. These findings have important implications for the refinement of health decision models and cost-effectiveness evaluation of prevention technologies.

Summary: The incidence and mortality of squamous cell carcinoma of the anus have been increasing globally due to high-risk HPV infections. Immunotherapies, such as immune checkpoint inhibitors, adoptive cell therapy, and vaccines, are being investigated for their potential role in the treatment of anal cancer. Research is focused on integrating immunotherapy into the treatment sequencing of anal cancer at different stages. The aim of this review is to provide a summary of the current state and future directions of immunotherapy in anal squamous cell cancers.

Summary: Anal cancer rates, especially in women, have been increasing. Women with a history of genital dysplasia or cancer are at higher risk of anal cancer. However, there is a lack of education on this increased risk, resulting in delayed diagnosis and advanced disease. This review presents current evidence on screening, treatment, and surveillance of anal precancerous lesions in high-risk women.

Summary: HPV infection leads to changes in host gene methylation, contributing to neoplastic progression of HPV-associated cancers. This study identified genome-wide methylation changes in anal and cervical cancer, as well as high-grade pre-neoplastic lesions, and found common overlapping hypermethylated genes in the progression of both cancers.

Summary: This systematic review evaluates the accuracy of DNA HRHPV, mRNA HPV, DNA HPV16, and p16 biomarkers in identifying anal intraepithelial neoplasia (AIN) 2 or 3 and cancer. The findings suggest that DNA HR HPV can be a useful tool for screening aHSIL and anal cancer, while mRNA HPV shows better performance as an isolated test. Subgroup analysis reveals similar accuracy in MSM with HIV.

Summary: The incidence of anal intraepithelial neoplasias associated with HPV is increasing worldwide, with higher risk among individuals living with HIV/AIDS. A study in Mexico found that 59.8% of men and women living with HIV had anal HPV infection, with multiple genotypes identified. Risky behaviors, such as receptive anal intercourse and inconsistent condom use, were associated with HPV infection. Cytology identified low-grade and high-grade squamous intraepithelial lesions in a significant proportion of participants.

Summary: This study aimed to investigate the factors associated with progression of HPV infection to anal dysplasia and cancer, as well as the differences in anal microbiome composition in this rare disease. The results showed significant differences in microbial community composition between anal cancer patients and high-risk normal individuals, suggesting a potential role of microbiome in inflammation and neoplasia progression.

Summary: Human papillomavirus can lead to HSILs in the anogenital area, and the microbiome may play a role. HIV-MSM individuals have a high risk of anal cancer, but current screening methods for HSILs lack specificity. This study found that elevated levels of microbiome-encoded proteins were associated with HSILs, and measuring these biomarkers improved the current strategy of anal cancer screening.

Summary: This study compared anal cytology, hrHPV DNA testing, and two novel cytology/hrHPV cotesting algorithms in detecting anal precancer/cancer among high-risk populations. The results showed that screening algorithms incorporating cytology and hrHPV testing significantly increased sensitivity but decreased specificity, particularly performing well in MSM.

Summary: The study found that in Australia, widespread uptake of the quadrivalent HPV vaccine has significantly reduced the prevalence of quadrivalent vaccine-targeted HPV genotypes in women aged 18-35. Vaccination has a big impact on HPV infection, but unvaccinated women have higher prevalence of other HPV types, influenced by factors such as age and number of sexual partners.

Summary: Studies suggest that HPV-18 variants from the A lineage have higher oncogenic potential compared to B variants, with uneven distribution in cervical adenocarcinomas and squamous cell carcinomas. Immortalized PHKs with HPV-18 A1 and B1 sublineages showed qualitative differences in colony formation, invasion ability, and resistance to apoptosis. Further investigation is needed to understand the mechanisms underlying these differences and their relevance to cervical cancer subtypes.

Summary: Smoking is associated with higher prevalence, persistence, and incidence of anal HPV in men. However, no significant differences were found between former smokers and never smokers. Smoking does not impact the prevalence or persistence of anal HPV in men who have sex with men but is associated with higher incidence of high-risk HPV. Among men who have sex with women, current smokers have a higher prevalence and incidence of low-risk HPV.

Summary: Prophylactic HPV vaccines are recommended for the prevention of HPV-related infections, tumors, and genital warts. Two doses of the vaccine have been shown to be as effective as three doses, leading to reduced vaccination schedules in many countries. Single-dose HPV vaccination has also shown promise in reducing the incidence of pre-cancer and cervical cancer.

Summary: This study found that the rs3819817 variant in the HAL gene is associated with vitamin D levels and bone mineral density. This variant is associated with low levels of 25-hydroxyvitamin D, vitamin D deficiency, and decreased hip and femoral neck BMD values. The study also found interactions between skin pigmentation and adiposity with this variant. Additionally, higher vitamin D levels were observed in the southern region compared to the northern region in indigenous postmenopausal women in Mexico.

Summary: This study investigated the potential mediating role of BMI in the association between SSB intake and 25(OH)D. The findings suggest that high total sugar intake from SSB and BMI independently contribute to lower 25(OH)D, and BMI mediates the inverse association between total sugar intake from SSB and 25(OH)D. Furthermore, an increased risk of having vitamin D deficiency was found in the population who consumed higher levels of sugar from SSB or soda drinks.

Summary: CD40, a member of TNFR family, is expressed in different types of cells, including immune cells and cancer cells. The higher expression of CD40 is observed in squamous cell carcinomas compared to adenocarcinomas. Moreover, the expression of CD40 and CD40L in squamous cervical carcinomas is associated with better overall survival. These proteins could be explored as prognostic markers in cervical cancers.
