Summary: A novel missense mutation (I423T) in the FSHR second transmembrane domain was identified in a woman with primary ovarian failure, leading to impaired receptor expression and signaling. Meanwhile, the D408Y mutation severely affected membrane expression and signal transduction. Molecular dynamics simulations showed important disruptions in both mutant FSHRs, explaining the differences in expression levels, signaling, and phenotypic expression between I423T and D408Y mutant FSHRs.

Summary: It has been traditionally believed that pituitary hormones act on their cognate receptors in classical target glands to carry out their various functions. However, recent studies have revealed that multiple types of pituitary hormone receptors are widely expressed in non-classical target organs. This article summarizes the additional biological functions of pituitary hormones (thyroid-stimulating hormone, follicle-stimulating hormone, luteinizing hormone, adrenocorticotrophic hormone, and prolactin) when they act on non-classical organs, defining them as part of an atypical pituitary hormone-target tissue axis. This novel concept provides insight into the mechanisms behind metabolic diseases and offers potential novel therapeutic targets.

Summary: This study discovered that deletion of the pituitary enhancer located in the 6th intron of the Nr5a1 gene in mice leads to the disappearance of NR5A1 in the pituitary gland and severely affects the development of reproductive organs. Transcriptomic analysis further identified genes that are specifically dependent on NR5A1 expression in pituitary gonadotropes, which play important roles in reproductive organ development through regulation of pituitary gonadotrope functions.

Summary: Recent studies have shown that the hypo-glycosylated FSH glycoform FSH21/18 is more bioactive than the fully-glycosylated FSH24. FSH21/18 predominates in women of reproductive prime while FSH24 is more common in peri-post-menopausal women. Differential FSH glycosylation modulates FSHR oligomerization, with high concentrations of FSH21/18 and FSH24 leading to dissociation of FSHR oligomers into monomers, and low concentrations promoting FSHR association into oligomers.

Summary: This study investigated the pharmacokinetics and pharmacodynamics properties, safety, and tolerability of cetagliptin. The results showed that cetagliptin has great potential for the treatment of type 2 diabetes patients due to its inhibition of DPP-4, increase in GLP-1 and insulin, and decrease in glucose. It may be more effective in DPP-4 inhibition than sitagliptin.
