Summary: Measurement of antibody content and function is crucial for diagnosing and selecting the most effective convalescent plasma (CP) units. Zhang et al. analyzed over 19,000 COVID-19 CP (CCP) samples and found a moderate correlation between antibody amount and neutralizing titer. About one-third of the samples had little or no neutralizing activity. The study highlights differences in the quality of CP units collected before antibody screening and suggests using CP units with top quintile of antibody content and neutralizing capacity for future infectious disease emergencies.

Summary: This study evaluated the antibody levels in COVID-19 convalescent plasma and found different neutralization activities against variants of SARS-CoV-2. 60% of infected individuals with the beta variant had IgG antibody levels above 100μg/ml. Anti-S-IgG levels were inversely correlated with neutralization activity for the wild-type and beta variants, while no correlation was observed with anti-N-IgG levels. IgG antibodies could prevent infection and cytopathic effects of six different variants of concern, except for the beta and omicron strains.

Summary: Early administration of high-titer CIP to non-critically ill patients with COVID-19 respiratory disease resulted in significantly increased anti-SARS-CoV-2 specific antibodies compared to baseline, and a nonsignificant reduction in ICU transfer and death compared to controls. This prospective phase II trial suggests that CIP may delay progression to critical illness and death in specific patient populations during the COVID-19 pandemic. The study provides insight into the optimal timing and potential population for the use of CIP in COVID-19, particularly in settings without access to other interventions or in planning for future coronavirus pandemics.

Summary: The study aimed to investigate the results and side-effects of using ruxolitinib as a treatment for polycythemia vera (PV). The study included 34 PV patients from 18 different centers. The response to ruxolitinib treatment was evaluated based on reduction in spleen volume (>= 35% splenomegaly size) by imaging and control of hematocrit levels (<= 45%) compared to baseline. The results showed that 55.9% of patients achieved reduction in spleen volume and hematocrit control at 3 months, and this increased to 61.8% at 6 months.

Combining 4D-STE and fQRS complexes enhances the identification of early left ventricular systolic dysfunction. These findings offer potential improvements in recognizing and managing cardiovascular complications in PV patients, with implications for future research and clinical practice. Further investigations are needed to elucidate underlying mechanisms and validate these markers in larger cohorts.
