The interpretation of in vitro cytotoxicity data of Cu(II)-1,10-phenanthroline (phen) complexes normally does not take into account the speciation that complexes undergo in cell incubation media and its implications in cellular uptake and mechanisms of action. We synthesize and test the activity of several distinct Cu(II)-phen compounds; up to 24 h of incubation, the cytotoxic activity differs for the Cu complexes and the corresponding free ligands, but for longer incubation times (e.g., 72 h), all compounds display similar activity. Combining the use of several spectroscopic, spectrometric, and electrochemical techniques, the speciation of Cu-phen compounds in cell incubation media is evaluated, indicating that the originally added complex almost totally decomposed and that Cu(II) and phen are mainly bound to bovine serum albumin. Several methods are used to disclose relationships between structure, activity, speciation in incubation media, cellular uptake, distribution of Cu in cells, and cytotoxicity. Contrary to what is reported in most studies, we conclude that interaction with cell components and cell death involves the separate action of Cu ions and phen molecules, not [Cu(phen)(n)] species. This conclusion should similarly apply to many other Cu-ligand systems reported to date.

Experimental and computational studies on pseudotetrahedral nickel(ii)-(S or R)-dihalogen-salicylaldiminates with Δ- or ?-chirality induction at-metal

Arezoo Saadati, Hadi Amiri Rudbari, Mahnaz Aryaeifar, Olivier Blacque, Isabel Correia, Mohammad Khairul Islam, Dennis Woschko, Takin Haj Hassani Sohi, Christoph Janiak, Mohammed Enamullah

Summary: In this study, the synthesis of three pairs of pseudotetrahedral nickel complexes with chirality induction at-metal was investigated. The crystal structures and electronic circular dichroism spectra confirmed the preservation of chirality induction and enantiopurity in these complexes. Supramolecular packing analysis and non-covalent interaction studies were also performed, demonstrating the presence of intermolecular interactions and the phase purity of the crystal samples.

Summary: In this study, a series of mononuclear non-oxido vanadium(IV) complexes are synthesized and characterized. The complexes show distorted octahedral or trigonal prismatic arrangement around the metal center. The EPR and DFT data suggest the coexistence of different isomers in solution, and the ESI-MS results indicate partial oxidation of the complexes. The complexes exhibit moderate binding affinity towards bovine serum albumin and show cytotoxicity against cancer cells by inducing cell death through apoptosis.

Summary: In this study, a combinatory antitumoral nanotherapy was developed using magneto-fluorescent nanoparticles as carriers for a combination of two drugs (doxorubicin and ofloxacin) and surface modification with two targeting agents (AZM-SH and Cetuximab). The results showed that the nanoparticles modified with AZM-SH had higher cellular uptake than those modified with Cetuximab, making them more effective for colorectal cancer cells. Furthermore, the oral administration of the nanoparticles with an enteric coating was successfully achieved under extreme conditions.

Enhancing water sensing via aggregation-induced emission (AIE) and solvatofluorochromic studies using two new dansyl derivatives containing a disulfide bound: Pollutant metal ions detection and preparation of water-soluble fluorescent polymeric particles

Frederico Duarte, Georgi Dobrikov, Atanas Kurutos, Hugo M. Santos, Javier Fernandez-Lodeiro, Jose Luis Capelo-Martinez, Elisabete Oliveira, Carlos Lodeiro

Summary: Polarity-sensitive dansyl derivatives L1 and L2 were synthesized and their ability to sense pollutant metal ions was investigated. Both L1 and L2 exhibited high sensitivity towards Cu2+ and Hg2+ metal ions, with L2 capable of detecting and quantifying Hg2+ concentrations as low as 2.5 μM. Additionally, both compounds showed positive sol-vatofluorochromic behavior, modulated in the presence of water, leading to fluorescence enhancement through aggregation-induced emission (AIE). Luminescent L1-based polystyrene-block-polybutadiene-block-polystyrene (SBS) microparticles were successfully prepared, demonstrating the potential applications of dansyl-derivatives in biomedicine and environmental fields.

Silvia Nuti, Adriaïn Fernandez-Lodeiro, Lidia E. Chinchilla, Ana B. Hungria, Jose-Luis Capelo-Martinez, Carlos Lodeiro, Javier Fernandez-Lodeiro

Summary: This study presents a simple method for fabricating Au@Pd bimetallic branched nanoparticles, which allow for tunable optical response, using polyallylamine-stabilized branched AuNPs as template cores for Pd overgrowth. The concentration of PdCl4(2-) and ascorbic acid (AA) can be manipulated to alter the palladium content, enabling the growth of a Pd shell up to approximately 2 nm thick. The presence of palladium on the surface of gold nanoparticles enhances their catalytic properties, as demonstrated through a comparison of nanoenzymatic activity.
