Molecular and culture-based surveys of metabolically active hydrocarbon-degrading archaeal communities in Sundarban mangrove sediments

Archaea remain important players in global biogeochemical cycles worldwide, including in the highly productive mangrove estuarine ecosystems. In the present study, we have explored the diversity, distribution, and function of the metabolically active fraction of the resident archaeal community of the Sundarban mangrove ecosystem, using both culture-independent and culture-dependent approaches. To evaluate the diversity and distribution pattern of the active archaeal communities, RNA based analysis of the 16S rRNA gene was performed on an Illumina platform. The active Crenarchaeal community was observed to remain constant while active Euryarchaeal community underwent considerable change across the sampling sites depending on varying anthropogenic factors. Haloarchaea were the predominant group in hydrocarbon polluted sediments, leading us to successfully isolate eleven p-hydroxybenzoic acid degrading haloarchaeal species. The isolates could also survive in benzoic acid, naphthalene, and o-phthalate. Quantitative estimation of p-hydroxybenzoic acid degradation was studied on select isolates, and their ability to reduce COD of polluted saline waters of Sundarban was also evaluated. To our knowledge, this is the first ever study combining culture-independent (Next Generation sequencing and metatranscriptome) and culture-dependent analyses for an assessment of archaeal function in the sediment of Sundarban.

A trinuclear copper (II) complex of naproxen-appended salicylhydrazide: Synthesis, crystal structure, DNA binding and molecular docking study

Archaeal Hsp14 drives substrate shuttling between small heat shock proteins and thermosome: insights into a novel substrate transfer pathway

Summary: The study investigated the crosstalk in the heat shock response pathway of thermoacidophilic crenarchaeon Sulfolobus acidocaldarius, focusing on the small heat shock protein Hsp14. It was found that Hsp14 interacts with Hsp20 and Hsp60 to facilitate substrate proteins' folding under stress conditions, demonstrating the dynamic oligomeric structure of Hsp14 and its ability to form hetero-oligomers with Hsp20 at higher temperatures. Moreover, enthalpy-driven direct physical interaction between Hsp14 and Hsp60 was revealed, showing that Hsp14 can transfer substrate proteins captured by sHsp to Hsp60 for refolding.

These genomes had a high GC content and encoded genes related to survival in the presence of hydrocarbons. Functional annotation revealed different pathways utilized by the strains for degrading aromatic compounds.

Anthropogenic impact accelerates antibiotic resistome diversity in the mangrove sediment of Indian Sundarban

Summary: Mangroves in India are vulnerable to pollution and deforestation. This study investigates the microbiome of the Sundarban mangrove and its antibiotic resistance genes. Physicochemical parameters play a crucial role in maintaining the mangrove microbiome. The Sundarban mangrove has a higher diversity of antibiotic resistance genes compared to Chinese mangroves, but lower abundance. Anthropogenic activities and pollution can degrade microbiome diversity, disrupt function, and enrich resistance genes.

Heat shock response in Sulfolobus acidocaldarius and first implications for cross-stress adaptation

Summary: This study used high-throughput transcriptomic analysis to investigate the gene expression changes of S. acidocaldarius under high temperatures and stress conditions. The results revealed key genes involved in stress adaptation and consistently overexpressed transcription factors across all stress conditions. This research provides the first insight into the molecular mechanisms of cross-stress adaptation in S. acidocaldarius.
