Summary: The causes of grey matter pathology and diffuse neuron injury in multiple sclerosis (MS) are not fully understood. This study found that axonal stress signals from white matter lesions may play a role in initiating diffuse grey matter pathology. The researchers analyzed the transcriptome of actively translating neuronal transcripts in mouse models of demyelinating disease and identified upregulated genes associated with the ISGylation pathway. They also noted changes in neuronal extracellular vesicle protein and miRNA cargo, with a skewed distribution towards proinflammatory and neurotoxic miRNAs.

This review focuses on the role of exosomal miRNAs in the onset and progression of IPF and discusses their potential as therapeutic agents.

Summary: The study found that long RNA transcripts enriched in extracellular vesicles (EV) have longer average half-lives and distinctive stability-related sequence and structure characteristics. These EV-enriched long RNA transcripts are enriched in HNRNPA2B1 binding motifs and are impacted by this RNA-binding protein. After modification of the cellular transcriptome due to signaling, the rate of EV enrichment relative to cells is altered for many transcripts, showing a negative correlation with intracellular abundance.
