Enhancing transcription in Escherichia coli and Pseudomonas putida using bacteriophage lambda anti-terminator protein Q

Summary: Functional characterization of metagenomic DNA often involves expressing heterologous DNA in genetically tractable microorganisms such as Escherichia coli. Anti-terminator protein lambda Q has been shown to efficiently transcribe heterologous genes with intrinsic terminators, and can enhance expression of heterologous DNA in metabolic engineering applications. This study demonstrates the utility of lambda Q in improving transcription efficiency and expression levels in both E. coli and Pseudomonas putida.

MgO(111) Nanocatalyst for Biomass Conversion: A Study of Carbon Coating Effects on Catalyst Faceting and Performance

Summary: This study reports the synthesis, characterization, and catalytic properties of commercial MgO, MgO(111), and carbon coated derivatives for 2-pentanone condensation. The results show that MgO(111) maintained impressive selectivity towards the dimer product after carbon coating, while the other catalysts experienced a decrease in conversion and selectivity.

Summary: This study examines the role of CuMAlOx catalysts in upgrading woody biomass via SCM-DHDO. The results show that CuCeAlOx exhibits the highest activity and is correlated with its high reducibility and acidity. Additionally, Cu sintering and biogenic impurity poisoning are identified as possible deactivation mechanisms. Overall, SCM-DHDO processing can produce suitable light oxygenates for gasoline bioblendstock applications.

Summary: This study reports the synthesis of a Cu(I)-loaded COF material with subnanometer pores and a three-dimensional network morphology, and investigates the sorption mechanisms of hydrogen, ethylene, and carbon monoxide with this material. The results show that all three gases desorb near or above room temperature, and can be desorbed on-demand by irradiation with ultraviolet light. This work represents a step towards developing more practical ambient temperature gas storage and efficient regeneration of sorbents.

Simple and Rapid Site-Specific Integration of Multiple Heterologous DNAs into the Escherichia coli Chromosome

Summary: This study developed a system for integrating up to 12 genetic constructs into the E. coli chromosome. Site-specific recombinases were used to catalyze recombination between specific DNA sequences, enabling the integration of heterologous DNA. The utility of this system was demonstrated by the insertion of eight DNA methyltransferases into the E. coli chromosome.

Tuning of Gene Expression in Clostridium phytofermentans Using Synthetic Promoters and CRISPRi

Summary: This article presents a set of methods for controlling gene expression in Clostridia. The authors developed a simple benchtop electroporation method for introducing replicating plasmids and resistance markers into Clostridia. They defined a series of promoters with a wide range of expression levels and demonstrated the ability to quantitatively alter gene expression using a Tet repressor and anhydrotetracycline. They integrated these methods into a regulatory system and successfully repressed the expression of reporter and fermentation genes in Clostridia using CRISPRi.
