Claudins are major integral proteins of tight junctions (TJs), the apical cell-cell adhesions that enable maintaining polarity of epithelial cells, their differentiation, and cell signaling. A number of studies have indicated that claudins might play a crucial role in both physiology and pathogenesis. Their tissue-specific expression was originally linked to the development of different types of cancer and triggered a hope to use them as diagnostic or prognostic markers. However, it seems that their expression is more complex than that, and undoubtedly, claudins participate in one of the most important molecular events in cells. This review summarizes the recent research evaluating the role of claudins in fertility and the most common endocrine-dependent cancers in the reproductive system and highlights the crucial role of claudins both in human fertility and the most common cancers.

Deoxynivalenol induces apoptosis and autophagy in human prostate epithelial cells via PI3K/Akt signaling pathway

Karolina Kowalska, Marta Justyna Koziel, Dominika Ewa Habrowska-Gorczynska, Kinga Anna Urbanek, Kamila Dominska, Agnieszka Wanda Piastowska-Ciesielska

Summary: The PI3K/Akt signaling pathway is one of the most deregulated pathways in prostate cancer, and DON can affect apoptosis and autophagy in normal prostate epithelial cells by modulating this pathway. High concentrations of DON induce apoptosis, while low concentrations induce autophagy.

The Influence of Angiotensin Peptides on Survival and Motility of Human High-Grade Serous Ovarian Cancer Cells in Serum Starvation Conditions

Kamila Dominska, Kinga Anna Urbanek, Karolina Kowalska, Dominika Ewa Habrowska-Gorczynska, Marta Justyna Koziel, Tomasz Ochedalski, Agnieszka Wanda Piastowska-Ciesielska

Summary: This study investigated the effects of angiotensin peptides on HGSOC cell lines and found that these peptides can impact cell proliferation, motility, and cell cycle distribution. However, they do not significantly affect the expression of genes associated with epithelial-mesenchymal transition. Furthermore, the effects of angiotensin peptides on cancer cells depend on the availability of growth factors and nutrients.

SGLT2 Inhibitors May Restore Endothelial Barrier Interrupted by 25-Hydroxycholesterol

Agnieszka Pawlos, Marlena Broncel, Ewelina Wozniak, Lukasz Markiewicz, Agnieszka Piastowska-Ciesielska, Paulina Gorzelak-Pabis

Summary: This study aimed to evaluate the impact of SGLT2 inhibitors on the integrity of endothelial cells disrupted by 25-OHC and assess the association with changes in VE-cadherin levels. The results showed that SGLT2 inhibitors, when used in conjunction with 25-OHC, significantly increased endothelial integrity and improved the integrity of endothelial cells disrupted by 25-OHC. This effect was associated with significant improvements in VE-cadherin levels.

Revealing the Role of Alternariol in the Local Steroidogenesis in Human Prostate Normal and Cancer Cells

Kinga Anna Urbanek, Karolina Kowalska, Dominika Ewa Habrowska-Gorczynska, Marta Justyna Koziel, Kamila Dominska, Agnieszka Wanda Piastowska-Ciesielska

Summary: This study evaluates the impact of AOH on the local steroidogenesis process in normal and prostate cancer cells. The findings suggest that while AOH has a significant impact on cell cycle, inflammation, and apoptosis in prostate cancer cells, its influence on steroidogenesis is not significant. However, when combined with another steroidogenic agent, AOH significantly affects steroidogenesis.

Estrogens, Estrogen Receptors and Tumor Microenvironment in Ovarian Cancer

Marta Justyna Koziel, Agnieszka Wanda Piastowska-Ciesielska

Summary: Ovarian cancer, a common cancer in women, is believed to be influenced by hormonal imbalance. Estrogens, a group of hormones with important roles in physiological and pathological processes, are involved in regulating proliferation, invasiveness, and the epithelial to mesenchymal transition in ovarian cancer. Estrogen signaling also plays a role in the biology of the tumor microenvironment. This review provides information on estrogen receptors, estrogens, and their association with the tumor microenvironment, as well as changes in estrogen receptor expression following exposure to various environmental chemicals.

Estrogen receptor α mediates alternariol-induced apoptosis and modulation of the invasiveness of ovarian cancer cells

Marta Justyna Koziel, Dominika Ewa Habrowska-Gorczynska, Kinga Anna Urbanek, Kamila Dominska, Agnieszka Wanda Piastowska-Ciesielska, Karolina Kowalska

Summary: This study found that alternariol may affect ovarian cancer cells by modulating estrogen receptor alpha, leading to apoptosis, oxidative stress, and decreased invasive ability.

Cell Cycle Status Influences Resistance to Apoptosis Induced by Oxidative Stress in Human Breast Cancer Cells, Which Is Accompanied by Modulation of Autophagy

Magdalena Kluska, Agnieszka Wanda Piastowska-Ciesielska, Paulina Tokarz

Summary: Cancer cells can temporarily arrest their cell cycle at the G0 or G1 phase, which may contribute to tumor heterogeneity and drug resistance. The influence of G0- and G1-arrest on chemosensitivity has not been fully understood.

Summary: By utilizing nanomedicine and nuclear medicine, we have developed a radioisotope nanocarrier for the treatment of prostate cancer. This nanocarrier can specifically target prostate cancer cells and exhibits excellent performance in vitro with high stability. Although further studies are required to validate its efficacy in vivo, our research represents a significant advancement in the field of radionanomedicine.
