Summary: LCs in the epidermis contain genes that contribute to antimicrobial responses, including chemokines such as CCL1, CCL17, CCL19, CCL2, CCL22, CXCL14, and CXCL2, which mediate both antimicrobial and inflammatory responses. These genes encode proteins with broad antibacterial activity, including against Staphylococcus aureus, the leading cause of skin infections.

Summary: Photobiomodulation (PBM) is a valuable tool in dermatology for treating inflammatory skin conditions using low levels of light, but the physiological pathways responsible for its anti-inflammatory effect have not been well defined. This study found that PBM activated the Nrf2 pathway to control inflammation and played a key role in reducing inflammatory response. The findings support the role of PBM in skin immunomodulation and suggest it as a promising alternative or complementary therapeutic approach for skin-related inflammatory diseases.

Langerhans cells in the skin and oral mucosa: Brothers in arms?

Summary: The skin and oral mucosa are constantly exposed to pathogens and foreign antigens, and both contain Langerhans cells (LC) that can promote both tolerogenic and inflammatory immune responses. While skin LC have been extensively studied, less is known about oral mucosal LC. This review summarizes the current knowledge on LC subsets in the skin compared to the oral mucosa, including their development, homeostasis, function, and interaction with the microbiota. It also updates recent advances on the role of LC in inflammatory skin and oral mucosal diseases.

Summary: Research has shown that the epigenetic regulator miR-23a is crucial for LC antigen uptake and bone marrow-derived LC repopulation during inflammatory conditions. miR-23a regulates LC phagocytosis and repopulation by targeting molecules involved in efferocytosis, endocytosis, and proapoptotic pathways. This study reveals the context-dependent regulatory role of miR-23a in LCs, providing insights into the TGF-b- and PU.1-mediated regulation during steady-state and inflammation-induced repopulation.

Summary: This study reveals that tissue expansion mechanically stimulates the activation of SFRP2 protein in the skin, which is secreted by Langerhans cells. SFRP2 may act as a newly discovered effector of mechanotransduction pathways. Additionally, stretching activates Wnt signaling and cell proliferation, and tissue-expanded Langerhans cells proliferate and rebuild their network in the growing skin.

Summary: This study found that GAS infection is associated with multiple clinical sequelae, including psoriasis, and healthy individuals and those with psoriasis have high frequencies of GAS-responsive T cells. Psoriasis patients have higher frequencies of circulating GAS-reactive T cells, which are enriched for activation markers, cytolytic potential, and tissue association. In addition, GAS infection promotes the proliferation and activation of CD1a-autoreactive T cells, linking GAS infection to the pathogenesis and treatment of psoriasis.

Summary: This study demonstrates that allogeneic hematopoietic stem cell transplantation (HSCT) can potentially cure patients with chronic lymphocytic leukemia (CLL), with favorable progression-free survival (PFS) and overall survival (OS) rates. The impact of novel agents on HSCT outcomes was found to be negligible. Reduced intensity conditioning (RIC) HSCT may be a successful treatment option for CLL patients with Richter's transformation and poor-risk CLL patients.

Langerhans dendritic cell vaccine bearing mRNA-encoded tumor antigens induces antimyeloma immunity after autotransplant

Summary: It has been found that posttransplant vaccination can improve immune responses and prolong disease-free survival in multiple myeloma patients after autologous stem cell transplantation (ASCT). A phase 1 vaccine trial was conducted to investigate the safety and effectiveness of autologous Langerhans-type dendritic cell (LC) vaccination after ASCT. The results showed that the vaccination was safe and induced antigen-specific immune reactivity.
