Labelling Bacterial Nanocages with Photo-switchable Fluorophores

The robustness and biocompatibility of bacterial nanocages holds promise for bio-nanotechnologies. The propensity of these nano-carriers to penetrate cells has been demonstrated, which calls for the development of tracking strategies, both in vitro and in vivo. Here, we label bacterial nanocages with photo-switchable fluorophores, to facilitate their imaging by super-resolution microscopy. We demonstrate the functionalization of the encapsulin from Brevibacterium linens with a spiropyran, which is not fluorescent, by covalent attachment to the amine residues at the outer encapsulin shell. Upon alternating irradiation with ultraviolet and visible light, the spiropyran switches forth and back to its fluorescent merocyanine photo-isomer and thus the fluorescence can be switched on and off, reversibly. We also show that the bacterial compartments preserve their structural integrity upon covalent modification and over at least five irradiation cycles.

Acceleration of lipid reproduction by emergence of microscopic motion

Summary: Self-reproducing lipids can initiate, sustain and accelerate the movement of octanol droplets in water. Reciprocally, the chemotactic movement of the octanol droplets increases the rate of lipid reproduction substantially. This coupling between molecular chemistry and microscopic motility offers alternative means of performing work and catalysis in micro-heterogeneous environments.

Optimization of Monochloroacetic Acid Biodegradation by Recombinant E. coli BL21 (DE3)/pET-bcfd1 Carrying Haloacid Dehalogenase Gene from Bacillus cereus IndB1

Summary: In this study, the optimal conditions for producing active haloacid dehalogenase by E. coli BL21 (DE3) carrying a recombinant plasmid were investigated, leading to the identification of specific conditions that resulted in successful degradation of monochloroacetic acid (MCA) and production of chloride ions. The expression of the protein was influenced by inducer concentration, pre-induction incubation time and temperature, as well as induction time and temperature, while MCA concentration and medium pH affected the ability of the recombinant strain to grow in a toxic environment. These findings provide a foundation for further exploration of dehalogenases for MCA biodegradation.
