Usefulness of melatonin as complementary to chemotherapeutic agents at different stages of the angiogenic process

Chemotherapeutics are sometimes administered with drugs, like antiangiogenic compounds, to increase their effectiveness. Melatonin exerts antitumoral actions through antiangiogenic actions. We studied if melatonin regulates the response of HUVECs to chemotherapeutics (docetaxel and vinorelbine). The inhibition that these agents exert on some of the processes involved in angiogenesis, such as, cell proliferation, migratory capacity or vessel formation, was enhanced by melatonin. Regarding to estrogen biosynthesis, melatonin impeded the negative effect of vinorelbine, by decreasing the activity and expression of aromatase and sulfatase. Docetaxel and vinorelbine increased the expression of VEGF-A, VEGF-B, VEGF-C, VEGFR-1, VEGFR-3, ANG1 and/or ANG-2 and melatonin inhibited these actions. Besides, melatonin prevented the positive actions that docetaxel exerts on the expression of other factors related to angiogenesis like JAG1, ANPEP, IGF-1, CXCL6, AKT1, ERK1, ERK2, MMP14 and NOS3 and neutralized the stimulating actions of vinorelbine on the expression of FIGF, FGFR3, CXCL6, CCL2, ERK1, ERK2, AKT1, NOS3 and MMP14. In CAM assay melatonin inhibited new vascularization in combination with chemotherapeutics. Melatonin further enhanced the chemotherapeutics-induced inhibition of p-AKT and p-ERK and neutralized the chemotherapeutics-caused stimulatory effect on HUVECs permeability by modifying the distribution of VE cadherin. Our results confirm that melatonin blocks proangiogenic and potentiates antiangiogenic effects induced by docetaxel and vinorelbine enhancing their antitumor effectiveness.

Summary: Melatonin acts as a promising sensitizer molecule for cancer cells, enhancing the efficacy of chemotherapy and radiotherapy by inhibiting angiogenesis. Its antiangiogenic effects help to overcome the pro-angiogenic factors induced by chemotherapy, making tumors more sensitive to treatment. Melatonin also regulates factors involved in angiogenesis and acts as a radiosensitizer, increasing the effects of radiation on tumor cells.

Novel potential oncogenic and druggable mutations of FGFRs recur in the kinase domain across cancer types

Summary: Fibroblast growth factor receptors (FGFRs) are frequently altered by single nucleotide variants (SNVs) in many human cancers, with mutations occurring at different frequencies depending on the cancer type. Mutations in the kinase domain of FGFRs, especially those occurring in tumors such as urothelial carcinoma, are potential targets for cancer therapy. The study used bioinformatics analysis to identify novel mutations in the kinase domain of FGFRs, highlighting the importance of understanding the functional impact of alterations clustering on similar residues in analogous proteins.

Melatonin Modulation of Radiation-Induced Molecular Changes in MCF-7 Human Breast Cancer Cells

Summary: Radiation therapy is a crucial part of cancer treatment, but it has adverse effects. Melatonin, as an adjuvant molecule, can limit damage to normal tissues and enhance the efficacy of radiation therapy in breast cancer. This study revealed that melatonin can potentiate the anti-proliferative effect of radiation, modulate gene and microRNA expression, and enhance the inhibition of cancer growth. Melatonin shows promise as both a radiosensitizer and radioprotector in breast cancer treatment.

The impact of adipokines on vascular networks in adipose tissue

Summary: Adipose tissue is an active endocrine organ, which secretes adipokines that have various functions in the body. The plasticity of adipose tissue is associated with angiogenesis, which supports tissue expansion. Abnormal production or loss of certain adipokines can lead to metabolic dysfunction related to obesity. Understanding the role of adipokines in angiogenesis may offer new therapeutic approaches for treating obesity and metabolic diseases.

Summary: In pediatric rhabdomyosarcoma, elevated Akt signaling promotes tumor malignancy by hyperactivating the mTOR/p70S6K pathway and enhancing cell invasion and migration. Inhibiting PI3K and Akt signaling can reverse the aggressive phenotype of tumor cells, while mTOR inhibition fails to block cell dissemination. Additionally, pronounced Akt1 signaling increases the susceptibility of tumor cells to certain treatments, such as 2-DG and lovastatin.

Summary: In vertebrates, two homologous heterotetrameric AP1 complexes play vital roles in regulating intracellular protein sorting in a vesicle-dependent manner. These complexes, composed of four subunits (gamma, beta 1, mu 1, and sigma 1), are ubiquitously expressed and essential for development. The knockout of specific subunits in animal models demonstrated their importance in multicellular organism development, neuronal and epithelial cell specification. Mutations in genes encoding for these subunits have been associated with various human diseases, including adaptinopathies, which affect intracellular vesicular traffic. A zebrafish model with an ap1g1 knockout was used to study the molecular mechanisms underlying adaptinopathies, revealing dysregulated cell adhesion and providing potential targets for therapeutic interventions.
