Endophytic Actinobacteria from the Brazilian Medicinal Plant Lychnophora ericoides Mart. and the Biological Potential of Their Secondary Metabolites

Endophytic actinobacteria from the Brazilian medicinal plant Lychnophora ericoides were isolated for the first time, and the biological potential of their secondary metabolites was evaluated. A phylogenic analysis of isolated actinobacteria was accomplished with 16S rRNA gene sequencing, and the predominance of the genus Streptomyces was observed. All strains were cultured on solid rice medium, and ethanol extracts were evaluated with antimicrobial and cytotoxic assays against cancer cell lines. As a result, 92% of the extracts showed a high or moderate activity against at least one pathogenic microbial strain or cancer cell line. Based on the biological and chemical analyses of crude extracts, three endophytic strains were selected for further investigation of their chemical profiles. Sixteen compounds were isolated, and 3-hydroxy-4-methoxybenzamide (9) and 2,3-dihydro-2,2-dimethyl-4(1H)-quinazolinone (15) are reported as natural products for the first time in this study. The biological activity of the pure compounds was also assessed. Compound 15 displayed potent cytotoxic activity against all four tested cancer cell lines. Nocardamine (2) was only moderately active against two cancer cell lines but showed strong activity against Trypanosoma cruzi. Our results show that endophytic actinobacteria from L.ericoides are a promising source of bioactive compounds.

Summary: This study used the structure of SARS-CoV-2 spike protein to examine the impact of mutations on the protein's stability and interaction with ACE-2 receptor. Molecular dynamics simulations and protein-protein docking experiments revealed that the mutations affected the stability of the protein and improved its interaction with ACE-2 receptor, particularly in the Gamma variant.

Neuroprotective Properties of Chlorogenic Acid and 4,5-Caffeoylquinic Acid from Brazilian arnica ( Lychnophora ericoides ) after Acute Retinal Ischemia

Summary: Lychnophora, also known as Brazilian arnica, is a South American flowering plant used in traditional medicine. Its active components, derived from chlorogenic acid and C-flavonoids, exhibit anti-inflammatory and analgesic properties. In this study, it was found that chlorogenic acid and 4,5-di-O-[E]-caffeoylquinic acid from Lychnophora plants have neuroprotective effects in a rodent glaucoma model. The pretreatment with these compounds protected the retinal layers against damage and reduced neuronal death after ischemic insult.

Repurposing of 5-nitrofuran-3,5-disubstituted isoxazoles: A thriving scaffold to antitrypanosomal agents

Summary: Chagas disease (CD), caused by Trypanosoma cruzi, requires new antitrypanosomal agents due to the adverse effects and toxicity of current drugs. 5-Nitrofuran-isoxazole analogs synthesized in this study showed significant in vitro antitrypanosomal activity against T. cruzi. Analog 7s exhibited the highest efficacy, with an IC50 value of 40 nM and selectivity index of 132.50. The active analogs showed substrate activity for type I nitroreductase, suggesting their potential as agents for CD treatment.

Summary: Cannabis legalization has led to increased cannabis use during pregnancy. This study found that prenatal exposure to synthetic cannabinoid WIN can affect the respiratory control system of neonatal and juvenile rats, with greater CO2 sensitivity in males and breathing and brainstem changes. In females, it leads to increased apnea and reduced serotonin neurons. These findings demonstrate long-term and sex-specific consequences of excessive stimulation of the endocannabinoid system during gestation.

Oxidation Products from the Neolignan Licarin A by Biomimetic Reactions and Assessment of in vivo Acute Toxicity

Summary: Licarin A, a dihydrobenzofuranic neolignan found in medicinal plants and nutmeg seeds, has strong activity against protozoans responsible for Chagas disease and leishmaniasis. Biomimetic reactions using metalloporphyrin and Jacobsen catalysts resulted in the production of seven products, including four isomeric epoxidation products of licarin A. In vivo acute toxicity assays showed potential liver toxicity, but no tissue damage indicative of toxicity was observed after 14 days of exposure. New metabolic pathways of licarin A were identified through in vitro biomimetic oxidation reactions and in vitro metabolism by rat or human liver microsomes.

Antimicrobial Activity of an Fmoc-Plantaricin 149 Derivative Peptide against Multidrug-Resistant Bacteria

Summary: Antimicrobial resistance is a major public health concern and there is a need for novel antimicrobial therapies. This study designed a new antimicrobial peptide Pln149-PEP20 and tested its activity against 60 different species of bacteria. The results showed that Pln149-PEP20 had excellent bactericidal activity and its target was likely genes associated with membrane metabolism.

Summary: In this study, a hydrogel formulation containing a proanthocyanidin polymer-rich extract of Commiphora leptophloeos was developed and characterized. The hydrogel demonstrated good physico-chemical and microbiological stability and showed higher antifungal potential compared to a free extract. The results suggest that this hydrogel has the potential to be used in treating vulvovaginal candidiasis.

Ravidomycin Analogs from Streptomyces sp. Exhibit Altered Antimicrobial and Cytotoxic Selectivity

Summary: Six new ravidomycin analogs and one fucomycin analog were isolated from Streptomyces sp. Am59. The structures were determined using spectroscopic and computational methods. The absolute configuration of deacetylravidomycin was found to have a positive specific rotation, contrary to previous reports. The compounds exhibited antibacterial, antifungal, and cytotoxicity activities, with some showing greater selectivity than deacetylravidomycin.
