The short synthesis of alpha-thujone relies on the functionalization of the readily available dimethylfulvene. Furthermore, the three main metabolites of the natural product were also synthesized. Since d(6)-acetone can be used as a starting material, the route developed allows for the facile incorporation of isotopic labels which are required for detecting and quantifying trace amounts via GC/MS analysis.

Concentration-Dependent Self-Assembly of an Unusually Large Hexameric Hydrogen-Bonded Molecular Cage

Severin Merget, Lorenzo Catti, Shani Zev, Dan T. Major, Nils Trapp, Konrad Tiefenbacher

Summary: A large hexameric cage based on intermolecular amide-amide dimerization is reported, with a diameter of 2.3 nm and a cavity volume of 2800 Å(3), larger than any previously known capsule/cage structure relying solely on hydrogen bonds. The self-assembly process in chlorinated organic solvents is strongly concentration-dependent, with the monomeric form prevailing at low concentrations. Additionally, the formation of host-guest complexes with fullerenes (C-60 and C-70) was observed.

Modelling folates reaction kinetics during cowpea seed germination in comparison with soaking

Fanny Coffigniez, Michael Rychlik, Christian Mestres, Lisa Striegel, Philippe Bohuon, Aurelien Briffaz

Summary: The study found that in the germination process of cowpea seeds, the enzymatic interconversion of folate forms into 5-methyltetrahydrofolate was intense. After 48 hours, there was a high enzymatic production of 5-methyltetrahydrofolate, 5-formyltetrahydrofolate, and tetrahydrofolate, suggesting that a long germination process may be more beneficial than soaking in increasing the production of bioavailable folates within the seed for human consumption.

Nutritional metabolites in Brassica rapa subsp. chinensis var. parachinensis (choy sum) at three different growth stages: Microgreen, seedling and adult plant

Li Zou, Wee Kee Tan, Yuanyuan Du, Hui Wen Lee, Xu Liang, Jiajia Lei, Lisa Striegel, Nadine Weber, Michael Rychlik, Choon Nam Ong

Summary: Significant metabolic profile shifts were observed in different developmental stages of choy sum, with most nutritional metabolites concentrated either in the microgreens or seedlings. Essential amino acids decreased while plant sugars increased from microgreens to seedlings, and carotenoids and essential minerals exhibited different distribution patterns among the developmental stages.

Analysis of 13 Alternaria mycotoxins including modified forms in beer

Sophie Scheibenzuber, Fabian Dick, Stefan Asam, Michael Rychlik

Summary: A multi-mycotoxin LC-MS/MS method was developed to quantify 13 free and modified Alternaria toxins in different beer types, showing no contamination with Alternaria toxins apart from tenuazonic acid (TeA) in concentrations between 0.69 μg/L and 16.5 μg/L in 50 beer samples. The exposure towards TeA through beer consumption was considered relatively low, as the threshold of toxicological concern (TTC) value might not be reached when consuming reasonable amounts of beer.

Characterization of the nutrient composition of German beer styles for the German nutrient database

Larissa E. Pferdmenges, Annett Schroeter, Regina Lohmayer, Lisa Striegel, Michael Rychlik, Alexandra Mueller, Ann-Katrin Meinhardt, Bernhard Trierweiler, Bernd M. Hartmann, Lara Frommherz

Summary: This study revealed significant differences in nutrient composition among different German beer styles, providing an improved basis for understanding the nutritional value of beer. The new data showed higher daily nutrient recommendation percentages for niacin, vitamin B6, and folate, but lower values for alcohol and elements like copper and iodine for the average beer consumer. These findings demonstrate the importance of updated data for future studies on nutritional value calculation in beer consumption.

Expanding the Protecting Group Scope for the Carbonyl Olefin Metathesis Approach to 2,5-Dihydropyrroles

Fabian Huck, Lorenzo Catti, Gian Lino Reber, Konrad Tiefenbacher

Summary: Chiral pyrrolidine derivatives are crucial for natural product synthesis, and carbonyl olefin metathesis is a powerful tool for constructing these building blocks. The supramolecular resorcinarene catalyst enables access to chiral 2,5-dihydropyrroles under Bronsted acid catalysis, even tolerating Lewis-basic protecting groups. The product yield and selectivity in this catalytic system depend on the size of the substrates due to reaction occurring inside a closed cavity.

Summary: The epidemiology of Fusarium Head Blight (FHB) in spring barley was assessed in this study. Soil and spray inoculation were used to increase disease pressure and provoke genotypic differentiation. The evaluation of stable quantitative FHB resistance in barley genotypes was achieved through the quantification of fungal DNA contents and multiple Fusarium toxins. The study also identified the impact of weather conditions on fungal colonization in mature barley grain.

Evolution of Alternaria toxins during the brewing process and the usability of optical sorting methods to reduce mycotoxin concentrations in beer

Marina Brettraeger, Sophie Scheibenzuber, Stefan Asam, Michael Rychlik, Martina Gastl, Thomas Becker

Summary: Brewing experiments were conducted to assess the impact of black-colored grain on Alternaria mycotoxin concentrations throughout the brewing process. The concentrations of 13 different Alternaria mycotoxins were analyzed in each step using liquid chromatography-tandem mass spectrometry, and the toxin concentrations were balanced over the entire process. The results showed that TeA was the only toxin to migrate into the final beer, while other toxins were mainly found in the spent grains. This study suggests that optical sorting of malt batches can be a useful tool for preventing elevated mycotoxin concentrations in the malting and brewing industry.

Reductive cyclodimerization of chalcones: exploring the self-adaptability of galvanostatic electrosynthesis

Mauro Garbini, Andrea Brunetti, Riccardo Pedrazzani, Magda Monari, Massimo Marcaccio, Giulio Bertuzzi, Marco Bandini

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.
