Differential Pathomechanisms of Desmoglein 1 Transmembrane Domain Mutations in Skin Disease

Stephanie E. Zimmer, Takuya Takeichi, Daniel E. Conway, Akiharu Kubo, Yasushi Suga, Masashi Akiyama, Andrew P. Kowalczyk

Summary: This study compares two dominant mutations in the DSG1 gene that cause different skin diseases, revealing distinct pathomechanisms of these mutations. The mutants showed differences in desmosome assembly due to reduced membrane trafficking and lipid raft targeting. The study highlights the importance of DSG1 in epidermal pathophysiology and emphasizes the need for further research on desmosomal cadherins.

Effects of Dupilumab on Itch-Related Events in Atopic Dermatitis: Implications for Assessing Treatment Efficacy in Clinical Practice

Ryoma Kishi, Sumika Toyama, Mitsutoshi Tominaga, Yayoi Kamata, Eriko Komiya, Takahide Kaneko, Yasushi Suga, Kenji Takamori

Summary: This study investigated the effects of Dupilumab treatment on itch-related events in patients with atopic dermatitis (AD). The results showed that Dupilumab improved clinical scores and reduced the levels of inflammatory markers in serum. However, it did not affect the number of intraepidermal nerve fibers (IENFs). Therefore, dynamic changes may be used to assess the efficacy of Dupilumab in treating itch and inflammation in AD patients.

Summary: In this study, T. interdigitale isolates were obtained from Japanese patients and their ITS types were investigated. The results showed that all 29 isolates belong to ITS type II, while one clinical isolate of T. mentagrophytes was similar to ITS type II*. Additionally, a terbinafine-resistant strain of T. interdigitale also belonged to ITS type II.

Summary: A dermatophyte antigen kit (DQT) was introduced in Japan as a diagnostic tool to identify tinea unguium. A study was conducted from July 2022 to February 2023 to evaluate the accuracy of DQT in confirming fungal elements. The results showed that DQT was beneficial for diagnosing onychomycosis where traditional microscopy methods were insufficient.

Human papillomavirus type 94-associated flat warts and review of reported cases

Tamae Usami, Ikuroh Ohsawa, Yasushi Suga, Tsuyoshi Mitsuishi

Summary: In this study, human papillomavirus type 94 was identified in a patient with flat warts using polymerase chain reaction analysis. Dermatoscopic findings of macular brown lesions on the lower leg, showing numerous dotted vessels, supported the diagnosis of flat warts. Previous studies have also detected human papillomavirus type 94 in various skin diseases.

Long Amplification PCR (LA-PCR) Detection of Azole Resistant Trichophyton indotineae

Tsuyoshi Yamada, Honoka Nojo, Rui Kano

Summary: Trichphyton indotineae, a newly designated species, exhibits resistance to terbinafine and azoles. This species has caused an epidemic of dermatophytosis in North India and has spread worldwide. A long amplification PCR method was developed to detect low-susceptibility strains of T. indotineae to azole antifungal agents by targeting the tandem repeat of the CYP51B gene.

Genomic Analysis of Antifungal Drug Resistance Induced in Trichophyton rubrum After Prolonged Culture with Terbinafine

Rui Kano, Honoka Nojo, Hiromitsu Noguchi

Summary: In this study, the researchers induced terbinafine resistance in a T. rubrum strain and compared the genomes of the resistant and susceptible strains. They found that the only variation associated with drug resistance in the resistant strain was a hotspot mutation in the SQLE gene.

In vitro Azole Susceptibility Testing of Japanese Isolates of Terbinafine-Resistant Trichophyton indotineae and Trichophyton rubrum

Summary: Multi-antifungal-resistant strains of Trichophyton indotineae and Trichophyton rubrum have been isolated in Japan. In this study, the in vitro susceptibility of terbinafine-resistant isolates of T. indotineae and T. rubrum to efinaconazole and luliconazole was examined. Results showed varying sensitivity of the isolates to the tested antifungal drugs.
