Summary: Statins are commonly prescribed medications that reduce cholesterol and prevent cardiovascular disease. However, statin-associated musculoskeletal symptoms can affect patient adherence and long-term effectiveness. Genetic variations in SLCO1B1, ABCG2, and CYP2C9 genes have been identified to impact the metabolism and adverse events of statins. This study summarizes the literature supporting these associations and provides therapeutic recommendations based on genotypes to improve the safety, adherence, and effectiveness of statin therapy.

Effect of SLCO1B1 T521C on Statin-Related Myotoxicity With Use of Lovastatin and Atorvastatin

Summary: The study identified an association between the c.521T>C variant allele in the SLCO1B1 gene and statin-induced myotoxicity. Summary: Statin-associated muscle symptoms (SAMS) are common in patients using statin drugs, but not all symptoms are caused by the drugs, negative expectations could also play a role. Physical activity is important for managing cardiovascular disease risk, but the evidence of increased creatine-kinase levels in statin-treated athletes has been wrongly generalized. The potential of reducing sedentary behavior and resistance training has been overlooked in current guidelines.

Reporting of Drug-Induced Myopathies Associated with the Combination of Statins and Daptomycin: A Disproportionality Analysis Using the US Food and Drug Administration Adverse Event Reporting System

Summary: This study evaluated the muscular toxicity of the combination therapy of daptomycin and statins using a large pharmacovigilance database. The results showed that the combination of daptomycin with rosuvastatin, atorvastatin, and simvastatin increased the reporting frequency of myopathy. Additionally, the combination of daptomycin with these statins also increased the frequency of rhabdomyolysis.

A Case Series of Statin-Induced Necrotizing Autoimmune Myopathy

Summary: The use of statins for cardiovascular disease prevention has been increasing over the past decade, but it has also been associated with various side effects, including muscle-related complications. Among these, statin-induced necrotizing autoimmune myopathy (SINAM) stands out due to its unique characteristics and aggressive management. This article presents two cases of SINAM and discusses the clinical aspects of diagnosis, investigation, and treatment. SINAM typically presents with proximal myopathy and elevated creatinine kinase levels that do not improve with discontinuation of statins. Diagnosis is confirmed through biopsy and detection of 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGCR) antibodies. Additional investigations are necessary to rule out other causes of proximal myopathy. In most cases, rechallenging with statins is unsuccessful and immunosuppressive treatment is required.

Statin-associated immune-mediated necrotizing myositis in Native Americans

Summary: This study compared the manifestations of immune-mediated necrotizing myopathy (IMNM) to idiopathic inflammatory myositis (IIM) in Native Americans. It found that IMNM patients were older and more likely to have diabetes, hyperlipidemia, and statin exposure. They also had significantly higher levels of creatine kinase and anti-3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) antibodies, and a higher prevalence of necrotizing myopathy. However, they had shorter disease duration and fewer cutaneous or vascular manifestations compared to IIM patients.

Summary: This study investigated the association between serum PCSK9 levels and subclinical cerebrovascular disease in a community-dwelling Japanese male population. The results showed that higher levels of circulating PCSK9 were significantly associated with an increased prevalence of intracranial artery stenosis, but not with cerebral small vessel disease. Therefore, measuring serum PCSK9 levels may help identify individuals at high risk for cerebrovascular disease in the general population.

Summary: Dietary habits significantly impact healthy life expectancy by affecting cardiovascular disease risk and chronic diseases. The NIPPON DATA80/90 cohort study found that a dietary pattern characterized by high intake of fruits, vegetables, fish (n-3 polyunsaturated fatty acids), and dietary fiber, and low intake of salt and sodium-to-potassium ratio, is associated with a lower risk of cardiovascular disease mortality. These findings can inform effective strategies for cardiovascular disease prevention.

Determinants of double product: a cross-sectional study of urban residents in Japan

The study concludes that high DP at rest is associated with insulin resistance, gamma-glutamyl transferase, and exercise in participants without underlying disease.
