STAT3 for Cardiac Regenerative Medicine: Involvement in Stem Cell Biology, Pathophysiology, and Bioengineering

Heart disease is the most common cause of death in developed countries, but the medical treatments for heart failure remain limited. In this context, the development of cardiac regeneration therapy for severe heart failure is important. Owing to their unique characteristics, including multiple differentiation and infinitive self-renewal, pluripotent stem cells can be considered as a novel source for regenerative medicine. Janus kinase/signal transducer and activator of transcription 3 (JAK/STAT3) signaling plays critical roles in the induction, maintenance, and differentiation of pluripotent stem cells. In the heart, JAK/STAT3 signaling has diverse cellular functions, including myocardial differentiation, cell cycle re-entry of matured myocyte after injury, and anti-apoptosis in pathological conditions. Therefore, regulating STAT3 activity has great potential as a strategy of cardiac regeneration therapy. In this review, we summarize the current understanding of STAT3, focusing on stem cell biology and pathophysiology, as they contribute to cardiac regeneration therapy. We also introduce a recently reported therapeutic strategy for myocardial regeneration that uses engineered artificial receptors that trigger endogenous STAT3 signal activation.

Intrinsic Electrical Remodeling Underlies Atrioventricular Block in Athletes

Summary: The study found that atrioventricular block in athletes is mainly caused by electrical remodeling. Trained animals and mice showed atrioventricular node dysfunction, characterized by prolonged PR intervals and reduced expression of key ion channels. miRNAs such as miR-211-5p and miR-432 were identified as direct regulators of Ca(V)1.2 and HCN4.

Common arterial trunk in a cat: a high- resolution morphological analysis with micro-computed tomography

Summary: A 6-month-old female cat presented with complex cardiac anomalies including biatrial and biventricular dilation, with branching abnormalities in the aorta. The cat died of severe congestive heart failure, with postmortem diagnosis confirming the arterial trunk abnormalities through micro-computed tomography analysis. Micro-computed tomography proved to be a valuable tool for ex vivo diagnosis of complex cardiac anomalies in this case.

c-Myc/microRNA-17-92 Axis Phase-Dependently Regulates PTEN and p21 Expression via ceRNA during Reprogramming to Mouse Pluripotent Stem Cells

Summary: Induced pluripotent stem cells (iPSCs) have great potential in regenerative medicine and disease modeling, but their reprogramming efficiency is still low. In this study, researchers investigated the role of miR-17-92 in iPSC induction. They found that upregulation of miR-17-92 increased reprogramming efficiency, while deletion of the miR-17-92 cluster decreased efficiency. Further analysis revealed that miR-17-92 targets genes PTEN and p21, and their downregulation is disrupted by introducing the 3′ UTR of PTEN and p21, suggesting they act as competing endogenous RNAs (ceRNA). These findings provide insight into the underlying mechanism of iPSC reprogramming.
