Identification of prognostic claudins signature in hepatocellular carcinoma from a hepatocyte differentiation model

Background Loss of terminal differentiation markers and gain of stem cell-like properties are a major hallmark of cancer malignant progression. Identification of novel biomarkers representing tumor developmental progeny and predictive of patients' prognosis would greatly benefit clinical cancer management. Methods Human embryonic stem cells were induced to differentiate into hepatocytes along hepatic lineages. Transcriptomic data from different liver developmental stages were analyzed combining with the RNA-seq data from The Cancer Genome Atlas (TCGA) project. Kaplan-Meier survival analysis and Cox regression analyses were used to analyze the clinical significance in HCC patients. Results A shifted expression pattern of claudin (CLDN) family genes were identified to be closely associated with liver development and tumor progression. Claudins with hepatic features were found to be significantly down-regulated and predicted better prognosis in HCC patients. Conversely, another set of claudins with embryonic stem cell features were found to be significantly up-regulated and predicted worse prognosis in HCC patients. A claudin signature score system was further established by combining the two sets of claudin genes. The newly established claudins signature could robustly predict HCC patients' prognosis in the training, testing, and independent validation cohorts. Conclusions In the present study, we developed a novel embryonic developmental claudins signature to monitor the extent of tumor dedifferentiation in HCC from an in vitro hepatocyte differentiation model. The claudins signature might present a great potential in predicting prognostic significance in HCC as cell surface biomarkers, and provide novel therapeutic targets for precision oncology further in the clinic.

Impacts of climate change on the food-water nexus in central Arizona

Summary: This study used the WEAP-MABIA model to simulate crop production in the main irrigation districts near Phoenix, Arizona, under different warming levels and water allocations. The results showed that crop production will decline in the coming decades, especially under higher warming scenarios, and will further decrease under water shortage measures, potentially reaching almost a 50% reduction by the end of the century.

Chassis Coordinated Control for Full X-by-Wire Four-Wheel-Independent-Drive Electric Vehicles

Summary: A full X-by-wire chassis coordinated control scheme is proposed in this paper by utilizing Direct Yaw-moment Control, Active Front Steering, Anti-Slip Regulation, and Active Roll Control. A vehicle state prediction module is established to generate reference vehicle states and a decentralized event-triggered discrete sliding mode control scheme is developed to track these references by coordinating the subsystems. The hardware-in-the-loop tests show that the proposed scheme can improve vehicle stability, handling performance, comfort, and rollover prevention.

Applying a time-saving and cost-effective postprocessing method to achieve a high-quality perovskite light emitting diode

Summary: This study presents an effective postprocessing method for recrystallizing the perovskite emissive layer. The method involves annealing and plasma treatment, which are time-saving and do not require vacuum tools. Applying this process reduces the grain size of the MAPbBr3 and increases the carrier recombination rate and photoluminescence quantum yields of the perovskite emitters. These results indeed improve the luminance, external quantum efficiency, and lifetime of the devices.

Cloud-Edge-End Intelligence for Fault-Tolerant Renewable Energy Accommodation in Smart Grid

Summary: Smart grid integrates distributed energy resources and massive information to facilitate energy flow in industries. Accommodating renewable energy is crucial for achieving energy efficiency, but optimal policies are difficult to obtain due to intermittency. To capture renewable energy statuses for decision-making, heterogeneous information is collected by end devices in smart grid, posing challenges for existing algorithms. This article proposes a cloud-edge-end computing scheme to efficiently repair missing values and obtain optimal policies in two separate layers using deep learning and deep reinforcement learning algorithms. Simulations on a real power grid dataset demonstrate the effectiveness of the proposed fault-tolerant renewable energy accommodation algorithm.

Screening of heat stress-regulating active fractions in mung beans

Summary: Heat stress caused by high temperatures has adverse effects on the safety and health of humans and animals. This study characterized the components of mung bean that regulate heat stress and found that polyphenols in mung bean have antioxidant and heat stress-regulating effects. Specific polyphenols like vitexin, orientin, and caffeic acid were identified as the main regulators of heat stress, with orientin having the best effect.

Effect of multi-scale reinforcement on fracture property of ultra-high performance concrete

Summary: This paper aims to achieve multi-scale crack control of concrete by using graphene oxide (GO) and macroscopic steel fiber in combination, and to reveal the multi-scale collaborative reinforcement mechanism of GO and steel fiber. The results showed that GO reinforced UHPC had the best fracture property, and steel fiber and GO showed good synergistic crack control effect. GO directly acted on controlling the cracking of concrete matrix at the nanoscale, thus improving the fracture energy of UHPC matrix. A prediction model for fracture energy of straight steel fiber UHPC was established, which showed that the initial fiber-matrix bond strength and slip hardening coefficient had a powerful influence on the UHPC fracture energy.

Summary: Improving wheat quality by adjusting its storage protein genes is crucial due to rising living standards. Gene pyramiding of high molecular weight subunits, NGli-D2 and Sec-1(s) successfully enhanced dough elasticity and improved overall protein composition. The sedimentation values and mixograph parameters were significantly increased, and celiac disease epitopes were reduced, offering potential for better wheat processing quality and reduced risk of celiac disease.

A comparative investigation into the material matching performance of different friction pairs under current-carrying friction

Summary: To improve the material matching performance of carbon skateboard/contact rails, three different contact pairs (powder metallurgy carbon skateboard/stainless steel, copper-impregnated carbon skateboard/stainless steel and copper-impregnated carbon skateboard/tin bronze) were studied for their current-carrying friction performance. Results showed that the copper-base powder metallurgy/stainless steel contacts maintained low and stable contact resistance and friction coefficient under different currents, while the copper-impregnated/stainless steel and copper-impregnated/tin bronze contact pairs were more sensitive to increased currents. Wear mechanisms of the three contact pairs were mainly abrasive wear, oxidative wear, and adhesive wear, according to tribological analysis.

Dynamic response of planar frame sub-structure isolated by friction pendulum bearings subjected to a middle column removal scenario

Summary: The aim of this study is to investigate the dynamic response of planar frame sub-structure isolated by friction pendulum bearings subjected to a middle column removal scenario. The study presents and evaluates the dynamic structural behaviors, load transfer mechanisms, progressive collapse resistance, and dynamic increase factor (DIF) results of FPBs-isolated planar frame. The results show that the compressive arching action (CAA) and catenary action (CA) cannot be fully developed for buildings isolated by FPBs due to the significant reduction in the lateral stiffness of the isolated layer caused by FPBs. The DIF of buildings isolated by FPBs initially decreases and then barely changes with an increase in the stabilized vertical displacement of the failed column. The presented fitted formula for DIF of planar frame isolated by FPBs describes the obtained results well. The friction coefficient and equivalent radius of FPBs show very limited influence on the dynamic response of planar frame isolated by FPBs under progressive collapse, but reinforcing the peripheral constraints of the failed column can enhance the progressive collapse resistance of isolated planar frame.

An Intention Recognition Method Based on Fuzzy Belief-Rule-Base

Summary: This paper proposes a new information processing method called fuzzy Belief-Rule-Base (BRB), which addresses the deficiency of traditional intention recognition methods in handling uncertain information. The method combines the advantages of fuzzy sets and Dempster-Shafer (DS) theory. Experimental verification shows that the fuzzy belief-rule-base achieves high accuracy, effectiveness, and reliability under the condition of small samples.

Construction of Flexible, Self-Supporting, and In-Plane Anisotropic PEDOT:PSS Thermoelectric Films via the Wet-Winding Approach

Summary: Flexible and self-supporting PEDOT:PSS thick films with evident thermoelectric (TE) property anisotropy were developed through a wet-winding approach. Highly oriented and ordered PEDOT:PSS chain packing facilitated intra-chain charge transport and resulted in anisotropic films with a power factor 2.4 times higher than isotropic counterparts. The fiber-wound films also exhibited high mechanical strength and excellent stability, allowing the assembly of a TE generator with a good match between optimal property direction and temperature difference direction.

Discovery of a photoactivatable dimerized STING agonist based on the benzo[b]selenophene scaffold

Summary: This study introduces a novel dimerized STING agonist, diBSP01, which shows promising binding and activation properties for STING in vitro with the benzo[b]selenophene scaffold. Additionally, the study presents the development of the first photoactivatable STING agonist, caged-diBSP01, which demonstrates no biological potency without light stimulation but regains its STING agonistic activity after 400 nm irradiation. The optically controlled in vivo anticancer activity of caged-diBSP01 is also validated in a zebrafish xenograft model.
