Summary: The mortality of patients with pancreatic ductal adenocarcinoma (PDAC) is strongly associated with metastasis, and transcriptional and epigenetic rewiring can contribute to the metastatic process. This study shows that HDAC2 plays a crucial role in undifferentiated PDAC by controlling cell cycle and metastasis. HDAC2 maintains the metastatic program by controlling the expression of several prosurvival receptor tyrosine kinases connected to mesenchymal PDAC, and impairs the tumor-suppressive arm of the TGFI3 pathway.

Summary: This study investigated the association of eight variants of four matrix metalloproteinase (MMP) genes with ischemic stroke (IS) and examined whether interactions among these single nucleotide polymorphisms (SNPs) increase the risk of IS. The results showed that an MMP-9 rs17576 polymorphism is associated with increased IS risk in the Han Hakka population, and there is also an interaction between MMP-9 rs17576 and MMP-12 rs660599 that is associated with increased IS risk.

It is identified that miR-467a-3p and miR-874-5p can respectively mediate the stemness and myogenesis of satellite cells (SCs) by performing bioinformatics analysis. EVs loaded with ant-467a and ant-874 are more effective than their naked counterparts in promoting the stemness and myogenesis of SCs.

Summary: An autoimmune condition called systemic lupus erythematosus (SLE) is characterized by overactive B cells and the production of harmful autoantibodies. Current treatments for SLE are ineffective or have adverse effects. The dysregulation of B cell activation plays a crucial role in the development of SLE. In this study, antagomiR-7 was used to target and suppress PTEN mRNA in B cells, leading to reduced B cell hyperresponsiveness and prevention of lupus onset. The development of a sialic acid-poly (D, L-lactide-co-glycolide) (SA-PLGA) nano delivery system helped deliver antagomiR-7 specifically to splenic B cells, addressing the challenges of miRNA stability and targeted delivery in vivo. The results demonstrated the therapeutic potential of inhibiting miR-7 for the treatment of SLE.

Challenges and feasibility of using miRNAs as therapeutic agents are discussed, along with updates on the novel neuro-miRNAs related to therapy against neurotoxicity induced by amyloid beta and Parkinson's disease.

Summary: The study showed that knockout of miR-29a in rats aggravated neurological damage in experimental stroke rat models by promoting M1 microglial polarization and increasing glutamate release. MiR-29a-5p levels decreased in the blood and brain tissue of rat MCAO models, as well as in the peripheral blood of acute stroke patients. Additionally, miR-29a knockout enhanced infarct volume and showed M1 polarization of microglia in the MCAO rat brain.

Summary: By measuring circulating cytokines in the plasma, we identified the alterations of cytokines in patients with super-acute ischemic stroke (AIS). Comparing to healthy controls, certain cytokines in AIS patients were significantly changed, and lymphocytes and neutrophils played a pivotal role in the inflammation of AIS.
