Rapid Inflammasome Activation following Mucosal SIV Infection of Rhesus Monkeys

The earliest events following mucosal HIV-1 infection, prior to measurable viremia, remain poorly understood. Here, by detailed necropsy studies, we show that the virus can rapidly disseminate following mucosal SIV infection of rhesus monkeys and trigger components of the inflammasome, both at the site of inoculation and at early sites of distal virus spread. By 24 hr following inoculation, a proinflammatory signature that lacked antiviral restriction factors was observed in viral RNA-positive tissues. The early innate response included expression of NLRX1, which inhibits antiviral responses, and activation of the TGF-beta pathway, which negatively regulates adaptive immune responses. These data suggest a model in which the virus triggers specific host mechanisms that suppress the generation of antiviral innate and adaptive immune responses in the first few days of infection, thus facilitating its own replication. These findings have important implications for the development of vaccines and other strategies to prevent infection.

Soil heavy metal pollution from Pb/Zn smelting regions in China and the remediation potential of biomineralization

The effect of extracellular polymeric substances (EPS) of iron-oxidizing bacteria (Ochrobactrum EEELCW01) on mineral transformation and arsenic (As) fate

Summary: Extracellular polymeric substances (EPS) play a crucial role in communication and material exchange between iron-oxidizing bacteria and the external environment, leading to the production of iron (oxyhydr) oxides that reduce the availability of arsenic (As). EPS secreted by iron-oxidizing bacteria (Ochrobactrum EEELCW01) primarily consists of polysaccharides, followed by smaller amounts of proteins. Low concentrations of As promote EPS secretion, and its composition includes polysaccharides, proteins, and a small amount of nucleic acids. The adsorption of As is influenced by functional groups in EPS, and XPS analysis reveals that As3+ is bound to EPS. The mineralization process of Ochrobactrum sp. results in the formation of ferrihydrite, goethite, galena, and siderite minerals, with siderite and goethite becoming the main mineral phases over time. As preferentially adsorbs to the edges of iron minerals. This study provides insights into the biomineralization mechanism of iron-oxidizing bacteria and As remediation in the environment.

Summary: This study investigated the effects of shading treatment on gene expression and chlorogenic acid (CGA) content in Lonicera macranthoides Hand.-Mazz. The results showed that shading treatment significantly decreased the CGA content in LM leaves, increased carotenoid content, and decreased soluble sugar and starch contents. Light provides energy and material for CGA accumulation in LM and affects the expression of CGA accumulation-related genes.

Altered Cytokine Production in Human Intervillous Blood T Cells in Preeclampsia

Summary: This study aimed to investigate the differences in T cell phenotype and function in uterine intervillous blood (IVB) between normal pregnancy and preeclampsia, and to explore the immunologic basis of preeclampsia. The results showed that preeclampsia was associated with a greater pro-inflammatory cytokine ratio in uterine IVB T cell subsets, indicating an inflammation-related mechanism of preeclampsia. Therefore, IVB sampling is a useful tool for studying human uterine T cell biology and may provide insights for immunotherapeutic strategies for preeclampsia.

Litsea Males Are Better Adapted to Pb Stress Than Females by Modulating Photosynthesis and Pb Subcellular Distribution

Summary: This study investigated the sex-specific responses of Litsea cubeba under lead (Pb) stress. The results showed that male plants exhibited greater adaptability to Pb stress by enlarging their leaf area and enhancing photosynthesis. Furthermore, more Pb reached the organelle fraction and damaged chloroplasts and mitochondria in female leaves under high-level Pb treatments. These findings provide a theoretical foundation for the utilization of L. cubeba in the phytoremediation of Pb-polluted soil.

Summary: The development of vaccines that protect vulnerable populations from SARS-CoV-2 is a public health priority. This study tested different adjuvants and SARS-CoV-2 antigens to identify a combination that induces antibodies and protection in both young and aged mice. An oil-in-water emulsion containing carbohydrate fatty acid monosulphate derivative (CMS:O/W) was found to be the most effective in enhancing immune response and protection across age groups.

Small Area Forecasting of Opioid-Related Mortality: Bayesian Spatiotemporal Dynamic Modeling Approach

Summary: This study developed Bayesian spatiotemporal dynamic models to predict opioid-related mortality at granular geographical and temporal scales for Massachusetts. The models showed better accuracy in predicting urban areas compared to rural areas with sparse population. The predictions suggested a stabilizing pattern in opioid-related overdose mortality for 2020 and 2021 in Massachusetts if there were no disruptive changes to the observed trends. These models provide valuable information for preemptive public health decision-making and resource allocation.

Summary: SARS-CoV-2 vaccines have reduced protection against the acquisition of the BA.5 subvariant, but remain effective against severe disease. The immune correlates of protection against BA.5 are unknown. This study demonstrates that both homologous and heterologous regimens involving Ad26.COV2.S and SpFN vaccines provide robust protection against a mismatched BA.5 challenge in macaques.
