Summary: Elevated expression of eIF3C in ICC tissues was associated with impaired cellular functions and worse clinicopathologic features. High levels of eIF3C or KI67 were predictive of shorter survival and higher recurrence rates in ICC patients, indicating their potential value as prognostic markers.

Summary: This study characterizes the bacterial metagenomics signature of intrahepatic cholangiocarcinoma (ICC) tissues and investigates their potential as anticancer agents. The presence of intratumoral bacteria, including P. fungorum, was found in multiple cell types. P. fungorum was shown to possess antitumor activity by inhibiting tumor growth through alanine, aspartate, and glutamate metabolism.

Summary: This study developed a novel model to predict lymph node metastasis in ICC patients, as well as predict their 5-year overall survival and disease-specific survival rates. The model performed well in both training and validation datasets, providing valuable information for clinical decision-making prior to resection of ICC.

Summary: This study investigated the incidence trend and prognosis of intrahepatic cholangiocarcinoma (ICC) and found that its incidence has been increasing in recent years while the prognosis remains poor. Surgery is beneficial for all stages of ICC, and lymph node dissection improves the prognosis for patients with positive lymph nodes.

Summary: This study characterizes the molecular classification and tumor microenvironment of intrahepatic cholangiocarcinoma (iCCA) using single cell RNA-sequencing. The authors identify S100P and SPP1 as markers for patient classification and demonstrate that different subtypes of iCCA have distinct immune cell infiltration and survival rates.

Summary: The study reveals the role of VIRMA in intrahepatic cholangiocarcinoma (ICC) and its mechanism of action. VIRMA stabilizes the expression of TMED2 and PARD3B through the m6A-HuR-mediated mechanism, promoting the development of ICC. These findings suggest that VIRMA and its signaling pathway could be potential therapeutic targets for ICC treatment.

Summary: This study dissected the tumor ecosystem in intrahepatic cholangiocarcinoma (ICC) treated with immune checkpoint blockades (ICBs) using single-cell RNA sequencing data and immunofluorescence staining. The results revealed a tighter cellular communication network and the involvement of VEGF signaling mediated by PGF-VEGFR1 in ICC following ICBs. SPP1(+) tumor-associated macrophages (TAMs) were identified as an adverse prognostic indicator for ICC patients.

Summary: This study investigates the expression of specific cells in the tumor microenvironment of intrahepatic cholangiocarcinoma (ICC) and their relationship with clinicopathological characteristics and prognosis. The results suggest that infiltration of Treg cells, CD8+ T cells, and macrophages is associated with ICC relapse. High ratio of FoxP3(+) Treg cells in the tumor tissue predicts longer relapse-free survival (RFS) and serves as an independent favorable prognostic factor for ICC patients. Among Treg cell subtypes, TregIII in peripheral blood is correlated with RFS of ICC patients.

Summary: This study showed that early-relapse hepatocellular carcinoma (HCC) tumors exhibit innate-like CD8(+) T cells with overexpression of KLRB1 (CD161) and a low cytotoxic state, unlike the exhausted state seen in primary HCC. These cells were associated with a worse prognosis. Additionally, potential immune evasion mechanisms were identified in recurrent tumor cells, which dampen dendritic cell antigen presentation and recruit innate-like CD8(+) T cells.

Summary: This study investigated the value of circulating tumor cells (CTCs) as predictors of tumor recurrence following liver transplantation (LTx) in hepatocellular carcinoma (HCC) patients. Results showed that post-operative CTC count >= 1 per 5 mL blood could be a potential biomarker for predicting tumor recurrence, especially in patients with no detectable CTCs before LTx and negative tumor serological markers. Moreover, post-operative serial CTC detection may be useful in post-surgical surveillance for HCC recurrence.

Summary: The ChimeraX(R)-i120 platform successfully combines CTC detection with single-cell molecular analysis, demonstrating high sensitivity, accuracy, and reproducibility. High CTC-positive rates were found in five types of cancer, while CTCs were rarely detected in healthy donors. In hepatocellular carcinoma patients, CTC status was significantly associated with tumor characteristics, prognosis, and treatment response.

Summary: The single-cell DNA library preparation method developed in this study, known as scDPN, eliminates the need for preamplification and allows for high-throughput CNV detection. It shows lower amplification bias and noise compared to the MDA method, and demonstrates high sensitivity and accuracy in evaluating cell lines and tumor tissues. Additionally, it was used to identify clonal subpopulations in hepatocellular carcinoma (HCC) samples, revealing clonal selection during tumor recurrence. Overall, this approach provides a comprehensive and scalable solution for understanding genome heterogeneity and evolution.
