Summary: SARS-CoV-2 continues to spread globally, with evolving variants that are more transmissive and show reduced vaccine efficacy. Targeting host proteins such as DDX3 with RK-33 can effectively reduce viral load and downregulate SARS-CoV-2 genes, making it a potential strategy to control infection.

Summary: The use of antibody arrays revealed that the RNA helicase DDX3 plays a role in modulating the expression of secreted signaling factors in oral squamous cell carcinoma (OSCC) cells. Furthermore, the interaction between DDX3 and the signal recognition particle (SRP) affects the expression of AREG mRNA, contributing to the progression of tumors.

Summary: The study showed that a strong reduction of DDX3 protein leads to similar changes in vitro and in vivo, including cell cycle deregulation and pathway alterations, while a more moderate downregulation resulted in discordant results between in vitro and in vivo, indicating the significant impact of DDX3 protein levels on cell phenotype.

Summary: The translation inhibitor Rocaglamide A shows promising antitumor activity by clamping eIF4A and DDX3 onto polypurine RNA for selective translational repression. Analysis of a de novo-assembled transcriptome from the natural source of RocA, plant Aglaia, reveals the amino acid critical for RocA binding. The dominant-negative effect of RocA on eIF4A and DDX3 leads to strong translational repression in cancer cells, indicating its tumor toxicity.

Summary: This study demonstrates the selective induction of cell death in latent HIV-1-infected cells by DDX3 inhibitors, and provides evidence for the reduction of the inducible HIV-1 reservoir in CD4+ T cells from PLWHIV. The pharmacological targeting of DDX3 shows promise in reversing HIV-1 latency and eliminating the latent reservoir.

Summary: The DDX3 gene in chinook salmon plays a role in enhancing innate immunity and antiviral response by activating the type I IFN signaling pathway and inducing the expression of type I IFN. This study reveals the biological role of fish DDX3 in regulating innate immunity and antiviral effect, providing a scientific basis for understanding the antiviral regulation mechanism of innate immunity in fish. These findings can guide the breeding of disease-resistant salmon and prevent viral diseases.

Summary: Antiretroviral therapy (ART) is effective in reducing HIV-1 infection. However, the emergence of treatment-refractory variants remains a challenge. HIV-1 uses cellular helicases, including DDX3X, to facilitate its replication. Exploring these helicases may reveal new therapeutic targets and help prevent viral escape.

Summary: The study reveals that the mismatch repair protein MutS beta is enriched at telomeres in ALT cancer cells, preventing the accumulation of G-quadruplex (G4) structures and R-loops, thus maintaining telomere integrity during alternative lengthening of telomeres.

Summary: In this study, researchers identified KLHL29 as a crucial tumor suppressor that regulates chemosensitivity in triple-negative breast cancer (TNBC). Low levels of KLHL29 were associated with unfavorable prognoses. Mechanistically, KLHL29 suppressed TNBC growth by degrading DDX3X and inhibiting cell cycle progression. The combination of DDX3X inhibitor RK33 and platinum-based chemotherapy showed promising results in suppressing TNBC.

Summary: The G-quadruplex DNA structure in cancer cells requires the involvement of DNA helicase for unwinding, and the helicase can bind to and unwind G4DNA. The binding and unwinding of G4DNA by helicase is influenced by the single-stranded DNA terminals in G4DNA.

Summary: YTHDC2 is an important regulator for breast cancer tumorigenesis and metastasis, and its expression level is significantly correlated with tumor stage. Knockdown of YTHDC2 attenuates sphere-forming and metastatic ability of breast cancer cells, but the mechanism by which YTHDC2 regulates these phenotypes is different between different subtypes of breast cancer.
