The thermodynamic stability of 1 : 1 and 2 : 1 boron-nitrogen (B -> N) adducts formed between aromatic boronic esters with mono- and diamines was studied in solution by NMR and UV-vis spectroscopy with association energies (Delta G degrees) ranging from -11 to -28 kJ mol(-1). The effect of different substituents in the boronic ester, the nature of the diamine linker, and the effect of the solvent was explored. Stable 2 : 1 B <- N adducts with diamines such as 1,3-diaminopropane were produced in solutions of hydrogen-bonding acceptor solvents (acetonitrile and ethyl acetate), which can be isolated in the solid state as crystalline solvates, whereas the use of noncoordinating solvents such as 1,2-dichloroethane afforded mainly 1 : 1 B <- N adducts. In suitable combinations, aromatic bis-pyridyl diamines produced stable 2 : 1 B <- N adducts that were isolated either as solvent-free solids, solvates, or cocrystals. In these crystalline forms, double-tweezer hosts were observed with an exceptional syn/anti conformational guest-adaptability driven by simultaneous donor-acceptor and C-H...pi interactions in the tweezer cavities, resembling preorganized covalent tweezer hosts. Interestingly, cocrystals with electron-rich guests such as tetrathiafulvalene and pyrene showed non-centrosymmetric crystal lattices with infinite pi-stacked donor-acceptor columns.

Molecular Dynamics Studies of Aromatic Guests in Three Isostructural Inclusion Compounds with Molecular Boron-Nitrogen Hosts

Eva Cecilia Vargas-Olvera, Frank Jose Salas-Sanchez, Abraham Colin-Molina, Salvador Perez-Estrada, Braulio Rodriguez-Molina, Jose Alejandre, Gonzalo Campillo-Alvarado, Leonard R. MacGillivray, Herbert Hopfl

Summary: This study examined the dynamics of three guests (benzene, naphthalene, and xylene) included in isostructural cavities. The results showed that benzene is highly mobile with fast in-plane jumps, while naphthalene experiences 180-degree jumps at high temperatures. Xylene can only undergo 120-degree in-plane jumps at temperatures close to decomposition due to the methyl groups acting as effective stoppers.

Zinc(II) complexes constructed from an adamantane-functionalized pyridine Schiff base-Influence of the counterion on the supramolecular organization by means of C-H ••• O, C-H ••• N, C-H ••• π and π•••π interactions

Tushar S. Basu Baul, Anurag Chaurasiya, Maria G. Vasquez-Rios, Herbert Hopfl

Summary: Four zinc(II) complexes were successfully synthesized using a specific ligand, with their structures characterized by various analytical techniques and confirmed by single crystal X-ray diffraction studies. The molecular arrangements of the complexes were found to be influenced by different anions attached to the zinc ions, as revealed through intermolecular interactions analysis.

Nanoconfinement of a Pharmaceutical Cocrystal with Praziquantel in Mesoporous Silica: The Influence of the Solid Form on Dissolution Enhancement

Reynaldo Salas-Zuniga, Karina Mondragon-Vasquez, Sergio Alcala-Alcala, Enrique Lima, Herbert Hopfl, Dea Herrera-Ruiz, Hugo Morales-Rojas

Summary: Nanoconfinement is a recent strategy to improve solubility and dissolution rate of active pharmaceutical ingredients (APIs) with poor biopharmaceutical properties. Combining cocrystals of racemic praziquantel (PZQ) with glutaric acid (PZQ-GLU) and confining them in mesoporous silica material leads to increased solubility and dissolution rate. The confined PZQ-GLU phase shows transient supersaturation behavior in simulated gastric fluid, providing a dissolution advantage over pristine PZQ.

Summary: A series of 15 novel 1,3-thiazole amide derivatives of the pentacyclic triterpene Ochraceolide A were synthesized, characterized, and evaluated in vitro against human cancer cell lines. The 5-nitrofuramide derivative showed better cytotoxic and antiproliferative activity than other compounds, while the butyramide derivative exhibited higher selectivity against SiHa cells.

Summary: This study focused on the synthesis of N-alkyl-arylsulfonamide derivatives of lupeol acetate for the treatment of Trichomonas vaginalis, with two compounds showing promising efficacy against both sensitive and resistant strains. These compounds have the potential to serve as novel antiparasitic agents.

Tailoring Chlorthalidone Aqueous Solubility by Cocrystallization: Stability and Dissolution Behavior of a Novel Chlorthalidone-Caffeine Cocrystal

Christian Rodriguez-Ruiz, Pedro Montes-Tolentino, Jorge Guillermo Dominguez-Chavez, Hugo Morales-Rojas, Herbert Hopfl, Dea Herrera-Ruiz

Summary: A cocrystal of the antihypertensive drug chlorthalidone and caffeine was synthesized, which showed improved solubility and dissolution behavior compared to chlorthalidone alone. The stability and dissolution properties of the cocrystal make it a promising candidate for the development of solid dosage forms with lower drug doses and fewer adverse effects.

Synthesis and structures of diorganotin(IV) Schiff base complexes [R2Sn(L)Cl2] and their proliferative responses on breast cancer cells

Tushar S. Basu Baul, Bietlaichhai Hlychho, Maheswara Rao Addepalli, Andrew Duthie, Olivia Sarkar, Arpan Dey Bhowmik, Ansuman Chattopadhyay, Maria G. Vasquez-Rios, Herbert Hoepfl

Summary: The synthesis and characterization of five diorganotin (IV) complexes with different ligands have been reported. The complexes exhibit two distinct configurations, with six-coordinate C2SnN2Cl2 environments. The molecular arrangements in the solid-state structures are determined by various weak interactions. The anti-proliferative activity of the complexes against breast cancer cells has been evaluated, with one complex showing the best activity and another significantly less active. The mechanism of cell death was found to be apoptosis.
