Emerging Viruses without Borders: The Wuhan Coronavirus

The recently emerged coronavirus in Wuhan, China has claimed at least 6 lives as of January 22 and infected hundreds if not thousands of individuals. The situation has drawn international attention, including from the virology community. We applaud the rapid release to the public of the genome sequence of the new virus by Chinese virologists, but we also believe that increased transparency on disease reporting and data sharing with international colleagues are crucial for curbing the spread of this newly emerging virus to other parts of the world.

Summary: The emergence of new Omicron subvariants, including BQ.1, BQ.1.1, BA.4.6, BF.7, and BA.2.75.2, has shown enhanced neutralization resistance against sera from vaccinated healthcare workers and COVID-19 patients. The mutations N460K, K444T, and F486S play a significant role in driving the increased neutralization resistance of these subvariants. These findings provide insights into the evolution of SARS-CoV-2 Omicron subvariants.

Summary: Viruses have posed various challenges for humanity, but virology research has played a crucial role in reducing disease burden. The COVID-19 pandemic has brought unprecedented attention to the field, with concerns about conducting research on human pathogens and uncertainties regarding the origin of SARS-CoV-2. This has led to public confusion and misguided criticism of virology. This article aims to promote rational discourse by explaining gain-of-function approaches, discussing the possible origins of SARS-CoV-2, and outlining the regulatory structures in the United States that oversee virological research. The intention is to provide guidance to policymakers and maintain evidence-based discourse to address public concern and support vital virology research.

Determinants and Mechanisms of the Low Fusogenicity and High Dependence on Endosomal Entry of Omicron Subvariants

Summary: The rapid spread and strong immune evasion of the SARS-CoV-2 Omicron subvariants has raised serious concerns for the global COVID-19 pandemic. The mutations H655Y and T547K in the C-terminal S1 subunit play critical roles in determining the low fusogenicity and enhanced dependence on the endosomal entry pathway of Omicron. The H655Y mutation also determines these characteristics in other Omicron subvariants, including BA.5 and BA.2.75.

Although virology research has made significant progress in combating these diseases through vaccines and antivirals, the COVID-19 pandemic has brought unprecedented attention to the field, with concerns regarding the safe conduct of research on human pathogens and uncertainties over the origins of SARS-CoV-2. This article aims to promote rational discourse by explaining gain-of-function approaches in science, discussing the possible origins of SARS-CoV-2, and outlining the current regulatory structures in place for virological research in the United States. The goal is to aid policymakers in navigating these controversial issues and ensure balanced, evidence-based discussions that support vital virology research.

Discovery of novel papain-like protease inhibitors for potential treatment of COVID-19

Summary: The emergence of new SARS-CoV-2 variants highlights the urgent need for more effective therapeutic agents to prevent COVID-19 outbreaks. SIMR3030 has been identified as a potent inhibitor of SARS-CoV-2 PLpro, showing the ability to inhibit inflammatory markers and viral activity. This study lays the foundation for developing new drugs to tackle the COVID-19 pandemic and may pave the way for the development of novel therapeutics for future outbreaks of new SARS-CoV-2 variants or other Coronavirus species.

STING Agonist-Derived LNP-mRNA Vaccine Enhances Protective Immunity Against SARS-CoV-2

Summary: Lipid nanoparticle (LNP)-mediated mRNA COVID-19 vaccines have provided large-scale immune protection. To efficiently deliver antigens and activate antigen-presenting cells, a library of SALs was synthesized and formulated into LNPs for mRNA delivery. SAL12-LNPs were identified as the most potent in delivering Spike glycoprotein mRNA while activating the STING pathway in DCs. Two doses of SAL12 S-LNPs elicited potent neutralizing antibodies against SARS-CoV-2 in mice.

Effects of activation and assisted reproduction techniques on the composition, structure, and properties of the sauger (Sander Canadensis) spermatozoa plasma membrane

Summary: The sperm plasma membrane is crucial for fertilization, but assisted reproduction techniques can harm it and reduce fertility. While there has been extensive research on fish sperm membrane, few studies have examined the effects of cryopreservation and other assisted reproduction techniques on membrane composition, characteristics, and organization. This study investigated the impact of testicular harvest, strip spawning, and cryopreservation on the sperm plasma membrane using Sauger sperm. The results revealed that these techniques can significantly affect the membrane, providing potential biomarkers for sperm quality and insight into sub-fertility caused by assisted reproduction techniques.

Enhanced evasion of neutralizing antibody response by Omicron XBB.1.5, CH.1.1, and CA.3.1 variants

Summary: This study demonstrates the nearly complete escape of XBB.1.5, CH.1.1, and CA.3.1 variants from neutralizing antibodies stimulated by mRNA vaccine or BA.4/5 wave infection, but neutralization can be rescued by a bivalent booster containing BA.5. CH.1.1 and CA.3.1 also show strong immune escape from monoclonal antibody S309. Furthermore, the spike proteins of XBB.1.5, CH.1.1, and CA.3.1 exhibit enhanced fusogenicity and processing compared to BA.2.

Selective suppression of de novo SARS-CoV-2 vaccine antibody responses in patients with cancer on B cell-targeted therapy

Summary: In this study, we evaluated the antibody responses induced by the SARS-CoV-2 ancestral virus and Omicron variant vaccines in 57 patients with B cell malignancies, both before and after booster immunization. We found that a significant proportion of vaccinated patients were seronegative before the booster, especially those receiving active cancer therapies. Booster immunization was able to induce detectable antibodies in some seronegative patients, but the overall benefit was more noticeable in patients who were already seropositive and not receiving active therapy. We also observed that while the antibody levels against the ancestral virus and Omicron variant were generally concordant among individual patients, neutralizing antibodies against Omicron tended to be reduced. Interestingly, we found comparable levels of Epstein-Barr virus (EBV)- and influenza-reactive antibodies in all patients, including those unable to generate detectable antibodies against SARS-CoV-2 spike, suggesting that B cell-targeting therapies primarily impair de novo antibody production rather than preexisting antibody levels.

Neutralization escape of Omicron XBB, BR.2, and BA.2.3.20 subvariants

Summary: New Omicron subvariants are continuously emerging worldwide. The XBB subvariant, a recombinant virus between BA.2.10.1.1 and BA.2.75.3.1.1.1, as well as the BA.2.3.20 and BR.2 subvariants with distinct mutations from BA.2 and BA.2.75, are increasing in proportion. Antibodies induced by 3-dose mRNA booster vaccination and previous BA.1-and BA.4/5-wave infection effectively neutralize BA.2, BR.2, and BA.2.3.20, but have reduced efficiency against XBB. The BA.2.3.20 subvariant exhibits enhanced infectivity in lung-derived CaLu-3 cells and 293T-ACE2 cells. Continued monitoring of immune escape and tissue tropism of emerging Omicron subvariants is crucial.

Summary: Coronaviruses can cross species barriers and infect animals and humans. A sensitive and specific ELISA was developed to detect antibodies to SARS-CoV-2. The study found antibodies to SARS-CoV-2 in deer populations in Ohio, highlighting the need for surveillance programs to monitor susceptible wildlife species globally.

Increased COVID-19 Mortality and Deficient SARS-CoV-2 Immune Response Are Not Associated with Higher Levels of Endemic Coronavirus Antibodies

Summary: This study evaluated the impact of pre-existing common cold coronavirus antibodies on SARS-CoV-2 immune responses and disease severity in hospitalized COVID-19 patients. The results showed a positive correlation between levels of common cold coronavirus antibodies and SARS-CoV-2 antibodies. Higher levels of common cold coronavirus antibodies were associated with better clinical outcomes in COVID-19 patients. Age, cancer, and immunosuppression were found to be associated with higher mortality and reduced levels of SARS-CoV-2 and common cold coronavirus antibodies.
