A feasible image-based colorimetric assay using a smartphone RGB camera for point-of-care monitoring of diabetes

Urinary glucose determination using a glucose test strip is simple and convenient in daily self-monitoring of diabetes. However, diabetic patients exhibit acquired impaired color vision (ICV), which results in the inability to discriminate between hues. Even with the assistance of a color chart, it is still not easy for these patients to read the urinary glucose results with the naked eye. In this study, a smartphone camera using an image-based colorimetric detection method was successfully developed for quantitative analysis of urine glucose. A horseradish peroxidase-hydrogen peroxide-3,3'5,5'-tetramethylbenzidine (HRP-H2O2-TMB) system was optimized for a reliable and gradual color fading process via a glucose oxidase (GOD) catalyzed oxidation reaction. The color changes of the peroxidase-H2O2 enzymatic reactions in the 96-well microplate were captured by a smartphone RGB camera with subsequent detection of red, green, and blue (RGB) intensities decreasing at each image pixel. The highly quantitative relationships between the glucose concentrations and the color characteristic values of the blue channel of the captured images were successfully established. The high accuracy of this method was demonstrated in urine glucose measurements with a linear response over the 0.039 mg mL(-1) to 10.000 mg mL(-1) glucose concentration range and a 0.009 mg mL(-1) detection limit. The method has great potential as a point-of need platform for diabetic patients with defective color vision and features high accuracy and low cost.

RGD and Scutellarin Conjugate (WK001) Targeting Platelet Glycoprotein IIb/IIIa Receptor Protects from Myocardial Ischemia/ Reperfusion Injury: Synthesis, Characterization, and Bioactivity Evaluation

Summary: We synthesized a novel peptide, WK001, by modifying the RGD structure to combine with scutellarin. Experimental and docking evidence showed that WK001 provides immediate and potent platelet inhibition, surpassing the inhibition of eptifibatide and scutellarin in platelet aggregation. In vivo studies demonstrated that WK001 administered intravenously prevents thrombus formation and attenuates myocardial fibrosis progression. Therefore, WK001 has the potential to be developed as an antiplatelet drug for thrombosis-associated diseases, such as stroke and myocardial infarction.

New insight of complement system in the process of vascular calcification

Summary: The complement system defends against pathogenic microbes and modulates immune homeostasis. Dysregulation or activation of the complement system contributes to autoimmune and cardiovascular diseases, particularly those related to vascular calcification. This review summarizes current evidence on the activation of the complement system in vascular calcification and discusses its complex interactions with various processes involved in the development of this condition. Understanding the relationship between the complement system and vascular calcification can provide insights for slowing its progression.

The study also addresses the concern of cell tracking or AuNP tracking and improves the accuracy and precision of imaging and target delivery using zwitterionic and glucose-functionalized AuNPs. This is the first study to noninvasively visualize the infiltration of macrophages into MI hearts using CT.

Insights into the Mechanism of Supramolecular Self-Assembly in the Astragalus membranaceus-Angelica sinensis Codecoction

Summary: Astragalus membranaceus and Angelica sinensis are a classic herb pair used to treat myocardial fibrosis. This study reports the formation of supermolecules, called AA-NPs, from the codoping reactions of these herbs. The self-assembly mechanism of AA-NPs was investigated, and it was found that AA-NPs were effective in treating myocardial fibrosis by improving cardiac function, reducing collagen deposition, and inhibiting endothelial-to-mesenchymal transition.

Summary: Radix Astragali (RA) is a commonly used Asian herbal therapy or food supply with astragalosides and flavonoids as its major components. This study investigated the bioaccessibility of these compounds during the process of oral digestion and examined their effects on intracellular reactive oxygen species levels in a human arterial endothelial cells model. The results indicate that the acetyl biotransformation of RA in the small intestine directly affects the response to oxidative stress, providing insights into the multi-component action of oral RA in cardiovascular health care.

Summary: In this study, a fluorescent biosensing system for high sensitivity detection of MT was established using a new Ag+-stabilized DNA triplex probe (Ag+-SDTP) combined with catalytic hairpin assembly (CHA) and CRISPR/Cas12a signal enhancements. The sensing method is able to selectively detect MT from other non-specific molecules and can achieve low level detection of MT in diluted human serums, demonstrating its potential for monitoring disease-specific MT biomarker at trace levels.

CRISPR-empowered electrochemical biosensor for target amplification-free and sensitive detection of miRNA

Summary: We developed a CRISPR-empowered electrochemical biosensor, named PER-E-CRISPR, for amplification-free and sensitive detection of miR-21. The biosensor combines the advantages of CRISPR/Cas13a and primer exchange reaction (PER) for target amplification. Under optimized conditions, the PER-E-CRISPR assay exhibits a linear range from 10-13 to 10-7 M and a limit of detection of 30.2 fM. The biosensor shows excellent performance in actual plasma, indicating promising prospects for miRNA detection in molecular diagnosis.

Application and evaluation of molecular docking for aptamer and small molecular interaction- A case study with tetracycline antibiotics

Summary: Molecular docking (MD) analysis is widely used for studying the interaction between aptamers and small molecules. This study established steady-state aptamers of tertiary structures (SATS) using specific aptamers of tetracycline antibiotics as docking models. Molecular docking results revealed multiple binding sites in the SATS of aptamers, with significant variations in binding free energy (BFE) and docking score (DS). The study also proposed a method for MD analysis based on SATS, providing insights into binding mode and predicting binding sites.

The construction of highly selective surface molecularly imprinted polymers based on Cu(II) coordination for the detection of bisphenol A

Summary: In this study, a highly selective molecularly imprinted polymer Cu-MIPs@CS was designed and synthesized for bisphenol A (BPA). Cu-MIPs@CS showed significantly enhanced imprinting factor and selective factor towards BPA compared to MIPs@CS. It also exhibited superior discrimination ability between BPA and its structural analogue. Additionally, Cu-MIPs@CS were successfully applied as a solid phase extraction adsorbent for the determination of BPA in drinking water samples.
