Almost 50% of infertility cases are associated with human male infertility. The sperm membrane is a key structure influencing sperm morphology and function in normal and pathological conditions. The fatty acid profile determines the performance not only of sperm motility but also of acrosomal reaction and sperm-oocyte fusion. This review presents available knowledge on the role of fatty acid composition in human sperm and spermatogenesis and discusses the influence of dietary fatty acids on the sperm fatty acid profile. Recent studies in biological sciences and clinical researches in this field are also reported. The topic object of this review has potential application in medicine by identifying potential causes of infertility.

Could Dietary Supplementation with Different Sources of N-3 Polyunsaturated Fatty Acids Modify the Rabbit Gut Microbiota?

Giulio Curone, Filippo Biscarini, Elisa Cotozzolo, Laura Menchetti, Alessandro Dal Bosco, Federica Riva, Paola Cremonesi, Stella Agradi, Simona Mattioli, Bianca Castiglioni, Alessia Di Giancamillo, Alice Cartoni Mancinelli, Susanna Draghi, Alda Quattrone, Giulia Collodel, Silvia Clotilde Modina, Cesare Castellini, Gabriele Brecchia

Summary: The study evaluated the effects of two dietary sources of n-3 polyunsaturated fatty acids (PUFAs) on the gut microbiota, caecal fermentations, and gastrointestinal histology in rabbits. Results showed that n-3 PUFAs supplementation led to an increase in beneficial bacterial populations and reduced Bacilli abundance and Firmicutes/Bacteroidetes ratio. These findings suggest that n-3 PUFAs diet supplementation may have positive effects on gut health, although appropriate dosage should be further investigated.

Oxidation of Polyunsaturated Fatty Acids as a Promising Area of Research in Infertility

Giulia Collodel, Elena Moretti, Daria Noto, Roberta Corsaro, Cinzia Signorini

Summary: This review examines the role of fatty acids in human pathological conditions, particularly infertility. Fatty acids and their derivatives play important roles in regulating cell membrane composition, cell signaling, cell death, immune responses, and inflammation. Understanding the metabolism of fatty acids can lead to new approaches in treating infertility.

Seminal Levels of Omentin-1/ITLN1 in Inflammatory Conditions Related to Male Infertility and Localization in Spermatozoa and Tissues of Male Reproductive System

Elena Moretti, Cinzia Signorini, Daria Noto, Sergio Antonio Tripodi, Andrea Menchiari, Ester Sorrentino, Giulia Collodel

Summary: This study explored the relationships among omentin-1/ITLN1, semen parameters, and oxidative stress markers in different patient groups. The levels of omentin-1/ITLN1 were found to be correlated with F-2-IsoP levels and negatively correlated with sperm parameters. Infertile patients showed lower levels of omentin-1/ITLN1, while those with varicocele and genitourinary infections had higher levels. Omentin-1/ITLN1 was localized differently in sperm and male reproductive system tissues.

Summary: This study aimed to investigate the effects of different types of liposomes on porcine prepubertal Sertoli cells (SCs) and to find a suitable carrier for in vitro testing of drugs. The results showed that anionic and cationic liposomes altered the distribution of actin and vimentin filaments and increased the phosphorylation levels of AMPK alpha. Zwitterionic liposomes, on the other hand, did not cause any changes in the cell cytoplasm. These preliminary data suggest that zwitterionic liposomes are the most suitable carrier for in vitro studies of SCs to understand the effects of molecules or drugs with potential clinical applications in the treatment of certain forms of male infertility.

Summary: This study investigated the changes and relationships of various molecules in the follicular fluid (FF) of infertile women with different reproductive conditions. It found that some biochemical components, hormones, antioxidant enzymes, F-2-Isoprostanes (F-2-IsoPs), and resolvin (Rv) D1 in the FF are associated with follicular growth, oocyte maturation, and female infertility. These findings highlight the importance of personalized therapeutic approaches and optimized assisted reproductive techniques (ART) outcomes.

Summary: This study conducted a quantitative research on 925 healthcare students from two institutions in Hong Kong and used the Social Interaction Anxiety Scale (SIAS-6) and Social Phobia Scale (SPS-6). The results showed that these scales have good psychometric properties among healthcare students in Hong Kong. The study also proposed quantitative, qualitative, and mixed methods research designs to guide researchers in contributing to the discussion of student's social interactions in IPE.

Human Sperm as an In Vitro Model to Assess the Efficacy of Antioxidant Supplements during Sperm Handling: A Narrative Review

Elena Moretti, Cinzia Signorini, Roberta Corsaro, Maria Giamalidi, Giulia Collodel

Summary: Spermatozoa produce reactive oxygen species (ROS) due to aerobic metabolism, which can have both beneficial and detrimental effects on cell function. Assisted reproductive procedures, such as cryopreservation, can lead to excessive ROS production and oxidative damage to spermatozoa. This review focuses on using human sperm as an in vitro model to study the supplementation of antioxidants in media. The presence and synergic effects of different antioxidant molecules may lead to more effective products in vitro and potentially in vivo.

Summary: This study investigated the association of apelin and resolvin (RvD1) in the testis and sperm with the oxidative status of semen and serum testosterone in rabbits fed different diets enriched with flaxseed or fish oil. The results showed that flaxseed diet increased the levels of apelin in the testis and enhanced serum testosterone. Both enriched diets showed higher levels of malondialdehyde and RvD1 in the testis. Apelin and its receptor were also detected in ejaculated sperm.

F4-Neuroprostane Effects on Human Sperm

Elena Moretti, Cinzia Signorini, Daria Noto, Roberta Corsaro, Lucia Micheli, Thierry Durand, Camille Oger, Jean Marie Galano, Giulia Collodel

Summary: After incubation with F-4-NeuroPs, selected human sperm showed increased motility, membrane mitochondrial potential, and reacted acrosome percentage. Chromatin integrity did not differ between treated and control samples. PSA staining highlighted a significantly higher number of sperm with reacted acrosomes in the treated group after 4 hours. Immunofluorescence analysis revealed a dense distribution of ryanodine receptors in the midpiece and postacrosomal sheath of sperm after 2 hours, followed by a dotted distribution after 4 hours. F-4-NeuroPs at an appropriate concentration can induce sperm capacitation via ion channels, which may enhance the success of assisted fertilization.
