Exploring the Anticancer Potential of Diiron Bis-cyclopentadienyl Complexes with Bridging Hydrocarbyl Ligands: Behavior in Aqueous Media and In Vitro Cytotoxicity

A series of diiron complexes based on the [Fe2Cp2(CO)(x)] skeleton (Cp = eta(5)-C5H5, x = 2, 3; eta(4)-C5H5Ph in place of one Cp in one case) and containing different bridging hydrocarbyl ligands (aminocarbyne, thiocarbyne, allenyl) were preliminarily investigated for their anticancer potential. The water solubility, stability in water and in the presence of a cell culture medium, and octanol/water partition coefficient were evaluated by spectroscopic techniques. The cytotoxicity was assessed in vitro toward the human ovarian carcinoma cell line A2780, the human triple negative breast cancer cell line MDA-MB-231, and the human vascular smooth muscle cell line SMC. Some aminocarbyne complexes exhibited a potent cytotoxicity, with IC50 values in the low micromolar/nanomolar range, and a strong selectivity for the A2780 cells in comparison to the SMC cell line. Several experiments were carried out in order to give insight into the mode of action of selected compounds, including an assessment of catalytic NADH oxidation and ROS production and studies of binding with DNA and with a model protein.

Diiron bis-cyclopentadienyl complexes as transfer hydrogenation catalysts: The key role of the bridging aminocarbyne ligand

Summary: The catalytic activity of a series of diiron complexes based on the {Fe2Cp2(CO)(x)} core (x = 2-3) and containing a bridging aminocarbyne ligand was screened in the transfer hydrogenation reaction of cyclohexanone from isopropanol. The best catalyst among these complexes was [Fe2Cp2(CN)(CO)(mu-CO){mu-CN(Me)(4-C6H4OMe)}], 3a, which exhibited moderate activity and showed a carbyne-centered mechanism, stabilized by the cyanide co-ligand, as supported by DFT calculations and IR analyses.

Hypocholesterolaemic treatment in coronary unit: from statins to anti PCSK9 therapies and bempedoic acid

Summary: The knowledge that a significant proportion of ACS survivors are at risk of recurrent ischemic cardiovascular events highlights the importance of lipid-lowering strategies in secondary prevention. Statin treatment has been shown to improve clinical outcomes. This review critically discusses the use of statin and non-statin lipid-lowering approaches, including PCSK9 inhibitors, in early management of ACS patients. Cost is a major barrier to the widespread prescription of non-statin agents in this field.

Iminopyridine Copper Complex-Based Catalysts for 1,3-Dienes Stereospecific Polymerization

Summary: Novel copper complexes were synthesized by reacting copper(II) dichloride and copper(II) dibromide with pyridyl-imine ligands with different substituents. The complexes were characterized and their molecular structure was determined. When combined with methylaluminoxane (MAO), the complexes acted as active and selective catalysts for the polymerization of 1,3-dienes, resulting in polymers with unusual structures.

Diiron Aminocarbyne Complexes with NCE- Ligands (E = O, S, Se)

Summary: Diiron mu-aminocarbyne complexes can be synthesized by reacting sodium cyanate and tetrabutylammonium thiocyanate with pre-prepared diiron aminocarbyne complexes, yielding different substituted diiron cyanide complexes with yields of 67-81%, through substitution of the acetonitrile ligand. When reacted with potassium selenocyanate at reflux temperature, cyanide complexes can be obtained with yields of 45-67%. When reacted at room temperature in acetone, a small amount of potassium selenocyanate complexes can be obtained. All products were fully characterized and the chiral structure was determined by single crystal X-ray diffraction. DFT calculations shed light on the coordination mode and stability of the {NCSe-} fragment.

Fixed Combination for the Treatment of Dyslipidaemia

Summary: Lipid-lowering fixed-dose combinations may be a better approach to control plasma lipids and reduce the burden of ASCVD. Recent studies suggest that combination therapy of moderate-intensity statin with ezetimibe is as effective as high-intensity statin monotherapy in reducing cardiovascular outcomes. Fixed-dose combinations of bempedoic acid with ezetimibe and fenofibrate with atorvastatin have shown superior results in lowering LDL-C and triglycerides respectively.

Adding Diversity to a Diruthenium Biscyclopentadienyl Scaffold via Alkyne Incorporation: Synthesis and Biological Studies

Summary: We synthesized and evaluated the anticancer potential of two series of diruthenium biscyclopentadienyl carbonyl complexes. Novel dimetallacyclopentenone compounds (2-4) were obtained from the thermal reaction of [Ru2Cp2(CO)(μ-CO)(μ-η^1:η^3-C(Ph)C(Ph)C(O)}], 1, with alkynes HCCR [R = C5H4FeCp (Fc), 3-C6H4(Asp), 2-naphthyl; Cp = η(5)-C5H5, Asp = OC(O)-2-C6H4C(O)Me]. The resulting complexes showed cytotoxic activity against cancer cells, different interactions with nucleic acids, and production of ROS.

Cyclic Ruthenium-Peptide Conjugates as Integrin-Targeting Phototherapeutic Prodrugs for the Treatment of Brain Tumors

Summary: Investigation of tumor-targeting photoactivated chemotherapy using a chiral ruthenium-based anticancer warhead conjugated to an RGD-containing peptide showed strong phototoxicity and tumor-killing effects both in vitro and in vivo. The designed ruthenium-chelating peptide had triple action by preventing biomolecule coordination, self-assembling into nanoparticles, and specifically binding to tumor cells. These results demonstrate the potential of light-sensitive integrin-targeted ruthenium-based compounds for brain cancer treatment.

A proficient multivariate approach for iron(ii) spin crossover behaviour modelling in the solid state

Summary: Iron(ii) bis-pyrazolilpyridyl (bpp-R) complexes [Fe(bpp-R)(2)](X)(2)·solvent undergo spin transition from high spin (S = 2, HS) to low spin (S = 0, LS) in the solid state, known as spin crossover (SCO). The distortion of the octahedral coordination environment is affected by crystal packing and modulates the SCO behavior. This study used a multivariate approach combining Principal Component Analysis and Partial Least Squares regression to analyze the structural data of HS structures and efficiently model and predict the spin transition temperature T-1/2 for complexes with different R groups, X- anions, and co-crystallized solvents.

Iminium substituent directs cyanide and hydride additions to triiron vinyliminium complexes

Summary: The reactivity of diiron bis-cyclopentadienyl cationic complexes containing a vinyliminium ligand coordinated to a diiron scaffold and decorated with a ferrocenyl group, [1a-g]CF3SO3, was investigated towards NBu4CN and NaBH4. Various addition products were obtained depending on the nature of the iminium substituents. The products were characterized using analytical and spectroscopic methods, and the molecular structures and thermodynamics were studied using DFT calculations.

The beneficial effect of cyclohexyl substituent on the in vitro anticancer activity of diiron vinyliminium complexes

Summary: Novel diiron vinyliminium complexes were synthesized and characterized by various spectroscopic techniques. Their solubility in D2O, Log Pow coefficients, and stability in aqueous media were determined. The antiproliferative activity of these complexes on cancer cell lines was evaluated and their mechanism of action was investigated by assessing their influence on mitochondrial respiration.

Managing transition metal interferences during chemical vapor generation using the hydrolysis products of tetrahydridoborate: a proof-of-concept study

Summary: The use of BH intermediates as derivatization reagents significantly improves the tolerance to interference from transition metal ions in the generation of SbH3 and BiH3, and shows high tolerance in a wide range of acidity. The method has been successfully applied to the determination of Sb and Bi in complex samples, providing further understanding of the chemical behavior of borohydrides.
