Summary: SARS-CoV-2 infection induces the formation of viral Z-RNA, leading to activation of the ZBP1-RIPK3 pathway and subsequent inflammatory responses. Inhibition of RIPK3 or deletion of MLKL reduces IL-1 beta release caused by SARS-CoV-2 infection. Deficiency of ZBP1 or RIPK3 reduces the production of inflammatory cytokines and chemokines, immune cell infiltration, and lung damage in SARS-CoV-2 infected models. Targeting the ZBP1-RIPK3 axis may have therapeutic potential in treating COVID-19.

Summary: In this study, the involvement and role of receptor-interacting protein 3 (RIP3)-mediated necroptosis in periodontitis were investigated. RIP3 phosphorylation and activation were observed in the periodontal ligament of mice infected with Porphyromonas gingivalis. Knockdown of RIP3 reduced osteoclastogenesis and inflammatory cytokines in the periodontal area, and alleviated alveolar bone loss. In vitro experiments showed that P. gingivalis-LPS induced RIP3-mediated necroptosis in fibroblast cells, and knockdown of RIP3 decreased the expression of inflammatory cytokines. These findings suggest that RIP3-mediated necroptosis plays a role in periodontitis, and targeting RIP3 may be a potential treatment approach.

Summary: This study found that curcumin has potential therapeutic effects in ulcerative colitis by inhibiting necroptosis of intestinal epithelial cells through blocking the phosphorylation of RIP3 and MLKL. In mouse models, curcumin also inhibited inflammatory responses caused by necroptosis signaling pathway and improved colitis pathology.

Mechanically, shikonin induced necroptosis and impaired autophagic flux through ROS generation. The crosstalk between necroptosis and autophagy was also observed.

Summary: Serum amyloid A (SAA) plays a critical role in acute pancreatitis (AP) by inducing necroptosis in acinar cells through a RIP3-dependent pathway, making it a potential drug target for AP treatment. In a caerulein-induced AP model, the specific SAA3 isoform was significantly increased, and SAA3-knockout mice displayed reduced pancreatic damage and inflammation, suggesting the importance of SAA3 in the development of AP.

Summary: In this study, a series of celastrol derivatives containing 1, 2, 3-triazole moiety were designed and synthesized, and their activities against glioma cells were evaluated. Compound 6i exhibited potent anti-glioma activity by inducing necroptosis through the RIP1/RIP3/MLKL pathway. Moreover, compound 6i showed acceptable BBB permeability in mice and inhibited glioma cell proliferation in an in vivo zebrafish xenograft model.

Summary: This study aimed to investigate the mechanism of heat-induced intestinal epithelial cell death. The results showed that heat stress upregulated TLR3 and facilitated the formation of TRIF-RIP3 complex, which activated the RIP3-MLKL signaling pathway, leading to necroptosis in intestinal epithelial cells. In addition, heat stress promoted the phosphorylation of p53 and increased TLR3 expression. Thus, heat stress induced necroptosis in intestinal epithelial cells through the p53-TLR3-TRIF-RIP3-MLKL signaling pathway.
