Background: Research has shown that mixed dementia is more common than previously believed but little is known of its early stages. Objective: To examine if incipient mixed dementia can be differentiated from incipient Alzheimer's disease (AD) and subcortical ischemic vascular dementia (SVD) using neuropsychological tests, cerebrospinal fluid (CSF) markers, and magnetic resonance imaging markers. Methods: We included 493 patients and controls from the Gothenburg MCI study and used the dementia groups for marker selection (CSF total-tau (T-tau), phospho-tau (P-tau), and amyloid-beta(42) (A beta(42)), 11 neuropsychological tests, and 92 regional brain volumes) and to obtain cut-off values which were then applied to the MCI groups. Results: Incipient mixed dementia was best differentiated from incipient AD by the Word fluency F-A-S test and the Trail making test A. CSF T-tau, P-tau, and A beta(42) differentiated incipient mixed dementia from incipient SVD. Conclusion: Incipient mixed dementia is characterized by an AD-like biomarker profile and an SVD-like cognitive profile. Incipient mixed dementia can be separated from incipient AD and incipient SVD using CSF markers and cognitive testing.

Cerebrospinal Fluid Sulfatide Levels Lack Diagnostic Utility in the Subcortical Small Vessel Type of Dementia

Johan Svensson, Maria Blomqvist, Petronella Kettunen, Carl Eckerstrom, Marcus Henricsson, Michael Jonsson, Maria Bjerke, Jan-Eric Mansson, Anders Wallin

Summary: This study investigated the diagnostic utility of CSF ST levels in subcortical small vessel type of dementia (SSVD). The results showed a correlation between CSF ST levels and WMH volume, but there was no significant difference in CSF ST levels between dementia groups and controls, suggesting that CSF total ST level may not be an ideal biomarker of demyelination in SSVD. Further research is needed to explore the mechanisms underlying the correlation between CSF ST levels and CSF/serum albumin ratio.

Effects of Holding Density on the Welfare of Zebrafish: A Systematic Review

Marica Andersson, Petronella Kettunen

Summary: The welfare of zebrafish is influenced by various environmental factors, including food access, water quality, and holding density. Despite the widespread use of zebrafish in research, the impact of holding density on its welfare remains poorly understood. Future recommendations on holding density may vary depending on the specific welfare aspects being considered, indicating a need for further research on the interactive effects of density on welfare indicators.

Long-term glycemic variability and the risk of mortality in diabetic patients receiving peritoneal dialysis

Summary: The study investigates the association between HbA1c variability and the risk of all-cause mortality in diabetic patients with PD. The results suggest that higher HbA1c variability is associated with increased risk of death in diabetic patients with PD.

Higher thyroid function is associated with accelerated hippocampal volume loss in Alzheimer's disease

Patrick Quinlan, Alexandra Horvath, Carl Eckerstrom, Anders Wallin, Johan Svensson

Summary: This study found that higher thyroid hormone levels were associated with lower brain volume and increased risk of Alzheimer's disease (AD) in elderly individuals. However, the relationship between serum thyroid hormones and hippocampal atrophy rates has not been previously investigated. The results showed that FT3/FT4 ratio was related to baseline hippocampal volume in all study groups, but only in AD patients, higher levels of thyroid hormones were associated with greater annual loss of hippocampal volume.

Orthotopic Transplantation of Human Paediatric High-Grade Glioma in Zebrafish Larvae

Susanna Larsson, Petronella Kettunen, Helena Caren

Summary: Brain tumours are a leading cause of death in children with solid tumours, and high-grade gliomas are particularly devastating. This study presents a clinically relevant in vivo model for paediatric glioma using zebrafish larvae and primary glioma stem cell cultures. The model allows for the monitoring of tumour invasion and evaluation of drug treatments. This research has the potential to improve understanding of cellular behaviour and personalized treatments for paediatric high-grade gliomas.

Summary: The housing density of zebrafish affects their welfare, with a density of 1 fish/L leading to higher stress levels and aggressive behavior. However, densities between 4 and 16 fish/L do not compromise the welfare of zebrafish.

Bone-Derived IGF-I Regulates Radial Bone Growth in Adult Male Mice

Summary: Insulin-like growth factor-I (IGF-I) levels and cortical bone dimensions play important roles in determining fracture risk in elderly people. Inactivation of liver-derived circulating IGF-I reduces periosteal bone expansion in young and older mice. This study investigates the effect of inducible inactivation of IGF-I specifically in bone on the bone phenotype in adult/old mice, finding that it results in reduced radial growth of cortical bone, suggesting the importance of locally derived IGF-I in regulating the cortical bone phenotype in older mice.

Blood-brain barrier dysfunction and reduced cerebrospinal fluid levels of soluble amyloid precursor protein-β in patients with subcortical small-vessel disease

Petronella Kettunen, Maria Bjerke, Carl Eckerstrom, Michael Jonsson, Henrik Zetterberg, Kaj Blennow, Johan Svensson, Anders Wallin

Summary: We found that SSVD patients have a different biomarker profile, with lower levels of sAPP-β compared to AD patients and controls. We also observed signs of blood-brain barrier dysfunction in SSVD. These findings support the concept that SSVD is a distinct form of vascular cognitive disorder (VCD).
