miR-20b and miR-451a Are Involved in Gastric Carcinogenesis through the PI3K/AKT/mTOR Signaling Pathway: Data from Gastric Cancer Patients, Cell Lines and Ins -Gas Mouse Model

Gastric cancer (GC) is one of the most common and lethal gastrointestinal malignancies worldwide. Many studies have shown that development of GC and other malignancies is mainly driven by alterations of cellular signaling pathways. MicroRNAs (miRNAs) are small noncoding molecules that function as tumor-suppressors or oncogenes, playing an essential role in a variety of fundamental biological processes. In order to understand the functional relevance of miRNA dysregulation, studies analyzing their target genes are of major importance. Here, we chose to analyze two miRNAs, miR-20b and miR-451a, shown to be deregulated in many different malignancies, including GC. Deregulated expression of miR-20b and miR-451a was determined in GC cell lines and the INS-GAS mouse model. Using Western Blot and luciferase reporter assay we determined that miR-20b directly regulates expression of PTEN and TXNIP, and miR-451a: CAV1 and TSC1. Loss -of-function experiments revealed that down-regulation of miR-20b and up-regulation of miR-451a expression exhibits an anti -tumor effect in vitro (miR-20b: reduced viability, colony formation, increased apoptosis rate, and miR-451a: reduced colony forming ability). To summarize, the present study identified that expression of miR-20b and miR-451a are deregulated in vitro and in vivo and have a tumor suppressive role in GC through regulation of the PI3K/AKT/mTOR signaling pathway.

The Influence of Calcium Ions on the Electrotransfer Efficiency of Plasmid DNA and Cell Viability

Summary: Gene electrotransfer (GET) is a promising technique for gene therapy, but its success rate is influenced by the understanding of electroporation mechanism and other factors. This study analyzed the impact of low concentrations of Ca2+ on DNA electrotransfer and found that even 0.25 mM CaCl2 can significantly decrease the efficiency of GET and cell viability. The observed decrease in pDNA electrotransfer was not due to cell death but secondary mechanisms. The findings highlight the importance of Ca2+ in gene electrotransfer and its potential application in vivo.

Factors Affecting Performance of DNA Methylation as a Potential Biomarker in Ascites for Peritonitis and Peritoneal Carcinomatosis

Summary: This study aimed to evaluate DNA methylation as a diagnostic tool for differentiating between benign and malignant ascites. The results showed that methylation levels were associated with ascites in patients with tumors, but not in patients without tumors. In addition, inflammatory cells were also found to be related to DNA methylation changes.

Unlocking the bentonite microbial diversity and its implications in selenium bioreduction and biotransformation: Advances in deep geological repositories

Summary: In this study, the impact of bentonite microbial communities on the allotropic transformation of Se(IV) bioreduction products under DGR relevant conditions was investigated. Moreover, the Se amendment-dependent shifts in the bentonite microbial populations were assessed. The results showed that the bentonite color changed from orange to black after selenite treatment, and the allotropic changes of Se bioreduction products were tracked using analytical techniques. The distribution of strains involved in Se(IV) reduction and Se(0) allotropic biotransformation potential was identified.

Comparison of genomic and transcriptional microbiome analysis in gastric cancer patients and healthy individuals

Summary: This study characterized the microbiota of gastric cancer using 16S rRNA gene and its transcript and identified differences in bacterial composition. The results showed that there was only partial overlap between DNA and RNA samples and differences between gastric cancer patients and healthy controls depending on the chosen sequencing modality. Furthermore, RNA sequencing was more sensitive for detecting differences in bacterial richness and specific bacteria. Overall, the choice of sequencing modality affects the characterization of the microbiota and the differences between case and control.

Fecal Microbiota Transfer (FMT) in Germany - Status and Perspective

Summary: The frequency of FMT use in Germany is lower compared to other European countries. One of the obstacles is the regulatory classification of FMT as a non-approved drug, resulting in higher costs and reimbursement difficulties. The proposed regulation by the European Commission to classify FMT as a transplant may change the regulatory situation in Germany and promote nationwide availability of this recommended therapeutic procedure.

Mucin-microbiome signatures shape the tumor microenvironment in gastric cancer

Summary: This study identified the mucin-microbiome signatures in gastric adenocarcinomas and their impact on clinical outcomes. The findings showed that intestinal mucin phenotype and high-level MUC13 expression were associated with poor survival, while gastric MUC5AC or MUC6 abundance was associated with a more favorable outcome. Furthermore, oral bacteria such as Neisseria, Prevotella, and Veillonella were found to play a potential role in MUC13 signaling and were associated with intestinal and mixed mucin phenotype tumors.

The Assessment of Calcium and Bleomycin Cytotoxic Efficiency in Relation to Cavitation Dosimetry

Summary: Microbubble and ultrasound facilitated intracellular Ca2+ delivery, known as sonoporation, offers a promising alternative to conventional chemotherapy due to its spatio-temporally controllable and side-effect-free nature. This study demonstrates that a 5 mM concentration of Ca2+ in combination with ultrasound or ultrasound and Sonovue microbubbles can be used as an alternative to the anticancer drug bleomycin. Ca2+ delivery via sonoporation induces cell death in Chinese hamster ovary cells, similar to the combination of bleomycin and sonoporation, without causing systemic toxicity. Furthermore, Ca2+ delivery via sonoporation alters vital characteristics of viable cells and elicits sudden cell death within a short period of time.

Ionomic Parameters of Populations of Common Juniper (Juniperus communis L.) Depending on the Habitat Type

Summary: In this study, the ionomic parameters of Juniperus communis needles in different habitats in Lithuania were analyzed. The concentrations of elements in the needles varied significantly between contrasting populations, and the concentrations of main nutritional elements were lower in transition mires and quaking bogs.

The Translational Impact of Plant-Derived Xeno-miRNA miR-168 in Gastrointestinal Cancers and Preneoplastic Conditions

Summary: This study aimed to evaluate the clinical relevance of xeno-miRNA miR-168 in the gastric mucosa during preneoplastic conditions and gastric carcinogenesis. The results showed that miR-168 levels were higher in patients with moderate and severe intestinal metaplasia, suggesting its potential involvement in mucosal lesions and gastric cancer development.

Summary: The study investigated the relationship between fecal microbiota diversity and composition and plasma bile acid levels in young adults. The results showed that fecal microbiota diversity and evenness, as well as the relative abundance of Firmicutes and Bacteroidetes phyla, were positively associated with plasma levels of secondary bile acids. However, further investigations are needed to confirm whether gut microbiota composition can regulate plasma bile acid concentrations in humans.

Comparative Analysis of Pulsed Electric Fields (PEF) and Traditional Pasteurization Techniques: Comparative Effects on Nutritional Attributes and Bacterial Viability in Milk and Whey Products

Summary: In this study, the influence of pulsed electric field (PEF) processing on the nutritional properties of raw milk was evaluated and compared to conventional thermal pasteurization methods. It was found that PEF treatment significantly reduced the content of beta-lactoglobulin in liquid whey samples. However, all pasteurization methods had minimal impact on the concentrations of vitamins, amino acids, cholesterol, and fatty acids. Additionally, thermal pasteurization effectively eliminated bacterial colonies.
