During almost 40 years of use, the micronucleus assay (MN) has become one of the most popular methods to assess genotoxicity of different chemical and physical factors, including ionizing radiation-induced DNA damage. In this minireview, we focus on the position of MN among the other genotoxicity tests, its usefulness in different applications and visibility by international organizations, such as International Atomic Energy Agency, Organization for Economic Co-operation and Development and International Organization for Standardization. In addition, the mechanism of micronuclei formation is discussed. Finally, foreseen directions of the MN development are pointed, such as automation, buccal cells MN and chromothripsis phenomenon.

Chlorpyrifos alters expression of enzymes involved in vitamin D3 synthesis in skin cells

Krzysztof Sawicki, Magdalena Czajka, Magdalena Matysiak-Kucharek, Jacek Kurzepa, Paulina Wojtyla-Buciora, Karol Zygo, Marcin Kruszewski, Lucyna Kapka-Skrzypczak

Summary: This study investigated the impact of the pesticide chlorpyrifos (CPS) on the expression of enzymes involved in synthesis and metabolism of vitamin D3 in human keratinocytes and fibroblasts. The results indicate that exposure to CPS significantly altered the expression of cytochrome P450 isoforms in cells, potentially disrupting vitamin D3 metabolism. Further research is needed to better understand these mechanisms.

Ivabradine prevents deleterious effects of dopamine therapy in heart failure: No role for HCN4 overexpression

Aleksandra Paterek, Barbara Sochanowicz, Marta Okninska, Witold Smigielski, Marcin Kruszewski, Urszula Mackiewicz, Michal Maczewski, Przemyslaw Leszek

Summary: Ivabradine may prevent the detrimental effects of dopamine treatment during exacerbations of chronic heart failure, potentially offering additional benefits beyond heart rate reduction. However, preventing ventricular HCN4 overexpression may not play a major role in this setting.

Increased DNA repair capacity augments resistance of glioblastoma cells to photodynamic therapy

Somayeh Shahmoradi Ghahe, Konrad Kosicki, Maria Wojewodzka, Bartosz A. Majchrzak, Anna Fogtman, Roksana Iwanicka-Nowicka, Agata Ciuba, Marta Koblowska, Marcin Kruszewski, Barbara Tudek, Elzbieta Speina

Summary: Resistance to photodynamic therapy in cancer cells may be caused by various mechanisms, including efficient repair of oxidative DNA damage and DNA breaks. Higher activity of APE1 endonuclease and increased expression and activation of DNA damage kinase ATM in the U-87 MGR cell line suggest that they are potential targets for sensitizing resistant cells to PDT.

Coating-Dependent Neurotoxicity of Silver Nanoparticles-An In Vivo Study on Hippocampal Oxidative Stress and Neurosteroids

Katarzyna Dziendzikowska, Jacek Wilczak, Wojciech Grodzicki, Joanna Gromadzka-Ostrowska, Malgorzata Wesierska, Marcin Kruszewski

Summary: The study found that silver nanoparticles disrupted the antioxidant system in the hippocampus of rats and induced oxidative stress in a coating-dependent manner, which could potentially lead to neurodegeneration and cognitive disorders. The coating material of the nanoparticles also modulated the concentrations of steroids and the expression of genes involved in steroid synthesis and metabolism.

Expression of Iron Metabolism Proteins in Patients with Chronic Heart Failure

Bogna Kozlowska, Barbara Sochanowicz, Leszek Kraj, Malgorzata Palusinska, Piotr Kolsut, Lukasz Szymanski, Slawomir Lewicki, Witold Smigielski, Marcin Kruszewski, Przemyslaw Leszek

Summary: This study investigated the changes in iron-related proteins in the myocardium of heart failure patients. The results showed reduced expression of several proteins responsible for iron transport and storage, along with increased expression of oxidative stress markers. The reduction of iron-gathering proteins and elevated oxidative stress in failing hearts is detrimental to myocardial cells.

Silver Nanoparticles Inhibit Metastasis of 4T1 Tumor in Mice after Intragastric but Not Intravenous Administration

Kamil Brzoska, Maria Wojewodzka, Malgorzata Szczygiel, Agnieszka Drzal, Martyna Sniegocka, Dominika Michalczyk-Wetula, Eva Biela, Martyna Elas, Malgorzata Kucinska, Hanna Piotrowska-Kempisty, Lucyna Kapka-Skrzypczak, Marek Murias, Krystyna Urbanska, Marcin Kruszewski

Summary: This study analyzed the impact of different silver nanoparticles on the development and metastatic potential of breast cancer using a mouse model. The results showed that citrate-coated silver nanoparticles had a significant effect on reducing tumor volume and lung metastases in mice treated intragastrically. However, no effect on lung metastases was observed after intravenous administration of silver nanoparticles. The findings suggest that the anticancer activity of silver nanoparticles may depend on the activation of the immune system.

Exposure to Chlorpyrifos Alters Proliferation, Differentiation and Fatty Acid Uptake in 3T3-L1 Cells

Magdalena Czajka, Krzysztof Sawicki, Magdalena Matysiak-Kucharek, Marcin Kruszewski, Jacek Kurzepa, Paulina Wojtyla-Buciora, Lucyna Kapka-Skrzypczak

Summary: Chlorpyrifos, a representative of organophosphorus pesticides, has effects on the viability, proliferation, differentiation, and fatty acid uptake in 3T3-L1 cells. Exposure to chlorpyrifos decreases cell viability but increases it at low concentrations. It inhibits proliferation and differentiation of preadipocytes, reduces lipid accumulation, and down-regulates key transcription factors. In fully differentiated adipocytes, chlorpyrifos increases basal free fatty acid uptake. These findings suggest that chlorpyrifos may play a role in the development of obesity.
