Qigong Mind-Body Exercise as a Biopsychosocial Therapy for Persistent Post-Surgical Pain in Breast Cancer: A Pilot Study

Purpose: To assess the feasibility, safety, and preliminary effectiveness of a 12-week multimodal Qigong Mind-Body Exercise (QMBE) program for breast cancer survivors with persistent post-surgical pain (PPSP). Methods: This was a single-arm mixed-methods pilot study. Primary outcome measures were feasibility (recruitment, adherence) and safety. Validated self-report questionnaires were used to evaluate a constellation of interdependent symptoms, including pain, fatigue, mood, exercise, interoceptive awareness, and health-related quality of life at baseline and 12 weeks. A subset of the instruments was administered 6 months postintervention. Shoulder range of motion and grip strength were objectively assessed at baseline and 12 weeks. Qualitative interviews were conducted at baseline and 12 weeks. Results: Twenty-one participants were enrolled; 18 and 17 participants, respectively, completed the 12-week and 6-month outcome assessment. No serious adverse events were reported. Statistically significant improvements were observed at 12 weeks in pain severity and interference, fatigue, anxiety, depression, perceived stress, self-esteem, pain catastrophizing, and several subdomains of quality of life, interoceptive awareness, and shoulder range of motion. Changes in pain, fatigue, pain catastrophizing, anxiety, depression, and quality of life were clinically meaningful. Postintervention effects were sustained at 6 months. Conclusions: QMBE is a safe and gentle multimodal intervention that shows promise in conferring a broad range of psychosocial and physical benefits for breast cancer survivors with PPSP. Results support the value of future studies evaluating the impact of QMBE on multiple outcomes relevant to breast cancer survivors with PPSP.

Summary: This study aimed to assess the feasibility of objectively measuring posture and explore the relationship between posture and affect in breast cancer survivors with persistent postsurgical pain during a 12-week Qigong training. The preliminary findings provide support for considering emotions and patient-centered outcomes in an embodied framework, and highlight the potential of Qigong as an intervention for addressing complex biopsychosocial interventions such as persistent postsurgical pain in breast cancer survivors.

Voice Biomarkers of Recovery From Acute Respiratory Illness

Immune checkpoint inhibitor gastritis is often associated with concomitant enterocolitis, which impacts the clinical course

Summary: Gastrointestinal immune-related adverse events caused by immune checkpoint inhibitors (ICIs) often require interruption of cancer treatment. This study examined the clinical features and treatment outcomes of ICI gastritis. It was found that ICI gastritis can occur alone or more commonly with concurrent enteritis/colitis, with different clinical courses. Gastric biopsies are necessary to diagnose clinically significant cases with normal endoscopic appearance. Most patients with isolated gastritis can continue ICI therapy uninterrupted, but some may require glucocorticoids.

Granulocyte-Macrophage Colony-Stimulating Factor Influence on Soluble and Membrane-Bound ICOS in Combination with Immune Checkpoint Blockade

Summary: With the successful development of immune checkpoint blockade, the addition of granulocyte-macrophage-CSF (GM-CSF) has been shown to improve efficacy and decrease adverse events. The presence of ICOS+CD4+ or ICOS+CD8+ T cells in the peripheral blood is significantly higher in patients treated with ipilimumab plus GM-CSF compared to those treated with ipilimumab alone. Soluble ICOS splice variants have suppressive effects and can serve as a biomarker for GM-CSF and immune checkpoint blockade-based therapies.

Summary: By studying 51 newly diagnosed DLBCL cases, it was found that the organization of tumor and immune cells can be modeled by 6 composite cell neighborhood types (CNTs) and divided into immune-deficient, DC-enriched, and Mac-enriched TIMEs. Through the analysis of this organization structure, the cellular composition and spatial distribution characteristics of DLBCL can be understood.

Pancreatitis and Hyperlipasemia in the Setting of Immune Checkpoint Inhibitor Therapy

Summary: This study evaluated the etiology of hyperlipasemia and the clinical characteristics, management, and outcomes of immune checkpoint inhibitor-induced pancreatic injury (ICI-PI) in patients receiving immune checkpoint inhibitor (ICI) therapy. The results showed that 29% of individuals presented with asymptomatic hyperlipasemia and pancreatitis, with pancreatic injury attributed to ICI use in 105 individuals.

YAP1 and WWTR1 expression inversely correlates with neuroendocrine markers in Merkel cell carcinoma

Summary: This study reveals a previously unrecognized heterogeneity in the gene expression of Merkel cell carcinoma (MCC) and identifies an inverse correlation between neuroendocrine gene expression and the Hippo pathway transcription factors YAP1 and WWTR1. The expression of YAP1 or WWTR1 induces cell-cycle arrest in MCPyV-positive MCC cell lines through the TEAD-dependent repression of MCPyV LT. These findings shed light on the development of MCPyV-positive MCC and have implications for diagnosis and potential therapeutic interventions.
